Specialist spectroscopy can distinguish genuine artesunate tablets from counterfeits without opening their blister packs. In the studies behind Simon Hadlington’s 29 October 2007 Chemistry World report, Raman spectroscopy, spatially offset Raman spectroscopy (SORS), and attenuated total reflection Fourier-transform infrared (ATR-FTIR) imaging supplied different kinds of evidence: SORS read chemical information through packaging and at different depths, while ATR-FTIR imaging showed where active ingredients and excipients were distributed across a tablet surface. Follow-up work paired FTIR imaging with desorption electrospray ionization mass spectrometry (DESI-MS), and a 2008 study evaluated a hand-held Raman instrument for field screening.

This is forensic and analytical research, not a home test for medicines. The historical studies show how laboratories can investigate suspected counterfeit artesunate; they do not establish that a consumer device can authenticate a treatment, that any instrument is currently regulator-approved, or that a scan can replace a licensed medicine supply chain.

Why counterfeit artesunate requires more than a visual inspection

Artesunate is an antimalarial active ingredient used in treatments in malaria-endemic regions. A tablet can look convincing while containing the wrong ingredient, too little of the right ingredient, an unsuitable substitute, or an uneven mixture. Packaging may also conceal the tablet from direct inspection. Those problems make authentication an analytical question: investigators need to determine what chemicals are present and how they are arranged, preferably without destroying the evidence.

Conventional visual checks can still find obvious defects such as misspellings, damaged seals, inconsistent logos, or unusual tablet colour. They cannot establish chemical identity. The research associated with the 2007 report instead treats a tablet as a measurable chemical object. Light or ions interact with the sample, producing signals that can be compared with genuine material or interpreted by an analyst.

What the 2007 report was describing

Simon Hadlington’s article, “Forging ahead of the counterfeiters,” was published by Chemistry World on 29 October 2007. Its indexed description identifies Raman and infrared spectroscopy being used to spot fake malaria tablets. Imperial College London’s vibrational-spectroscopy group identifies its FTIR-imaging work on fake antimalarial drugs as the application discussed in that article.

The most detailed related evidence comes from a 2007 paper by Ricci and colleagues on genuine and fake artesunate tablets. The researchers combined ordinary Raman spectroscopy, SORS, and ATR-FTIR imaging. These techniques are complementary rather than interchangeable: one can examine a packaged tablet and obtain depth-related chemical information, while another can make a map of chemical domains at the surface.

The report should therefore be read as an account of laboratory and investigative capability. It is not a claim that a single scan, a phone attachment, or a retail gadget can certify a medicine.

How the spectroscopy methods work together

Raman spectroscopy: a molecular fingerprint

Raman spectroscopy illuminates a sample with a laser and measures the very small shift in wavelength produced when light interacts with molecular vibrations. The resulting spectrum can contain bands associated with an active pharmaceutical ingredient, fillers, binders, coatings, or other components. In the artesunate work, Raman spectroscopy was one part of a combined characterization strategy for genuine and fake tablets.

Raman is valuable because it can provide chemically specific information with little or no sample preparation. A spectrum is not automatically a verdict, however. Fluorescence, tablet coatings, mixtures, instrument settings, and the quality of the reference library can all affect interpretation. The cited studies report analytical use, not a universal accuracy percentage.

Spatially offset Raman spectroscopy (SORS): looking through packaging

SORS collects Raman signals at a position spatially separated from the point where the laser enters. That geometry helps separate signals from the outer package and the contents beneath it. In the artesunate study, SORS is described as depth-resolving and non-invasive, providing composition information through several types of packaging, including blister packs.

That capability matters operationally. An investigator can examine a sealed package, preserve the tablet, and screen material without first transferring it to a laboratory container. The measurement can also provide information from different depths rather than being limited to the topmost visible layer. Packaging type, thickness, colour, surface finish, and the tablet’s own fluorescence still influence what can be measured, so an analyst must validate the method for the packaging encountered in the case.

ATR-FTIR imaging: mapping the tablet surface

Fourier-transform infrared (FTIR) spectroscopy records how a sample absorbs infrared wavelengths. In attenuated total reflection (ATR), the tablet contacts an infrared-transmitting crystal and the evanescent wave probes a shallow region at the contact surface. FTIR imaging collects spectra across many positions, turning those measurements into a chemical map.

