A supplier list is not a supply-chain map. Two companies on purchase orders may buy the same active ingredient from one factory, depend on the same starting material, or use the same transport corridor. To find a genuine single-source dependency, trace every critical material from its upstream starting materials through intermediates, API manufacture, finished dosage form, packaging, and distribution, then count independently usable manufacturing sources rather than vendor names.

The U.S. Food and Drug Administration (FDA) expects records that identify suppliers, actual manufacturers, material use, batches, and distribution history. Its Q7A API Good Manufacturing Practice guidance says that when a critical-material supplier is not the manufacturer, the manufacturer’s name and address should be known. That distinction is the foundation of a defensible dependency assessment.

What a pharmaceutical supplier map must show

A useful map connects four kinds of records: materials, organizations, physical sites, and products or processes. Each edge in the map should answer what moved, who made it, where it was made, and which medicine or process consumed it.

  • Material: the chemical, solvent, reagent, key starting material, intermediate, API, inactive ingredient, or packaging component.
  • Organization: the supplier of record, broker, distributor, legal manufacturer, contract manufacturer, and parent entity where relevant.
  • Site: the physical plant, address, production line or campus, and country or region.
  • Product or process: the API batch, finished dosage form, market authorization, drug presentation, or manufacturing step that depends on the material.

Keep the distributor or broker separate from the original manufacturer. A purchase-order vendor can be an intermediary, not an independent source. A source is operationally “single-source” for the defined scope when only one independently usable, qualified source exists at the relevant tier. State whether the scope is one ingredient, one manufacturing site, one market, or one finished drug; the result can differ by scope.

The map should extend upstream and downstream. The U.S. Government Publishing Office’s Mapping of the Supply Chain describes the progression from raw materials and intermediates to API and finished doses. Packaging is part of the dependency picture when a container-closure system is needed to preserve product quality.

Map every tier from starting material to finished medicine

1. Raw materials and key starting materials

Begin with solvents, reagents, process chemicals, and regulatory or key starting materials entering API production. Record the material name, internal code, specification, grade, source organization, and manufacturing site when known. Note whether the material is made in-house, purchased directly, or obtained through a trader.

For each shipment or batch, retain the manufacturer, identity and quantity, supplier name and control or identification number if known, receipt number and date, test or release results, and the records that show where the material was used. These are the kinds of traceability details specified in FDA Q7A.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

2. Critical intermediates

List every intermediate whose loss would stop or materially restrict API production. An intermediate is not the API: it is a material undergoing further molecular change or purification before becoming the drug substance. Identify the organization and site that make each intermediate, the process step, batch linkage, and any upstream starting material that could create a shared dependency.

3. API or drug-substance manufacture

Connect each API to its actual manufacturing entity and site, not merely to the distributor named on a quality agreement or invoice. Capture manufacturing lines or suites where they affect independence, batch records, release status, approved markets, and the finished products that use the API. If a contract manufacturer performs only one step, represent that step separately so that a second API supplier is not mistaken for a second complete source.

4. Finished dosage form and inactive ingredients

Map the site that combines the API with inactive ingredients and converts it into tablets, capsules, injectables, liquids, or another finished form. Include critical excipients, sterilization services, specialized processing, and any single-use or device component whose absence would halt production. A finished-dose manufacturer can be a separate dependency even when the API has several qualified sources.

5. Container-closure and packaging materials

Identify vials, stoppers, syringes, blister films, caps, labels, and other components that are essential to maintaining quality or regulatory compliance. Record the actual maker, site, approved specification, and alternative qualification status. Packaging concentration can create a finished-product single source even when ingredients are diversified.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

6. Distribution and transport

Record every distributor, warehouse, freight route, and handoff to the next tier while preserving the distinction between movement and manufacture. FDA Q7A calls for traceability of API distributors and intermediaries, including original-manufacturer identity and transportation or distribution records. Include temperature control, ports, customs steps, and route constraints when they can interrupt supply.

Use a dependency record, not a vendor spreadsheet

A flat vendor list loses relationships. Use one record per material-and-site relationship, with stable identifiers for legal entities and facilities. The following compact schema works in a database, graph tool, or controlled spreadsheet.

Field What to capture
Material and tier Name, internal code, specification, and whether it is a starting material, intermediate, API, excipient, or packaging component.
Supplier of record Company on the purchase order, quality agreement, or certificate.
Actual manufacturer Legal manufacturing entity and confirmation of whether the supplier is an intermediary.
Site and location Physical address, production campus or line, country or region, and approved operations.
Upstream dependencies Critical starting materials, intermediates, utilities, specialist processes, and shared facilities.
Downstream drug or process API batch, dosage form, market, manufacturing step, and products consuming the material.
Qualification status Approved, conditionally approved, under qualification, unqualified, or rejected; include scope and restrictions.
Evidence date Date of supplier confirmation, audit, batch record, certificate, regulatory filing, or other supporting evidence.
Mitigation owner Named procurement, quality, regulatory, manufacturing, or logistics owner responsible for the next action.

