Short answer: Vitamin D2 (ergocalciferol) and vitamin D3 (cholecalciferol) both raise blood levels of 25-hydroxyvitamin D, the usual marker of vitamin D status. The NIH Office of Dietary Supplements (ODS) says most evidence shows D3 raises that level more and keeps it higher for longer than D2. That difference does not make D3 automatically right for every person. Your choice should depend on whether you are treating a diagnosed deficiency, following a prevention recommendation, and whether the ingredient source fits your dietary or ethical requirements.
The Endocrine Society’s 2011 guideline accepted either form for treating or preventing deficiency. Its 2024 guideline has a different scope: prevention in people without an established indication for vitamin D treatment or 25(OH)D testing. Those recommendations are population-specific and are not a universal instruction to take high-dose supplements.
What vitamin D2 and D3 are
Vitamin D2 and vitamin D3 are two forms of vitamin D used in supplements and prescription products. D2 is ergocalciferol; D3 is cholecalciferol. After either form is consumed, the body converts vitamin D into metabolites, including 25-hydroxyvitamin D, usually written as 25(OH)D. A blood test for 25(OH)D is the standard marker used to assess vitamin D status.
They are related compounds, but they are not the same molecule and are not interchangeable in every practical situation. The ODS describes both as capable of raising serum 25(OH)D. The strongest consistent difference in the ODS summary is that D3 generally produces a larger increase and maintains higher concentrations for longer than D2.

That laboratory difference is one part of a decision, not a stand-alone prescription. A form that performs better on a blood marker may still be inappropriate for someone whose clinician selected the other form, whose product has a different source, or whose treatment plan requires monitoring.

The most important distinction: prevention or treatment?
Before comparing forms, identify why vitamin D is being considered. “Should I take vitamin D to prevent disease?” is a different clinical question from “My clinician diagnosed deficiency; which regimen should I follow?” The two questions are covered by different guidance.
Do these 3 things before closing this tab:

If a clinician has identified vitamin D deficiency or another established indication, the form, amount, schedule and follow-up belong in that treatment plan. The Endocrine Society’s 2011 guideline suggested either D2 or D3 for treatment and prevention of deficiency. That statement means both forms were considered acceptable options in that guideline’s scope; it does not establish that every dose or product is equivalent for every patient.
A diagnosed deficiency can coexist with factors that affect absorption, metabolism, adherence or safety. For that reason, do not replace a prescribed regimen with an over-the-counter product simply because D3 is often more persistent in population evidence. Ask the prescriber whether changing form would alter the intended schedule or follow-up testing.
Routine prevention in people without an established indication
The Endocrine Society’s 2024 guideline addresses empiric supplementation for disease prevention in people without an established indication for vitamin D treatment or 25(OH)D testing. It is not a replacement for dietary reference intakes, and it does not apply to people whose clinician has already identified a reason for treatment.

For prevention, the guideline suggests empiric vitamin D supplementation for several population groups: children and adolescents aged 1–18, adults older than 75, pregnant people, and people with high-risk prediabetes. The rationale is potential population-specific benefit. These are conditional, population-level recommendations, not instructions that every person in one of those groups should select a particular product or dose without clinical context.

For healthy adults younger than 75, the 2024 guideline suggests against empiric supplementation above current reference dietary intakes solely to lower disease risk. “Above reference intakes” is important: the statement is about extra supplementation for prevention, not a directive to stop a clinician-prescribed treatment or to disregard a documented deficiency.
What the 2024 guideline does—and does not—tell you
It does not identify one universally optimal dose
The guideline says optimal empiric doses remain unclear for the populations it considered. That uncertainty is a reason to avoid treating a single internet dose as a universal answer. The appropriate amount can depend on the purpose of supplementation, age, pregnancy status, medical history, other sources of vitamin D and the product’s instructions.
It does not recommend routine testing for everyone in those populations
The guideline suggests against routine 25(OH)D testing in the populations it considered when outcome-specific benefits based on a measured level have not been established. This is a prevention recommendation, not a ban on testing. If a qualified clinician orders a test because of symptoms, a diagnosed deficiency, a malabsorption concern, medication effects or another established indication, that clinical reason takes precedence.
It is not a high-dose supplement recommendation
Population-specific prevention guidance should not be converted into a blanket recommendation for high-dose vitamin D. The 2024 document’s scope and uncertainty about optimal doses both argue for following the relevant guideline category and discussing personal circumstances with a clinician.

