It can identify a broad maternal-line haplogroup and show genetic matches in a testing company’s database, but it does not map all of your mother’s relatives, estimate your whole-family ethnicity, or prove that you descend from a particular named ancestor.
What mitochondrial DNA testing actually follows
mtDNA is found outside the cell nucleus and is inherited from a person’s mother. Both men and women have mtDNA, but only women pass it on. A test therefore traces one unbroken chain of mothers—not every branch of your family on your mother’s side.
For example, your mtDNA comes from your mother, who received hers from her mother, who received hers from her mother. Your mother’s father is not on that direct maternal chain, even though he is part of your maternal family. The same is true of your father’s side and of other branches that do not follow mother-to-child-to-mother. A man can take an mtDNA test to investigate his own direct maternal line; he simply will not pass his mtDNA to his children. To test the direct maternal line of a relative who can pass it on, choose a living descendant of that line whose maternal chain is intact.

What an mtDNA result can tell you
Your maternal-line haplogroup
A haplogroup places your mtDNA on a branch of a large genetic family tree. It can offer broad context about the ancient history and migrations associated with that branch. It is not an ethnicity estimate for your whole family, nor proof that your ancestors belonged to a particular modern nation or ethnic group. A map that describes a haplogroup’s broad history is also different from locations reported by individual testers, which depend on participant-provided information. Potential matches in a company’s database
A match means your tested mtDNA sequences are sufficiently similar under that provider’s rules. It is consistent with a shared direct maternal-line ancestor, but does not usually identify which person in a family tree that ancestor was. Because mtDNA changes slowly, the shared ancestor may have lived hundreds or thousands of years ago; a match alone does not provide a precise date.

A match list is limited to people who tested with that company, took a comparable test, and are included under its matching and visibility settings. It is not a list of everyone alive who shares your maternal-line ancestor. FamilyTreeDNA says people who have not signed a release or have opted out of matching are not displayed; its rules are provider-specific. Evidence to guide family-history research
mtDNA can help assess a hypothesis about a direct maternal line, especially when you compare appropriately tested relatives whose documented maternal lines are relevant. Treat a match as a clue to investigate alongside birth, marriage, death, census, and other records—not as proof of descent from a named person. The genetic result and the documentary evidence answer different parts of the question.

What the test cannot establish
- Your whole family tree: the test follows one maternal chain and says little about the many other branches you inherited from.
- A specific recent relationship by itself: a matching sequence does not generally tell you which ancestor you share or exactly when that person lived.
- Your overall ethnicity or a single country of origin: a haplogroup is one lineage, not a summary of all your ancestors.
- Every possible relative: the result depends on who has tested, which sequence was tested, and whether people appear in the database.
- A conclusion independent of records: genetic similarity can support a genealogical investigation, but does not substitute for identifying people and relationships through evidence.
Why test coverage and matching rules matter
Not every mtDNA test compares the same amount of sequence. FamilyTreeDNA describes its mtFull Sequence test as covering HVR1, HVR2, and the coding region. It describes HVR1-only and HVR1/HVR2 comparisons as lower resolution. More sequence can make comparisons more informative, but it does not convert genetic distance into a precise number of years or generations. FamilyTreeDNA’s test overview sets out its coverage.
Matching thresholds also vary by provider. For example, FamilyTreeDNA says its full-sequence matching allows up to three differences, including certain heteroplasmy cases, and excludes insertion/deletion locations 309 and 315 from its count. Those are that company’s rules, not universal standards for all mtDNA tests. Compare the test’s coverage and the provider’s matching explanation before interpreting a match. How mtDNA compares with other ancestry tests

There is no universally best DNA test; the useful choice depends on the question. If you want potential relatives across many branches, autosomal testing is generally a better fit than mtDNA. If you want one direct maternal line, mtDNA is the relevant lineage test. How to use a match in a family-history investigation
- Define the line: write down the specific mother-to-mother chain you want to investigate, rather than treating “my mother’s side” as one lineage.
- Check what was tested: note whether the result covers HVR1, HVR1 and HVR2, or the full sequence, and read the provider’s match criteria.
- Review the match context: check the available maternal-line information and any reported origins, remembering that these details are not independently established simply by appearing in a match list.
- Compare records: look for a plausible shared maternal ancestor in documented trees and evaluate the records connecting each generation.
- Test a relevant relative if appropriate: a person whose documented direct maternal line matches the question may provide a useful comparison; testing a relative from a different branch will not answer the same lineage question.
Consent and privacy considerations
DNA results can reveal unexpected relationships or information that affects relatives as well as the person tested. Get permission before arranging or managing another person’s test. Before submitting a sample, read the provider’s current consent, matching, data-use, and visibility options; some services allow users to opt out of match display, but terms can change. FamilySearch advises reviewing each company’s policies and considering the possibility of unexpected findings in its DNA testing FAQ.
