Researchers at The University of Queensland have built an experimental herbicide that pauses weed growth instead of reliably killing the plant. Their molecule, cyclooctatetraene-metsulfuron methyl (COT-MM), is a modified version of metsulfuron methyl in which the parent compound’s phenyl ring is replaced by an eight-carbon cyclooctatetraene ring that continually changes shape.

In laboratory tests on invasive rubber vine (Cryptostegia grandiflora), COT-MM stopped growth temporarily and caused less mortality than metsulfuron methyl. The result is an example of herbistasis—holding a weed in a low-growth state rather than pursuing complete kill. It is a chemistry proof of concept, not a field-ready product: field performance, crop selectivity, environmental fate, registration and commercial availability have not been established.

What is a shape-shifting carbon ring?

A shape-shifting carbon ring is a molecular ring whose atoms can adopt changing three-dimensional arrangements instead of presenting one rigid geometry to a biological target. In this study, the ring is cyclooctatetraene, an eight-carbon structure attached to the metsulfuron methyl framework.

Craig Williams, who led the University of Queensland work, described the ring as being “in constant dynamic equilibrium – shape shifting – and thus engages enzyme and receptor active sites differently to that of a static molecular moiety.” The important point is not that the molecule visibly transforms like a macroscopic object. Its bonds continually sample different conformations, changing how much of the ring can occupy a binding site at any instant.

The researchers used that molecular movement deliberately. Instead of leaving the herbicide’s binding geometry unchanged, they altered one part of a known active ingredient to make enzyme binding less complete and less persistent. That design creates a compound with a different biological outcome from the parent herbicide.

How was COT-MM built from metsulfuron methyl?

COT-MM keeps the part of metsulfuron methyl that targets acetolactate synthase (ALS), but replaces the parent molecule’s phenyl ring with cyclooctatetraene. The substitution was intended to preserve enough target recognition for the molecule to affect ALS while reducing the strength and duration of the interaction.

What ALS does in a plant

ALS is an enzyme required to make branched-chain amino acids. Metsulfuron methyl inhibits ALS, reducing production of those amino acids and thereby disrupting the growth processes that depend on them. The parent herbicide is designed for strong inhibition; severe disruption can lead to plant death.

What the ring substitution changes

Kinetic measurements, binding experiments and molecular-docking analysis indicated that the cyclooctatetraene-containing molecule cannot fit its aromatic portion fully into the ALS binding channel. Its interaction is therefore weaker than that of metsulfuron methyl. COT-MM can move into and out of the channel, allowing some enzyme activity to continue instead of producing the parent compound’s more severe blockade.

This is a change in binding behavior, not a change from chemical weed control to a completely different target. COT-MM still acts through ALS; the proposed innovation is controlling the intensity and reversibility of that inhibition.

What does herbistasis mean?

Herbistasis is a strategy of temporarily suppressing a weed’s growth rather than trying to kill it outright. A herbistatic treatment can leave the plant alive but unable to make normal vertical or biomass gains for a period, after which growth may resume.

The term is uncommon in practical weed-control discussions. Chemistry World reported that only four previous examples had been identified before this work, according to the researchers’ review. Williams emphasized that the concept is still unsettled because so few synthetic herbistatic agents have been made and studied.

How herbistasis differs from mortality

Mortality asks whether the treated plant dies. Herbistasis asks how long growth is held back, how completely it is suppressed, and whether the plant recovers. Those are different endpoints, so a compound can be successful at pausing growth while producing lower mortality than a conventional herbicide.

What happened when researchers treated rubber vine?

The team synthesized COT-MM and tested it on rubber vine (Cryptostegia grandiflora), an invasive species. The experiment was conducted as a laboratory proof of concept; the available reports do not establish field-scale performance or a recommended application rate.

Duration of the growth pause

The University of Queensland’s summary says COT-MM stopped rubber-vine growth for six weeks. A detailed account in Chemistry World says vertical growth resumed eight weeks after treatment and describes the response as reversible. These statements use different reporting points: one gives the reported period of stopped growth, while the other records when measurable vertical growth returned.