The Ricci study reports that ATR-FTIR imaging mapped surface domains of active ingredients and excipients, with high sensitivity and enhanced spatial resolution. A map can reveal whether components are evenly distributed, concentrated in islands, or arranged in a pattern inconsistent with a genuine reference. That spatial evidence is different from a single bulk spectrum: two tablets might have similar overall ingredients but differ in how those ingredients are distributed.

Because ATR requires contact, it is not the same kind of through-package test as SORS. The tablet may need to be removed from its blister and positioned against the ATR accessory, and the measured region is near the surface in contact with the crystal. The method is consequently powerful for laboratory characterization but not a simple non-contact consumer check.

FTIR imaging plus DESI-MS: distribution and identity

A separate 2007 paper combined FTIR imaging with desorption electrospray ionization linear ion-trap mass spectrometry. In that workflow, FTIR imaging detected where components were distributed, while DESI-MS confirmed component identities. Mass spectrometry adds information based on ions and mass-to-charge ratios, helping analysts verify the chemical identity suggested by the infrared map.

The pairing illustrates a general forensic principle: orthogonal methods reduce the chance that one ambiguous signal is mistaken for proof. FTIR contributes a spatial picture; DESI-MS supplies identity confirmation. The paper reports those analytical roles, not a consumer protocol or a guaranteed pass/fail threshold.

Method comparison

Method Role reported in the artesunate research Practical strength Important qualification
Raman spectroscopy Used with SORS and ATR-FTIR imaging to characterize genuine and fake artesunate tablets. Chemically specific molecular spectra. Interpretation depends on fluorescence, mixtures, instrument conditions, and suitable references.
Spatially offset Raman spectroscopy (SORS) Provided depth-resolving, non-invasive composition information through several packaging types. Can examine packaged tablets, including through blister material. Packaging and sample properties affect signal quality; validation is case-specific.
ATR-FTIR imaging Mapped surface domains of active ingredients and excipients, with high sensitivity and enhanced spatial resolution as described in the abstract. Shows spatial distribution rather than only an averaged spectrum. Requires contact with an ATR crystal and probes a shallow surface region.
FTIR imaging with DESI-MS FTIR imaging detected component distributions; DESI-MS confirmed component identities. Combines spatial information with an independent identity check. Requires specialist laboratory instrumentation and expert interpretation.

What a real laboratory examination can look like

The publications do not prescribe one universal operating procedure, but their methods imply a staged analytical workflow. Each stage answers a different question and creates evidence for the next one.

  1. Preserve the sample and record its context. Investigators document the package, batch information, seals, tablet appearance, and chain of custody. A sealed blister may be retained for a non-invasive SORS measurement.
  2. Screen without opening when possible. SORS can collect composition information through the packaging and provide depth-related data. The result is compared with spectra from authenticated material or an established reference set.
  3. Collect complementary Raman data. Conventional Raman measurements can characterize accessible tablet material and help identify spectral differences between genuine and suspected counterfeit samples.
  4. Map the surface with ATR-FTIR imaging. If laboratory examination is warranted, the tablet is placed against the ATR interface. The instrument records spectra across a grid and produces maps showing domains associated with active ingredients and excipients.
  5. Confirm identities with an independent technique. In the related 2007 workflow, DESI-MS was used to confirm component identities after FTIR imaging revealed their distributions.
  6. Interpret all evidence together. Analysts compare spectra, chemical maps, packaging effects, reference tablets, and control measurements. A suspicious result should trigger confirmatory testing and supply-chain investigation rather than an unsupported public accusation.

This sequence also explains why no single method is described as universally sufficient. Through-package screening, surface mapping, and molecular confirmation answer different analytical questions.

What the 2008 hand-held Raman study adds

In 2008, Ricci and colleagues assessed a portable Raman instrument for in-situ screening of potentially counterfeit artesunate tablets. The work was validated using FT-Raman spectroscopy and ambient mass spectrometry. “In situ” indicates an attempt to bring screening closer to where samples are encountered, rather than relying exclusively on a full laboratory setup.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

The study is historical evidence that portability was being investigated, not proof that a current hand-held device is commercially available, regulator-accepted, or suitable for the public. Portable instruments still require calibration, reference materials, trained operators, documented decision rules, and a pathway for confirmatory testing. A field screen can prioritize samples for laboratory analysis; it should not be treated as a medical authorization to take, sell, or distribute a product.

