Researchers can share an AI-designed protein more safely by treating release as a staged decision, not a choice between publishing everything and publishing nothing. First inventory the sequence and accompanying materials, assess how they could enable harmful use, obtain the relevant institutional review, and screen both sequences and recipients when synthesis or transfer is involved. Then release only the information needed for the legitimate scientific purpose, using open publication, controlled access, redaction, or delay as appropriate. Record the decision and reassess it if the design, recipient, or risk context changes.
What counts as an AI-designed protein release?
A release is more than posting an amino-acid sequence. A research package may include a predicted or experimentally resolved structure, claims about function, expression-system information, protocols, optimization data, troubleshooting notes, and metadata. Any of these can make a design easier to reproduce or use, so the disclosure decision should cover the whole package rather than the sequence alone.
Start with an asset inventory. For each item, note its scientific purpose, who needs it, whether it is necessary for validation, and whether it materially lowers a barrier to harmful synthesis, expression, optimization, or evasion of screening. A sequence may be difficult to interpret without context; a protocol or host detail could supply that context. Conversely, withholding too much can make legitimate results impossible to assess. The goal is not maximum secrecy but a defensible balance between reproducibility and foreseeable risk.
- Design assets: amino-acid sequence, structure files, model outputs, and the stated function or intended application.
- Execution assets: expression-system information, experimental procedures, optimization details, and troubleshooting material.
- Distribution assets: recipient information, repository settings, access records, and version history.
- Context: the scientific purpose, relevant expertise and equipment, plausible misuse pathways, and uncertainties in the assessment.
Keep a clear distinction between what has been experimentally demonstrated and what is only predicted or claimed. That distinction matters both for sound scientific communication and for assessing how operationally useful the package is.

How should a team assess misuse risk?
Assess the design as a potential capability, not just as a novel sequence. The review should consider plausible harmful functions, how readily the design could be synthesized or expressed, whether accompanying information makes use easier, what expertise and equipment are needed, and the severity of a credible harmful outcome. Record uncertainty instead of disguising it as a precise score.
Ask questions about the complete package
- Function: What activity is claimed, and how strong is the evidence supporting that claim?
- Feasibility: Could the sequence or its encoded product be readily obtained, expressed, or adapted? Which materials in the package change that answer?
- Operational value: Would a protocol, optimization parameter, host detail, or troubleshooting note significantly reduce trial and error?
- Screening and evasion: Is there a credible concern that the sequence or a related construct could be used to evade screening? What evidence supports or limits that concern?
- Consequence: If the design were misused, what is the plausible severity, and who could be affected?
- Recipient and setting: Is the proposed recipient, institution, and stated purpose appropriate for the requested level of access?
Risk is not determined by novelty or AI involvement alone. The practical question is whether this particular design, with these accompanying materials, could meaningfully enable harmful activity, and whether the proposed controls fit that risk. Involve people with relevant scientific, biosafety, security, and governance expertise; a researcher should not have to resolve a difficult dual-use question alone.
Interpret screening evidence cautiously
AI-generated sequences complicate screening because sequence similarity alone may not reliably capture function. NIST’s 2026 program page describes 2025 evaluations involving 30,000 enzyme variants and a dataset of approximately 40,000 AI-generated enzyme variants. NIST reported that synthetic homologs with predicted structures similar to native templates did not necessarily retain activity; reliable rewriting that both maintained activity and evaded screening was not demonstrated in that evaluation. That is a bounded finding from the described work, not proof that screening is unnecessary or that every evasion concern is resolved.

The practical implication is to use screening as one layer of review, not as a substitute for assessing function claims, accompanying information, recipient legitimacy, or institutional context. Where a screening result is unclear, pause the transfer and route the question to the appropriate institutional or provider contact rather than treating an automated result as a final risk decision.
Who should review the decision before release?
Route the proposed release through the review functions that apply at your institution and in the relevant jurisdictions. Depending on the asset and transfer, this may include institutional biosafety, dual-use review, legal or export-control review, and information security. Assign a decision owner who can collect advice, document the rationale, and confirm the release conditions.