Lower mortality than the parent herbicide

Rubber vines treated with COT-MM had lower mortality than plants treated with metsulfuron methyl. That result is consistent with a herbistatic response: the modified molecule restricted growth without producing the parent herbicide’s stronger lethal effect.

The experiment therefore demonstrates a biological effect, not a complete weed-management program. It does not show that a single application permanently controls rubber vine, prevents reproduction, or outperforms existing treatments under field conditions.

Why did the ring change weaken ALS inhibition?

The proposed explanation combines molecular shape with binding kinetics. Metsulfuron methyl presents a relatively fixed phenyl-containing portion to ALS. In COT-MM, the cyclooctatetraene ring continually changes conformation, so the ring cannot maintain the same full fit in the enzyme’s binding channel.

Less complete fit

Molecular-docking work indicated that the modified ring prevents the aromatic portion from occupying the channel completely. A partial fit can still block ALS, but it leaves more opportunity for the enzyme to resume activity than a tightly fitting inhibitor would.

Faster movement in and out

The kinetic and binding results indicated that COT-MM can enter and leave the ALS channel. That dynamic behavior reduces the time the enzyme remains occupied. The resulting inhibition is less severe, so branched-chain amino-acid production is reduced rather than shut down to the extent associated with the parent treatment.

Why this is a design principle

The work treats molecular flexibility as a controllable variable. Instead of optimizing a compound only for the strongest possible binding, the researchers explored whether a deliberately less persistent interaction could produce a useful, temporary plant response. The rubber-vine result suggests that target engagement can be tuned, although it does not yet show how broadly the approach works across weeds or enzymes.

How does herbistasis compare with conventional weed killing?

Herbistasis and conventional herbicide treatment differ in the outcome they seek, the evidence needed to judge success and the uncertainties that remain for COT-MM.

Comparison axis Conventional metsulfuron methyl treatment COT-MM experiment What is not established
Primary outcome Stronger ALS inhibition associated with greater mortality Temporary growth suppression with lower mortality than the parent treatment Whether temporary suppression gives better long-term control in a managed site
Reversibility and duration The cited experiment does not report a matching recovery interval for the parent treatment The University of Queensland reported six weeks of stopped growth; Chemistry World reported vertical growth resuming at eight weeks How duration changes with dose, weather, plant age or repeated treatment
ALS binding The phenyl-containing parent structure supports stronger target engagement The cyclooctatetraene ring fits less completely and can move in and out of the binding channel Whether the same binding behavior occurs in other plant ALS variants
Resistance-selection idea Repeated strong selection can favor resistant survivors; the cited study does not quantify this for metsulfuron methyl Researchers propose that a growth pause might reduce selection pressure, but the proposal remains untested Whether resistance actually evolves more slowly under herbistatic programs
Weed and crop selectivity No new selectivity comparison is supplied by this experiment Only rubber vine was tested in the reported work Crop safety, effects on non-target plants and useful weed-spectrum data
Field and regulatory status Metsulfuron methyl is the established parent compound, but this study is not a use-label assessment No field trial, registration or commercial formulation is reported Environmental fate, residues, exposure, formulation, application rules and commercial availability

The table’s central distinction is that COT-MM was evaluated for a reversible physiological response, not for the same immediate kill criterion used to judge a conventional herbicide.

Could a growth-pausing herbicide reduce resistance?

Possibly, but the study does not demonstrate it. Williams suggested that restricting growth instead of trying to kill every susceptible plant might reduce the selective pressure that drives resistance. That is a hypothesis about how treatment intensity could influence evolution, not a measured result from the rubber-vine experiment.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Why the hypothesis is plausible

A lethal treatment can leave a population composed of survivors that tolerated or resisted the herbicide. If a treatment instead holds many plants at low growth without killing them, the immediate difference between survivors and susceptible plants could be smaller. In principle, that might alter the conditions under which resistance-associated traits are favored.

