Humans usually repair injuries in more limited, tissue-specific ways and do not form this kind of blastema. The difference is not explained by one magic gene—or by scarring alone.
How a salamander limb regenerates
Axolotls, a type of salamander widely used in research, show how limb regeneration differs from simply closing a wound. After amputation, a blood clot seals the surface. In the sequence described in an axolotl study, specialized wound epithelium covers the injury within about a day. With nerve input and other local signals, cells in the stump activate and multiply, forming a blastema: a population of cells that contributes to replacement tissues.
The blastema is not an unstructured lump, and its cells do not all become any tissue at will. Progenitors are substantially fate-restricted, and their coordinated activity helps rebuild an organized limb.
Nerves, immune cells, and the wound covering all matter
Experiments show that regeneration depends on more than cells capable of dividing. Salamander limbs deprived of nerve input before amputation fail to form a blastema and may heal with fibrotic tissue. Depleting macrophages, or preventing proper wound epithelium from forming, can also cause regenerative failure. These results make “stem cells grow back the limb” an incomplete explanation: the injury environment helps determine whether rebuilding proceeds.

Why human healing usually does not rebuild a limb
Most mammals, including humans, do not form blastemas, according to the U.S. National Institute of General Medical Sciences (NIGMS) in its Regeneration overview, last updated February 13, 2025. Human repair is generally tissue-specific: adult stem cells tend to contribute to the tissues or organs where they reside, and wounds may close with scar formation. That can restore a barrier or repair a structure without recreating the precise arrangement of bones, joints, muscles, nerves, blood vessels, and skin in a complete limb.
Scarring is part of the contrast, but available evidence does not establish it as the sole reason humans do not regrow limbs. The broader difference involves how injury signals, progenitor cells, tissue patterning, and wound repair work together. Researchers have not identified one switch whose activation would make a human limb regrow.

Humans do regenerate some tissues
Human regenerative capacity is limited, not absent. Skin and hair are renewed; bones can knit after fractures; and some organs can compensate after damage. The liver, for example, can show compensatory growth after part of it is removed or injured. There is also evidence of regeneration of amputated fingertip portions during childhood. None of these examples amounts to reconstructing a whole arm or leg.

Axolotls are valuable models because they regenerate limbs and have structures homologous to those in humans. NIH’s Office of Research Infrastructure Programs describes their usefulness for studying regenerative medicine, while emphasizing that much remains to be learned before regenerative processes might be reproduced in mammals. A 2024 review in Annals of Anatomy describes pharmacological, tissue-engineering, and cell-therapy approaches as facing significant challenges.

Whole-limb regrowth in people remains a research aspiration, not an established treatment. Even salamander regeneration is not guaranteed under every condition: a 2017 axolotl study found that repeated amputation under its experimental conditions could compromise later regeneration initiation. That result underscores how context-sensitive the process can be; it does not show that the same mechanism explains human healing.