Do not assume that one approval automatically covers every subsequent action. A paper, a repository deposit, a transfer to a synthesis provider, and a direct handoff to a collaborator may involve different information, recipients, or controls. The review should identify which version of the package is approved, where it may be shared, and what would trigger renewed consideration.
- Prepare a release inventory. List each sequence, file, protocol, claim, and metadata field proposed for release, and identify the intended scientific use.
- Write down the risk questions. Describe plausible misuse pathways, uncertainty, relevant screening concerns, and the effect of releasing each operational detail.
- Consult the responsible institutional offices. Request biosafety or dual-use review and add legal, export-control, or information-security review when applicable.
- Name the decision owner. Record who is responsible for accepting, rejecting, or conditioning the release and who handles questions or incidents afterward.
- Set the approved scope and review conditions. Specify the release mode, version, permitted recipients or uses where relevant, and when temporary restrictions will be reconsidered.
Requirements vary by country, institution, funder, and the specific protein or sequence. The U.S. Department of Health and Human Services (HHS) says its 2024 synthetic nucleic acid screening framework is being revised or replaced under the May 5, 2025 executive order. Researchers should verify the currently applicable federal guidance and local procedures at the time of release rather than relying on an older summary.

What screening is needed before synthesis or transfer?
When a design is going to a synthesis provider or another recipient, consider both what is being transferred and who is receiving it. HHS guidance recommends sequence screening, customer and recipient legitimacy checks, responsible handling and transfer, and records. The Federal Register framework also describes following up on sequences of concern and documenting screening, follow-up, and final disposition.
Protein designs are often discussed as amino-acid sequences, while the cited screening guidance addresses synthetic nucleic acid sequences. Coordinate with the provider or institutional screening process to determine what sequence representation and associated information are relevant for the proposed order or transfer. Do not assume that checking a protein sequence in isolation satisfies the screening process for a nucleic-acid construct, or that a successful sequence screen establishes the recipient’s legitimacy.
- Identify the transfer pathway. Determine whether material is being ordered from a provider, sent to a collaborator, placed in a repository, or shared by another route.
- Use the appropriate screening channel. Follow the provider’s and institution’s applicable process for the sequence and construct involved. If the process raises a concern, stop and seek review.
- Verify recipient legitimacy. Confirm the identity, institutional affiliation, purpose, and authorization appropriate to the transaction. Resolve inconsistencies before proceeding.
- Document follow-up and disposition. Retain the outcome, any additional inquiries, the decision reached, and what was ultimately transferred or withheld.
- Transfer responsibly. Use the approved recipient and channel, and provide only the release package authorized for that transfer.
NIST describes screening as a combination of standards, databases, tools, testing, and secure-transfer capacity for identifying and tracking sequences of concern. Its program page reports monthly baseline testing beginning in August 2025 and collaborations with IBBIS, MITRE, and SecureDNA. These are descriptions of NIST program activity, not a guarantee that every provider uses the same tool or that any single screening result is conclusive.

Which release mode fits the scientific purpose?
Choose the least restrictive mode that still addresses the identified risk and allows the legitimate work to proceed. Open publication is only one option: controlled access, vetted-recipient transfer, partial release, and delay can preserve useful collaboration while limiting immediate exposure. Compare each option by scientific benefit, reproducibility, misuse plausibility and severity, reversibility, access controls, monitoring, and the time available for review.