The ODS summarizes evidence showing that both forms raise serum 25(OH)D, while most studies find D3 raises it more and maintains higher levels for longer than D2. This is a comparison of the measured biomarker, not proof that D3 delivers superior results for every health outcome or every clinical condition.

The distinction matters when a clinician is trying to correct or maintain a measured level and has chosen a formulation accordingly. It matters less as a reason to self-treat when you have no established indication, because the 2024 prevention guideline does not define one optimal dose and does not recommend routine testing across the considered populations.
The ODS also states that both forms “raise serum 25(OH)D levels, and they seem to have equivalent ability to cure rickets.” That sentence is specific to rickets. It should not be expanded into a claim that D2 and D3 are identical for all outcomes, doses or patients.
Ingredient source and dietary preference
How D2 is made
According to the ODS, D2 is manufactured by exposing ergosterol from yeast to ultraviolet light. People who prefer a yeast-derived form may regard that source as relevant when comparing labels.

D3 is typically produced by exposing 7-dehydrocholesterol from lanolin, a substance obtained from sheep’s wool, to ultraviolet light. Lanolin is not the same as a meat ingredient, but people following a strict vegan standard may still want to confirm the source and processing details with the manufacturer.
Lichen-derived D3
The ODS notes that animal-free D3 sourced from lichen is available. A label that says “vegan D3” may refer to this type of source, but wording and certification practices vary. Ask the manufacturer whether the vitamin itself is lichen-derived and whether other ingredients or processing aids meet your requirements.
Source preference does not establish a different medical dose. Choose a product that fits the treatment plan or prevention context first, then verify that its source and excipients meet your dietary needs.

A practical decision framework
- Identify the reason for use. Write down whether you are responding to a laboratory-confirmed deficiency, following advice for a specific condition, or considering routine prevention without a diagnosed indication.
- Check whether a clinician selected the form. If a prescription or written plan names ergocalciferol or cholecalciferol, follow that plan until you have discussed a change with the prescriber.
- Place your situation in the 2024 guideline’s scope. The prevention recommendations specifically discuss ages 1–18, adults older than 75, pregnancy and high-risk prediabetes. Healthy adults younger than 75 are advised against empiric supplementation above reference dietary intakes for disease-risk reduction.
- Compare the form’s expected blood-level behavior. If all other considerations are equal and you are choosing an over-the-counter form for a clinician-approved reason, remember that ODS evidence generally favors D3 for a larger and more persistent 25(OH)D increase.
- Verify ingredient source. Check whether the D3 is lanolin-derived or lichen-derived, or whether the D2 is yeast-derived. Contact the manufacturer when the label does not answer a dietary or ethical question.
- Confirm the amount and schedule. Use the amount specified by your clinician, the applicable reference intake or the product directions appropriate to your situation. Do not infer a personal dose from the D2-versus-D3 comparison alone.
- Plan follow-up when treatment requires it. Ask whether and when 25(OH)D or other laboratory monitoring is needed. Do not order repeated tests simply because a supplement advertisement suggests frequent measurement.
When testing is useful—and when it may not be routine
Serum 25(OH)D is the usual blood marker of vitamin D status, but a test result is meaningful only in context. A clinician may use testing when deficiency is diagnosed, when a condition or medication affects vitamin D handling, or when treatment response needs to be assessed. The result can then inform a specific plan.
The 2024 Endocrine Society prevention guideline does not support routine 25(OH)D testing for everyone in the populations it considered when no outcome-specific benefit based on a level has been identified. That recommendation is narrower than “testing is never useful.” If your healthcare professional has identified an established indication, follow that testing plan even if you also fall into a population covered by the prevention guideline.
How to read a vitamin D supplement label
- Look for the form: the label should identify vitamin D2/ergocalciferol or vitamin D3/cholecalciferol.
- Check the serving information: note how many capsules, tablets or drops provide the stated amount, and do not assume two products use the same serving size.
- Confirm the source when it matters: conventional D3 is commonly lanolin-derived; lichen-derived D3 is an animal-free option noted by the ODS.
- Separate form from dose: a D3 label does not tell you that its amount is appropriate for your medical situation.
- Review the full ingredient list: capsule materials, oils, flavorings and processing aids can matter for allergies, dietary restrictions or other personal requirements.
- Check directions and storage: follow the package instructions and keep the product away from children. If the product conflicts with a clinician’s written directions, ask before taking it.
Common mistakes to avoid
Assuming “D3 is better” answers every question
D3’s generally stronger and more persistent effect on 25(OH)D is useful information, but it is not a universal treatment rule. The ODS evidence concerns the biomarker, while clinical decisions also involve indication, source, monitoring and safety.
Using prevention guidance to replace deficiency treatment
The 2024 guideline’s prevention scope does not override the 2011 deficiency-treatment guidance or a current clinician’s plan. A person with diagnosed deficiency should not downgrade or discontinue treatment based on a recommendation intended for people without an established indication.
Taking a high amount because no dose is listed in a comparison article
The 2024 guideline says optimal empiric doses remain unclear for the populations it considered. Lack of a universal number is not permission to experiment with large amounts. Use the amount appropriate to your documented indication and professional advice.