Store aliases, former legal names, site codes, and parent-company relationships so that one manufacturer cannot appear as several independent sources. Keep evidence attached to the record rather than relying on an unverified name copied from a catalog.

Step-by-step method for building the map

  1. Define the assessment boundary. State the product, strength, presentation, market, time period, and failure event being tested. Decide whether the question concerns API availability, finished-dose output, or a specific packaging or logistics constraint.
  2. Inventory critical inputs. Start with the bill of materials, process descriptions, batch records, deviations, and packaging specifications. Mark inputs whose loss would stop production, force a long requalification, or create a patient-impacting shortage.
  3. Request manufacturer-level evidence. Ask each supplier to identify the actual manufacturing site, upstream critical materials, approved capacity, lead time, and distribution route. Reconcile the answer with quality agreements, certificates, audit reports, regulatory filings, receiving records, and batch documentation.
  4. Trace lots through the process. Link the received lot or batch to test and release evidence, the manufacturing step that consumed it, the resulting API or finished-dose batch, and the market in which that batch was released. This establishes material use rather than merely asserting a commercial relationship.
  5. Create the graph. Represent materials, organizations, sites, products, routes, and process steps as nodes; represent “made at,” “supplied by,” “consumed by,” “shipped through,” and “depends on” as edges. Keep an intermediary node between a distributor and the actual manufacturer.
  6. Normalize and deduplicate. Standardize legal-entity names, addresses, site identifiers, material codes, and transliterations. Investigate apparent duplicates and apparent alternatives until you know whether they are separate facilities or aliases.
  7. Test independence. Count actual manufacturers and sites, then inspect common starting materials, specialist equipment, utilities, parent production networks, warehouses, ports, and transport corridors. Record the evidence for each shared dependency.
  8. Check usability. An alternative counts only if it is qualified for the product and market, has an acceptable specification and quality history, can supply within the required lead time, and can meet applicable regulatory conditions. Label a merely possible source as unqualified rather than as a backup.
  9. Assign risk and action. Give each dependency an owner, evidence date, trigger for rechecking, mitigation, and target completion date. Examples include a second-site qualification, safety stock, a route change, a technical agreement, or a regulatory filing.
  10. Refresh the map. Revalidate it after supplier changes, site transfers, deviations, major capacity changes, regulatory submissions, geopolitical events, or material substitutions. A static map becomes misleading as soon as a site or upstream input changes.

How to expose hidden single-source dependencies

Count manufacturing sources, not purchase-order names

If three distributors all identify one factory as the manufacturer, the product has one manufacturing source. If two factories are owned by the same organization but use one critical intermediate plant, they may be two sites with one upstream bottleneck. Report both facts instead of collapsing them into a single score.

Look for shared sites and production networks

Compare addresses, plant identifiers, production lines, utilities, sterilization providers, and contract-process partners. A nominally different company may be a sister entity on the same campus or part of a network that shares a constrained operation. The shared-dependency test is a practical risk-analysis method, not an FDA-prescribed scoring formula.

Trace common upstream inputs

Follow each alternative API or intermediate back to critical starting materials. Two API manufacturers can remain coupled if both buy the same specialized reagent, precursor, fermentation input, or purification service from one upstream source. Capture that relationship even when the final manufacturing sites are in different countries.

Check logistics convergence

Map ports, airports, cold-chain hubs, customs steps, carriers, and roads. Separate geographic diversity from route diversity: factories on different continents can still depend on one port or one temperature-controlled lane. Geography is one risk dimension, not proof by itself that a source is fragile or resilient.

Separate potential from qualified capacity

A company that could theoretically make a material is not an immediately usable source. Record qualification status, available capacity, lead time, technical-transfer work, stability data, audit findings, and market authorization. FDA notes that new sources of raw materials, regulatory starting materials, or active pharmaceutical materials may require FDA approval to increase supply; the requirement depends on the product and change.

Assess concentration with a risk register

Use the graph to populate a risk register. Rank the consequence of losing each node and the time required to restore supply, rather than assigning a number solely from supplier count.

Risk axis Questions to answer Evidence or mitigation
Independent qualified sources How many actual manufacturers and usable sites remain after shared dependencies are removed? Manufacturer confirmations, audits, qualification files, and approved-source lists.
Substitutability Can another source meet the same specification, process capability, and market requirements? Technical-transfer plan, comparability work, validation, stability data, and change-control assessment.
Site and geographic concentration Do sources share a campus, region, utility, upstream site, port, or transport lane? Site maps, route records, business-continuity plans, and alternative lanes.
Quality and reliability Are there recurring deviations, recalls, inspection findings, or low process capability? Quality metrics, audit results, corrective-action status, and supplier performance history.
Capacity and demand Can the source increase output, and how does demand volatility affect the allocation? Validated capacity, lead times, allocation rules, forecasts, and contingency contracts.
Inventory and transport exposure How many days of usable stock cover demand, and what route or temperature risks exist? Lot-level inventory, expiry profile, transit times, buffer policy, and monitored lanes.
Clinical and operational criticality What is the patient impact if this medicine or presentation is interrupted? Therapeutic alternatives, substitution limits, treatment volume, and escalation criteria.
Regulatory recovery What filings, approvals, inspections, or change controls are needed for a replacement? Regulatory strategy, submission timeline, agency correspondence, and implementation owner.

