A genome is the complete set of DNA instructions in a cell, including genes and DNA that helps regulate their activity. There is no universal minimum number of genes that every kind of cell needs to live: an engineered bacterium called JCVI-syn3.0 grew as an independently replicating cell in nutrient-rich laboratory conditions with 473 genes, but that figure is not a minimum for human cells or for life in every environment.

What is a genome?

A genome is the full set of DNA instructions in a cell. DNA is built from four chemical bases, usually abbreviated A, T, C and G; their order stores biological information. The National Human Genome Research Institute (NHGRI) defines a genome as the entire set of DNA instructions found in a cell. In humans, a complete account includes DNA in both the nucleus and the mitochondria. NHGRI’s genome glossary explains the term.

A genome is not just a catalogue of genes. It also contains DNA sequences involved in controlling when, where and how genes are active. A gene may encode a protein, or it may encode a functional RNA. Some DNA does not encode either kind of product, but that does not make all such DNA automatically useless: the genome includes more than the instructions for building proteins, and some sequences help regulate those instructions.

That distinction matters whenever someone asks how many genes a cell needs. A gene count is one measure of genome content, not a complete description of the DNA or of what a cell can do. Two gene counts are only meaningfully comparable when it is clear whether they count protein-coding genes, RNA-coding genes or both, and what sort of cell and conditions they describe.

How many genes does a cell need to live?

No single minimum gene count has been established for all cells. The genes a cell can do without depend on its species, its surroundings, the nutrients available to it and whether another organism supplies functions it lacks. “Can live” also needs a definition: surviving, reproducing independently and dividing regularly are not interchangeable measures of cellular fitness.

The best-known engineered example is JCVI-syn3.0, a reduced-genome bacterium developed from a Mycoplasma mycoides derivative. The J. Craig Venter Institute’s 2016 account reports that the viable strain had 473 genes in 531,560 base pairs: 438 protein-coding genes and 35 RNA-coding genes. It grew as an independently replicating cell in nutrient-rich laboratory medium. The institute’s account of the minimal synthetic cell describes the result as a working approximation to a minimal cell.

The qualifier “in nutrient-rich laboratory medium” is essential. JCVI-syn3.0 was not shown to be self-sufficient in every environment. If a cell’s surroundings provide a nutrient or perform a function it otherwise would have to manage itself, the cell may be able to dispense with some of the relevant genetic capacity. A cell that can grow under one set of supportive conditions does not thereby establish what a cell needs in a different habitat.

The 473-gene result also does not describe the first cell created from raw chemicals. Researchers designed and synthesized a reduced genome and installed it in an existing recipient cell. That distinction separates the achievement—building a viable cell controlled by a designed, reduced genome—from the much broader claim that every component of a cell was created from scratch.

Why isn’t 473 a universal minimum?

Because different organisms live under different conditions, they can rely on different sets of genes. A bacterium that depends on a host may lose functions that the host supplies; a cell expected to grow independently must retain the functions needed for its own growth in the conditions it faces.

A 2023 paper by Moger-Reischer and colleagues notes that some obligately endosymbiotic bacteria have fewer than 200 protein-coding genes. That number does not contradict JCVI-syn3.0’s 473 total genes: it refers to a different group of organisms, counts protein-coding genes rather than all genes, and concerns bacteria that depend on a host. Those are different conditions and different counting units.

The reduced-genome example also shows why “viable” does not mean “works as well as possible.” The 473-gene strain divided irregularly. Adding 19 genes produced the JCVI-syn3A derivative, and investigators identified seven genes important for restoring regular division. NIST’s 2021 account describes the work on a synthetic-genome cell that grows and divides normally. These results distinguish the smallest reported viable configuration in that experiment from a configuration with more regular division.

Another derivative, JCVI-syn3B, has 493 genes. Its count should not be substituted for syn3.0’s: the strains are different versions, and the number depends on which derivative is being discussed. The comparison is useful only with the strain name attached.

What does the human gene count mean?

NHGRI estimates that humans have approximately 20,000 protein-coding genes. This is an estimate of the number of genes that encode proteins, not the total number of DNA instructions in a human genome and not an experimentally measured minimum for keeping a human cell alive. The information encoding those protein-coding genes occupies only about 1.5% of the human genome, according to NHGRI’s gene glossary.