Repeating blood tests without a clinical question
Routine testing is not recommended across the 2024 guideline’s prevention populations when a level-based outcome benefit has not been established. Testing should answer a clinical question rather than simply accompany every purchase.
Examples of how the decision changes by situation
An adult with a documented deficiency
The starting point is the clinician’s diagnosis and treatment plan. Either D2 or D3 was considered acceptable in the Endocrine Society’s 2011 deficiency guideline, while ODS evidence generally favors D3 for raising and sustaining 25(OH)D. Discuss the form, amount and follow-up testing before changing a prescribed product.
A healthy 45-year-old considering extra supplementation for general disease prevention
This person is younger than 75 and has no stated indication. The 2024 guideline suggests against empiric supplementation above current reference dietary intakes solely to lower disease risk. The D2-versus-D3 comparison does not create an exception or establish a need for a high-dose product.

A pregnant person choosing an over-the-counter product
Pregnancy is one of the populations for which the 2024 guideline suggests empiric vitamin D supplementation. That recommendation still does not specify a universally optimal dose or require one form for every pregnancy. Confirm the product, amount and source with the prenatal-care clinician, especially if a prenatal vitamin already contains vitamin D.
A vegan shopper without an established indication
First determine whether supplementation is indicated under the relevant prevention or dietary guidance. If a clinician-approved product is appropriate, look for lichen-derived D3 or another clearly documented animal-free source, and verify the rest of the ingredients.
A child or adolescent aged 1–18
This age range is included among the populations for which the 2024 guideline suggests empiric supplementation. The child’s clinician or caregiver should select an age-appropriate product and amount; adult product directions should not be extrapolated to children.
Questions to ask a clinician or pharmacist
- What is the reason for supplementation in my case: deficiency treatment, another indication or prevention?
- Should I use ergocalciferol or cholecalciferol, and does the form affect my schedule?
- What amount and duration apply to my age, pregnancy status, condition and other sources of vitamin D?
- Do I need a 25(OH)D test, and what decision will the result change?
- Could my medications, digestive condition, kidney or liver health, or other supplements affect the plan?
- Does this product’s lanolin, lichen, yeast or capsule source fit my dietary requirements?
- When should treatment response or continued need be reviewed?
Bottom line for choosing D2 or D3
Both D2 and D3 raise serum 25(OH)D, and the NIH ODS says most evidence favors D3 for a larger increase that remains higher for longer. For a diagnosed deficiency or another established indication, use the form and regimen selected with a qualified clinician; the Endocrine Society’s 2011 guideline accepted either form. For prevention, apply the Endocrine Society’s 2024 population-specific guidance rather than assuming everyone needs extra supplementation or a particular dose. Finally, check the ingredient source: D2 is made from UV-treated yeast, conventional D3 commonly comes from lanolin, and lichen-derived animal-free D3 is available.
