A 2013 Oxford study described an experimental way to weaken Escherichia coli without attacking the cell directly. A cyclodextrin molecule was found to block Wza, a pore in the bacterium’s outer membrane that exports the K30 polysaccharide used to build its capsule. When capsule production is interrupted, the protective layer becomes reduced or absent, exposing an underlying lipopolysaccharide (LPS) surface that antibodies and complement can attack.
This is a laboratory-stage antibacterial strategy, not an approved medicine or a proven human treatment. The idea that it could make resistance harder to evolve is a rationale for further study, not a demonstrated clinical result. Whether the approach works in a living animal was still an open question in the report.
What the Oxford team discovered
The work was led by Ben Davis and Hagan Bayley at Oxford and reported by James Urquhart in Chemistry World on 1 July 2013. The associated research paper is L. Kong and colleagues, published in Nature Chemistry in 2013 (DOI: 10.1038/nchem.1695).

The target was the Wza pore in the outer membrane of E. coli. Wza normally provides an exit route for K30, a long polysaccharide that the bacterium assembles into its capsular polysaccharide layer, or CPS. The CPS forms an outer protective coating around the cell. The researchers found that a cyclodextrin could mimic K30 closely enough to block the pore. That interference prevents normal capsule export, weakening the coating.

The important shift is strategic: the compound does not need to destroy the bacterium by itself. It removes part of the bacterium’s defence and leaves the host immune system with a better view of the cell surface.
How the “sugar” strips away bacterial armour
1. Wza is the export gate
Wza is a pore embedded in the outer membrane. Its normal job is logistical rather than destructive: it allows the K30 polysaccharide to move out of the cell so the CPS can be built around the bacterium. Blocking Wza therefore interrupts construction of the capsule at its export step.
2. Cyclodextrin imitates the exported material
Cyclodextrins are ring-shaped carbohydrate molecules. In this study, one was able to mimic K30 sufficiently to occupy or obstruct the Wza channel. The result is not that the capsule is chemically dissolved; rather, the supply route needed to maintain it is blocked.
3. The capsule barrier is reduced
With export disrupted, the bacterium loses or reduces the CPS layer that normally sits outside other surface components. Davis explained why attacking the capsule itself is unattractive: “If you go after the CPS itself there is loads of this armour so it’s hard to hit and even if you do it just keeps piping out.” Targeting the export machinery aims at the process that continually supplies that armour.

4. A previously shielded surface becomes reachable
Under the CPS is a lipopolysaccharide layer. When the capsule no longer masks it as effectively, antibodies can bind exposed bacterial surfaces. Complement, a group of immune proteins, can then help kill the weakened cell. The cyclodextrin is therefore acting as an immune-sensitising agent: it changes the bacterium’s vulnerability rather than serving as a stand-alone poison.
Why this differs from a conventional antibiotic
Most familiar antibiotics are designed to damage a vital bacterial process directly. The Wza proposal takes a different route by disabling an external defence and relying on host immunity to finish the job.
| Comparison point | Wza-blocking cyclodextrin strategy | Conventional antibiotic approach |
|---|---|---|
| Primary target | Wza, the capsule-export pore in the outer membrane | A bacterial target inside or on the bacterium that the drug directly disrupts |
| Immediate action | Reduces export of K30 and weakens the CPS shield | Directly interferes with a bacterial process |
| How the cell is cleared | Exposed surfaces can be recognised by antibodies and attacked with complement | The antibiotic’s own activity is expected to damage or kill susceptible bacteria |
| Evidence described for this strategy | Laboratory, or in-vitro, findings | The table does not claim a particular drug or clinical result |
| Resistance rationale | Potentially less direct selection pressure because the compound itself does not kill the cell | Resistance can be selected against the direct antibacterial activity |
The distinction is not that one category is always superior. A host-assisted compound may depend on a functioning immune response and on the bacterium exposing the right surface. A direct antibiotic may work through a mechanism that does not require antibodies or complement. The Wza idea is valuable because it adds a different design option to the antibacterial toolbox.

That is the proposed biological effect. In the reported experiments, blocking Wza removed the protective advantage supplied by the CPS and allowed immune mechanisms to engage exposed LPS. Antibodies can recognise the newly accessible surface, while complement can contribute to bacterial killing.