One single-copy human nuclear genome has roughly 3 billion nucleotides across 23 chromosomes, according to NHGRI’s 2023 figure. Most human cells carry two nuclear copies. The complete human-genome description also includes mitochondrial DNA, so “one genome” can refer to a different scope depending on whether the speaker means a single nuclear copy or the full set of DNA in a cell.

Do not read the approximately 20,000 protein-coding genes as a survival threshold. It tells you about estimated genome content in humans; it does not report how many could be removed while sustaining a human cell. The bacterial experiment cannot fill that gap: it involved a different species, a different cell system and specific laboratory conditions. The available figures answer different questions rather than forming a direct human-versus-bacterium ranking.

Nor does having more DNA or more genes automatically mean an organism is more biologically complex. NHGRI cautions that genome size and biological complexity do not correlate consistently. A gene total is therefore not a simple scale of sophistication, quality or independence.

How did researchers decide what to remove?

Creating JCVI-syn3.0 involved reducing a bacterial genome through design, synthesis and testing. The process was not simply a matter of deleting every gene initially labeled “nonessential.” Early candidate reductions failed, and the team learned that genes important for robust growth had to be retained even when simpler essential-versus-nonessential classifications suggested otherwise.

This is a useful distinction between a strict survival requirement and a contribution to reliable performance. A gene’s effect can depend on the cell’s genetic background and environment. A cell may remain viable without a gene under one condition while growing less robustly or dividing less regularly. The experiment’s result is consequently best understood as a carefully qualified minimal-cell approximation, not a definitive list of genes every cell needs.

Unknown function is another reason the list is not a universal blueprint. The JCVI’s 2016 account reported 149 genes with biological functions not assigned at that time; this did not mean that 149 genes had been shown to be dispensable. In an August 2026 study, Mushegian and colleagues estimated that 50–60 protein products still lacked predicted functions. The 2016 count and 2026 estimate describe different things—genes versus protein products, and assigned versus predicted function—and should not be treated as directly interchangeable. The 2026 authors proposed possible membrane-transport and immunity roles based on sequence and structural analyses, which is not experimental proof of every protein’s role.

How should you interpret a claim about a cell’s “minimum genome”?

Start by asking what the number counts and what the cell was required to do. A claim about a minimum can sound universal even when it refers only to one organism under a tightly specified environment.

  • Check the organism and strain. “Bacterium” is not a single biological type, and different derivatives of the same research strain can have different gene totals.
  • Check the counting unit. A total gene count may include RNA-coding genes as well as protein-coding genes; a protein-coding count does not include both categories.
  • Check the environment. Nutrient-rich laboratory medium, a host-dependent lifestyle and an independent life in other surroundings impose different demands.
  • Check the outcome. Remaining viable does not necessarily mean growing robustly or dividing regularly.
  • Check what is known about each gene. Unknown or predicted function is not evidence that a gene is unnecessary.

With those questions answered, numbers can be compared without implying more than they establish. The 473-gene JCVI-syn3.0 figure is a significant experimental result about one engineered bacterium under supportive laboratory conditions. It is not a universal biological constant, a human-cell requirement or proof that a smaller count would work for every organism.

Does a cell need every gene in its genome?

Not necessarily in every environment or for every outcome. The reduced-genome work shows that a particular bacterium could remain viable after extensive genome reduction in supportive laboratory conditions, while the difficulty of preserving robust growth shows why “not strictly required for survival” is not the same as “has no useful role.”

Frequently Asked Questions

Are all the cells in your body genetically identical?

Many human cells share the same inherited nuclear genome, but cells can acquire DNA changes as they divide, and some cell types have specialized differences. Cells can also use the same genes differently, which is one reason the same genome can support distinct cell types.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Can a cell’s genome change during its lifetime?

Yes. DNA can acquire mutations, and some changes can be passed to daughter cells when a cell divides. A genome is the cell’s DNA complement at a given point, not a guarantee that every cell in an organism carries a perfectly identical sequence.

Does genome sequencing tell you what every gene does?

No. Sequencing can determine DNA sequence, but assigning a biological function to a sequence is a separate task. The continuing uncertainty about some JCVI-syn3.0 protein products illustrates that a known sequence does not automatically reveal a confirmed role.

What is the difference between a chromosome and a genome?

A chromosome is a packaged DNA molecule containing many sequences and genes. A genome is the complete set of DNA instructions in the cell, which can include multiple chromosomes and DNA outside the nucleus, such as mitochondrial DNA in humans.