These dimensions reflect disruption factors identified by the FDA, including manufacturing and quality problems, economic incentives, demand increases, limited suppliers, foreign dependence, weather and environmental events, public-health responses, transportation, and geopolitical situations. The FDA says shortage response depends on the cause, patient-care impact, and public-health risk; no single concentration metric captures all three.

Regulatory change control is part of the dependency decision

A technically capable replacement cannot be treated as production-ready until quality and regulatory work is complete. FDA Q7A states: Changing the source of supply of critical raw materials should be treated according to Section 13, Change Control. Apply that instruction to supplier changes, site transfers, new starting-material manufacturers, and material substitutions. Determine whether the change affects specifications, validated processes, filings, stability, sterility assurance, or product release testing.

For each proposed alternate, document the gap between the current source and the replacement: analytical comparability, process performance, impurity profile, safety information, audit outcome, capacity, and transport conditions. Then obtain the approvals or notifications required for the relevant product and market before counting the source as independently usable.

What shortage statistics do—and do not—tell you

The FDA Drug Shortage Task Force’s report, Drug Shortages: Root Causes and Potential Solutions, identifies three broad root causes: weak incentives to produce less-profitable medicines, inadequate market reward for mature quality systems, and logistical or regulatory challenges that make recovery difficult.

For drugs that experienced shortages between 2013 and 2017, the FDA’s February 2020 recalculation found that 42 percent had a significant production increase. In that report, “significant production increase” meant new suppliers entering and/or existing suppliers increasing output enough to restore at least half of the unavailable quantity. Thirty percent had their quantity fully restored to pre-shortage levels. These are retrospective figures about that shortage cohort, not a success rate for supplier mapping or a guarantee that a second source will restore a particular product.

Use public shortage data as a signal, not as the map

The FDA’s openFDA Drug Shortages data can reveal that a product, presentation, or market is under pressure. The agency says manufacturers provide most shortage information and that the database reflects situations known to FDA. Public entries generally do not establish every confidential supplier relationship, upstream source, approved site, current capacity, or product-specific qualification status.

For an actual dependency decision, reconcile public signals with purchasing records, receiving and batch records, supplier audits, quality agreements, regulatory filings, direct manufacturer confirmation, and current capacity evidence. The FDA also states in its Supply Chain: FDA’s Role material that it cannot ensure continuity or resilience of entire industrial supply chains and cannot require companies to produce products or report information that the law does not require.

Do not use country of origin as a shortcut for resilience. A source in a supposedly low-risk country can share an upstream precursor or route with another source, while a geographically distant source may have genuinely independent materials, sites, and logistics. The graph and its evidence should support the conclusion.

Operational checklist for a defensible assessment

  • Every critical material has an identified actual manufacturer and physical site, or an explicitly documented evidence gap.
  • Intermediaries, brokers, distributors, contract steps, and legal manufacturers are represented as different entities.
  • Raw materials, intermediates, APIs, finished dosage forms, inactive ingredients, container-closure components, and distribution routes are connected.
  • Each lot or batch can be traced from receipt and testing to downstream use.
  • Aliases, former names, parent companies, and site codes have been normalized and reviewed for duplicates.
  • Alternative names have been tested for shared sites, starting materials, utilities, specialist processes, ports, warehouses, and carriers.
  • Qualification status, evidence date, lead time, capacity, and market restrictions are recorded for every proposed alternative.
  • Regulatory approval and change-control requirements are identified before an alternate is counted as usable.
  • Risk owners, mitigation actions, trigger events, and review dates are assigned.
  • Public shortage information is treated as an early-warning input and reconciled with confidential company records.

How to maintain the map over time

Make supplier mapping part of change management rather than a one-time exercise. Trigger a review when a supplier changes legal entity, a manufacturing site moves, a critical material specification changes, a contract manufacturer is added, a major deviation occurs, capacity is allocated, a transport lane is disrupted, or a new regulatory filing is submitted. Require procurement, quality, manufacturing, regulatory, and logistics teams to update the same controlled record so that a commercial change cannot silently invalidate the dependency analysis.

A mature map does not merely say that a product has “two suppliers.” It shows whether those suppliers are independent at the manufacturing, upstream-material, site, and route levels; whether each is qualified for the product and market; and how long replacement would take. That level of evidence is what turns a vendor list into a usable supply-continuity decision.