Why these techniques are complementary instead of competing

Counterfeit tablets can differ from genuine ones in several ways at once. The wrong active ingredient is an identity problem. An incorrect amount is a quantitative problem. Poor mixing is a spatial-distribution problem. Packaging interference is a measurement problem. No method in the cited work is presented as solving every one of these alone.

  • Need information through a sealed blister? SORS is the method specifically described as providing composition information through packaging.
  • Need to see where ingredients sit on a tablet surface? ATR-FTIR imaging supplies spatial maps of active-ingredient and excipient domains.
  • Need an independent identity check? The FTIR-imaging and DESI-MS study used mass spectrometry to confirm component identities.
  • Need a field-oriented screen? The 2008 hand-held Raman assessment explored portability, while retaining validation by laboratory Raman and ambient mass spectrometry.

These are analytical roles, not a ranking in which one instrument replaces all others.

Limits, uncertainty and safe interpretation

No published accuracy figure in the accessible record

The accessible title record and abstracts do not provide a central numerical performance statistic or sample-size figure for the 2007 spectroscopy findings. It would be misleading to supply a made-up sensitivity, specificity, detection limit, or counterfeit-prevalence estimate. The studies support the described capabilities, but the available summaries do not establish a universal pass rate.

Not a consumer authentication test

Raman and infrared instruments are specialist analytical systems. Correct interpretation depends on instrument configuration, sample presentation, reference spectra, operator training, and controls. A consumer should not shine a generic laser device at a tablet and infer that the medicine is genuine. A visually convincing spectrum, an app score, or a marketing claim is not a substitute for validated pharmaceutical analysis.

Not evidence of current regulatory approval

The 2007 and 2008 publications document historical research. They do not establish present-day availability, regulatory acceptance, or clinical validation of a particular instrument. Requirements also differ by jurisdiction and by intended use: investigative screening, quality-control release, customs inspection, and clinical treatment decisions are not the same activity.

A suspicious result needs confirmation

Spectroscopic differences can arise from packaging, coatings, moisture, manufacturing variation, fluorescence, or instrument drift. A credible investigation therefore preserves the sample, repeats measurements under controlled conditions, compares authenticated references, and uses an independent confirmatory method when the decision has legal or public-health consequences.

Independent reader supportYour contribution helps us test, update, and keep practical guides available for everyone.Support on Ko-Fi

What readers should do if they suspect a medicine is counterfeit

Do not rely on a home experiment to decide whether to take a medicine. Keep the packaging and tablets together, avoid altering or discarding them, and contact a pharmacist, national medicines regulator, or other qualified public-health authority for instructions. If someone may have taken a harmful or ineffective antimalarial, seek urgent medical advice; suspected treatment failure in malaria can become an emergency.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

For laboratories and enforcement teams, the practical lesson is different: preserve chain of custody, use validated reference materials, document packaging and measurement conditions, and treat portable screening as a triage step unless the responsible authority has approved it for a defined purpose.

What the 2007–2008 studies established—and what they did not

  • It established a combined analytical approach using Raman spectroscopy, SORS, and ATR-FTIR imaging to characterize genuine and fake artesunate tablets.
  • It described SORS as depth-resolving and non-invasive, with composition information obtainable through several packaging types.
  • It described ATR-FTIR imaging as a way to map surface domains of active ingredients and excipients with high sensitivity and enhanced spatial resolution.
  • It showed, in a separate study, how FTIR imaging and DESI-MS can provide complementary distribution and identity evidence.
  • It documented a 2008 assessment of hand-held Raman screening validated by FT-Raman and ambient mass spectrometry.
  • It did not establish a consumer product, a universal accuracy number, current regulatory approval, or a safe home-authentication procedure.

The lasting contribution is methodological: counterfeit detection becomes stronger when non-invasive depth information, spatial chemical imaging, and independent identity confirmation are combined. That is why the work remains relevant to forensic laboratories and medicine-quality programs, even though the instruments and regulatory context must be evaluated afresh for any current deployment.