Why the hypothesis needs direct testing

Living weeds still matter to management. A plant that resumes growth can recover, reproduce or require another intervention. Resistance outcomes would depend on treatment timing, exposure patterns, repeated applications, the genetic diversity of the weed population and interactions with other control methods. None of those long-term variables was established by the laboratory proof of concept.

Has COT-MM been tested in fields or approved for use?

No. The reported work is a laboratory study on one invasive species. The available evidence does not establish field efficacy, crop selectivity, environmental safety, environmental breakdown, residue behavior, registration or commercial availability.

What the current evidence does show

  • The molecule can be synthesized by modifying metsulfuron methyl.
  • It retains ALS inhibition but binds less strongly according to kinetic, binding and docking analyses.
  • It temporarily suppressed rubber-vine growth in the reported laboratory tests.
  • It produced lower rubber-vine mortality than metsulfuron methyl.

What the current evidence does not show

  • That COT-MM controls rubber vine in pasture, rangeland, forest or other field settings.
  • That crops would tolerate an application or that desirable native plants would be protected.
  • That the compound is safe for soil organisms, wildlife, water or people.
  • That a registered formulation, label, supply chain or consumer product exists.

What experiments are needed before this could become a management tool?

The next studies would need to test whether the temporary response is useful outside the original laboratory conditions and whether its trade-offs are acceptable.

Measure recovery and reproduction

Follow treated rubber vines beyond the first recovery point and record regrowth, flowering, seed production and the effect of repeated treatments. A growth pause that looks substantial at six or eight weeks could have a very different management value if plants rapidly reproduce afterward.

Test dose, timing and formulation

Determine how concentration, plant size, season, rainfall and application interval change the duration of suppression. The reported sources do not provide a field dose or a formulation, so those parameters cannot be inferred from the proof of concept.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Examine species and crop responses

Screen the compound against multiple weed species, crop varieties and native plants. Because ALS sequences and uptake can differ among plants, activity on rubber vine cannot establish either broad weed control or crop safety.

Track environmental fate and exposure

Measure persistence, breakdown products, movement through soil and water, and effects on non-target organisms. These data are prerequisites for a meaningful environmental-risk assessment and are not supplied by the laboratory report.

Test resistance directly

Use controlled selection experiments and population genetics to compare how quickly resistance appears under lethal versus herbistatic regimes. The resistance-reduction idea should remain clearly labeled a hypothesis until such evidence exists.

What should readers take from this study?

COT-MM shows that changing a herbicide’s molecular shape can change the biological outcome from stronger killing toward temporary growth suppression. Its cyclooctatetraene ring weakens and destabilizes ALS binding enough to pause rubber-vine growth, while the parent metsulfuron methyl causes greater mortality.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

The study opens a research direction rather than delivering a ready-to-use weed treatment. Herbistasis could eventually become one tool for managing invasive plants or exploring resistance strategies, but claims about field control, crop safety, environmental impact or regulatory approval would require evidence that this experiment did not provide.

Frequently Asked Questions

Why are the reported six-week and eight-week timings different?

The University of Queensland summary describes six weeks in which rubber-vine growth was stopped. The more detailed account reported by Chemistry World uses the return of vertical growth at eight weeks as its marker, so the figures refer to different observations rather than a demonstrated contradiction.

Is COT-MM available as a garden or agricultural product?

No retail formulation or commercial availability is reported for COT-MM. It is an experimental research compound, so readers should not treat the laboratory result as a use recommendation or look for it under an approved product label.

Does lower mortality mean the treatment failed?

Not under a herbistatic objective. The experiment was testing whether growth could be held back temporarily, so lower mortality is consistent with the intended effect; it would be a limitation if the management goal were permanent eradication from one application.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

What does COT-MM stand for?

COT-MM is short for cyclooctatetraene-metsulfuron methyl. The name reflects the design change: a cyclooctatetraene ring was substituted into the metsulfuron methyl structure while retaining its ALS-targeting framework.

Who published the underlying chemical study?

H. Xing and colleagues published the work in Chemical Science in 2025, volume 16, page 649, under DOI 10.1039/d4sc06923d. The reported experiment was led by Craig Williams’s team at The University of Queensland.