This does not mean that every E. coli infection would respond in the same way. The finding concerns the K30 capsule-export system studied by the Oxford group. A treatment would have to reach the relevant bacteria, block Wza effectively, and operate in a host environment where antibodies and complement can function. Those requirements are part of why an in-vitro result cannot be treated as proof of clinical benefit.
Does the approach prevent antibiotic resistance?
No measured resistance rate was reported, and the study did not establish that resistance would be prevented. The proposed advantage is mechanistic. Because the cyclodextrin does not need to penetrate deeply and directly kill the bacterium, Davis argued that resistance “will not be down to the compound alone.” In his description, the host’s immune response supplies an essential part of the killing pressure.
That logic could reduce the selection pressure associated with a compound whose own toxicity eliminates susceptible cells, but “could” is the key qualification. Bacteria might still adapt in ways that reduce Wza binding, alter capsule export, change surface exposure, or otherwise limit immune access; the report did not measure which routes occur or how often. A host-assisted mechanism can make resistance biology more complicated, not make resistance impossible.
The responsible conclusion is therefore limited: Wza is a plausible target for exploring lower-direct-selection antibacterial strategies. The 2013 work did not provide a resistance-prevention result in animals or people.

What evidence existed, and what was still unknown?
The evidence described was from laboratory experiments. It showed the intended chain of events under experimental conditions: a cyclodextrin can interfere with Wza, capsule export can be disrupted, and immune-mediated attack can become possible when the underlying surface is exposed.
David Livermore, an antibiotic-resistance expert at the University of East Anglia, identified the central limitation: “The obvious question is: ‘Does it work in vivo as well as in vitro?’” In-vivo performance means working in a living organism, where distribution, stability, immune status, tissue penetration, dosing and safety all affect the outcome. None of those questions was answered by the report’s laboratory findings.
The source provides no animal-survival result, efficacy percentage, human-trial result, resistance frequency or market-availability figure. It also does not establish that a Wza-blocking cyclodextrin reached clinical development. Readers should treat the work as an early proof of concept and a direction for research rather than as evidence of an available therapy.

That distinction matters because “cyclodextrin” is a broad chemical family. Generic cyclodextrin ingredients, laboratory reagents or products that happen to contain a cyclodextrin are not automatically the experimental molecule used to interfere with Wza. Buying or taking a generic product would not reproduce the reported antibacterial strategy and should not be treated as self-treatment for an E. coli infection.
Why Wza was considered an attractive target
The capsule is an appealing defensive structure but a difficult object to attack directly. Davis’s explanation captures the engineering problem: the bacterium has a large amount of CPS, and it can continue piping material out. Wza offers a narrower chokepoint. Instead of trying to remove a large, replenished layer from the outside, a molecule can obstruct the route that supplies it.
The target is also positioned in the outer membrane. Davis noted that the compound “doesn’t need to penetrate so deeply into the bug like some antibiotics.” A surface-accessible target can be advantageous in principle, although the report did not establish how well a drug would reach Wza in an infected body or how long blockade would persist.
Most importantly, the strategy links bacterial physiology to immune clearance. CPS is not merely a structural coating in this model; it is the feature that keeps immune effectors from readily reaching the LPS layer beneath. Remove that shield, and the bacterium may become much easier for the host to recognise and damage.
Questions future studies would have to answer
- Does it work in vivo? Laboratory activity must translate into a measurable benefit in an animal before claims about living hosts can be made.
- How broad is the target? The report focused on the Wza system associated with K30 export in E. coli; it did not establish coverage across all E. coli strains or other bacteria.
- Can enough compound reach the pore? A useful medicine would need suitable distribution and exposure at infected sites, issues not resolved by the in-vitro work.
- How much immune function is required? Because antibodies and complement participate in the proposed killing mechanism, the host response is part of the treatment concept rather than a background detail.
- What resistance pathways emerge? The rationale predicts a possible advantage, but the frequency, speed and biological cost of resistance were not measured.
- Is the compound safe and developable? The report supplies no human safety, dosing, manufacturing or approval data.
What readers should take away
The “sweet spot” in this antibiotic research is a vulnerable point in bacterial defence. A cyclodextrin was reported to block Wza, stop normal K30 capsule export and expose an LPS layer that antibodies and complement can attack. That is a host-assisted antibacterial concept, not a conventional killing antibiotic.
Its significance lies in the target and the mechanism: disable the capsule’s supply route instead of trying to overpower the capsule itself. Its limits are equally clear. The evidence was laboratory-stage, the in-vivo question remained unanswered, no human efficacy or availability was shown, and the resistance benefit was proposed rather than measured.