|---|---|---|
| Open publication | When the review supports broad disclosure and the information is appropriate for unrestricted scientific use. | Disclosure is difficult to reverse once files are copied or redistributed. |
| Controlled-access repository | When researchers need a shared resource but access should be limited to approved users or purposes. | Access controls and monitoring must be sufficiently reliable for the risk, and legitimate users may face extra friction. |
| Vetted-recipient transfer | When specific collaborators need details that are not appropriate for unrestricted release. | Recipient verification, transfer records, and clear handling expectations become especially important. |
| Partial release or redaction | When some results support evaluation but selected operational details would materially increase misuse capability. | Redaction can reduce reproducibility or make scientific claims harder to independently assess. |
| Delayed release | When a limited period would enable additional review, safer dissemination arrangements, or resolution of a concern. | A delay needs a responsible owner and a review date; otherwise temporary restriction can become indefinite by default. |
Make the decision at the level of individual materials where possible. A paper may report scientific findings while a repository holds a restricted file; a collaborator may receive a validated subset while additional operational details remain controlled. Explain the scientific reason for any restriction to affected collaborators and reviewers, without distributing the sensitive information through the explanation itself.
How can a team preserve reproducibility without releasing every detail?
Separate information needed to evaluate the scientific claim from information that materially enables execution or optimization. A responsible release can retain enough evidence for readers to judge what was done and what the results support, while controlling details that create a disproportionate operational benefit for misuse.
For each withheld or restricted item, document why it is sensitive, what scientific purpose it serves, who needs it, and how that need can be met safely. Consider whether a summary, a validated result, access through a controlled repository, or transfer to a vetted collaborator would support the legitimate purpose with less exposure. Review the effect of redaction on interpretability: if a reader cannot tell whether a central claim is substantiated, the release may need a different design or further review.
Do not treat metadata as harmless by default. Descriptive fields, file names, annotations, links between versions, or protocol context can disclose information that is absent from the headline sequence. Review the complete release package and repository settings before making it public or granting access.

What records and follow-up should accompany the release?
Maintain a decision record that makes the release understandable later, including when the design, recipient, or threat information changes. HHS guidance calls for documentation of screening, follow-up, transfers, and final disposition; a research team can connect those records to its institutional review and release history.
- Release inventory, including the version of each sequence, structure, protocol, and associated file reviewed.
- Risk rationale, scientific purpose, uncertainties, and the reason for the selected release mode.
- Institutional approvals, advice received, decision owner, and any conditions imposed.
- Screening outcome, recipient-verification steps, follow-up, transfer details, and final disposition where applicable.
- Recipient list, repository access settings, access logs, version history, and an incident contact.
- A review date for temporary controls and a record of changes made after reassessment.
Reopen the assessment if the sequence or protocol changes, new recipients are added, access settings change, new evidence alters the function claim, or credible threat information emerges. A release decision applies to the version and circumstances reviewed; it should not silently carry over to a materially different package or transfer.
What current guidance should researchers check?
Several public resources are relevant, but their scope and currency differ. NIST’s Biosecurity for Synthetic Nucleic Acid Sequences page describes technical and program work on screening; HHS and the Federal Register describe screening and transfer expectations; the International Gene Synthesis Consortium (IGSC) lists updated industry resources; and the Responsible Biodesign Workshop 2025 recommendations call for next-generation screening algorithms for AI-generated sequences of concern and collaboration with synthesis providers.
The IGSC resource listing identifies a 2026 Harmonized Screening Protocol v3.1 alongside related supply-chain and screening resources. That update is a reminder to check the version currently used by the relevant provider rather than assuming an older protocol remains current. Likewise, the HHS page notes revision activity following the May 5, 2025 executive order. Confirm the current guidance, institutional procedure, and provider requirements immediately before a consequential release or order.

Frequently Asked Questions
Does an AI-designed protein need a different review from a conventionally designed protein?
The source material does not establish a separate universal review category solely because AI was used. The relevant assessment is the design’s plausible function and misuse potential, the release package, and the transfer context; AI-generated sequences do add challenges for screening methods.
Can I publish a protein sequence but keep the structure file private?
That can be a possible partial-release arrangement, but whether it is appropriate depends on how much the structure contributes to reproducibility or operational usefulness in this case. Document the scientific and risk rationale for treating the two assets differently.
Who is responsible if a collaborator requests restricted design files?
Responsibility depends on institutional policy, the transfer arrangement, and applicable law, so the researcher should not infer it from authorship alone. Identify a decision owner through the institution’s review process before sending the files.
