A scientific photograph or data image is reliable evidence when it faithfully represents the original observation or measurement, its acquisition and processing are transparent, and enough source material and context are available to assess the claim. A polished figure by itself cannot establish reliability: readers need to know how it was made, what was changed, whether comparisons are fair, and whether the image supports the conclusion being drawn.

Why scientific images should be treated as data

A photograph, microscopy panel, gel, blot, scan, or visualization is not merely an illustration. It is a representation of observations or measurements, and changes to it can change what a reader thinks was observed. The U.S. Office of Research Integrity (ORI) says, “Digital scientific images should be treated as data,” and advises retaining original files: ORI’s Guidelines for Best Practices in Image Processing.

Reliability is therefore not a matter of whether an image looks convincing. It depends on traceability to the original, a defensible acquisition and processing history, fair comparisons, and documentation that lets others judge the inference. An irregularity may warrant scrutiny, but it does not by itself establish intent or misconduct.

How to assess an image

  1. Check provenance

    Look for access to the source image or underlying data, and for enough identifying context to connect it to a sample, acquisition, date or sequence, and experimental condition. For gels and blots, check whether the journal requires original uncropped files.   2. Examine acquisition details

    Methods or legends should identify the relevant instrument and acquisition settings. Exposure, detector settings, sample preparation, sampling choices, and other acquisition decisions can affect the pattern shown. Without those details, it may be difficult to distinguish a meaningful signal from an acquisition effect. Nature’s policy asks authors to document tools and key settings: Nature Digital Image Integrity.

  3. Trace processing from original to figure

    Check whether adjustments are disclosed, applied consistently to the whole image, and applied equally to controls. If panels come from different samples, times, or acquisitions, they should not be presented as though they were one observation; boundaries and legends should make the construction clear. Relevant filtering, thresholding, gamma changes, deconvolution, reconstruction, or pseudo-coloring should be explained when they affect interpretation. Compare the presented figure with source files where available.

  4. Test whether comparisons are fair

    Images compared with one another should be acquired under equivalent conditions. For quantitative intensity claims, measurements should be made from raw data and calibrated against a known standard; a display’s brightness or color alone is not a measurement. Consider resolution, magnification, scale bars, artifacts, controls, and how numeric values map to color or intensity. These principles are reflected in Nature’s image policy and ORI’s image-processing guidance.

  5. Ask whether the image supports the interpretation

    Check whether the figure includes enough context to assess alternative explanations and artifacts. A crop can be legitimate if it is representative and does not conceal relevant context; selective omissions can distort what the reader sees. The CLIP principles emphasize that biological variability, acquisition, image quality, interpretive standards, evaluator experience, and artifacts all affect how consistently an image can be interpreted: CLIP principles article abstract.

Which image adjustments can be acceptable?

Some processing is compatible with reliable presentation, but acceptability depends on the image type, community standards, and the specific journal’s rules. Nature’s figure guide states: “The final image must correctly represent the original data and conform to community standards.” Its guidance permits a degree of processing, including whole-image brightness or contrast changes, when these do not obscure information and are applied consistently, including to controls.

By contrast, local touch-ups that remove, add, clone, heal, or selectively brighten observed content can misrepresent the underlying observation. ORI warns that such practices can make “a misstatement to the reader about what was seen.” Nature’s cited figure guidance also prohibits generative image content. Consult the policy for the journal and figure type rather than assuming one rule covers every publication.

  • Potentially acceptable: uniform whole-image brightness or contrast adjustment that preserves relevant information and is applied equally across comparisons.
  • Requires clear disclosure where relevant: pseudo-coloring, gamma changes, filtering, thresholding, deconvolution, reconstruction, and combining or juxtaposing separately acquired images or lanes.
  • Warning signs: selectively altering a region, concealing boundaries between separate sources, or adding or removing a feature.

What documentation makes a figure interpretable?

A useful figure legend and methods account should let a reader understand how the displayed image relates to the observation. Depending on the image and claim, look for:

  • Original, unprocessed image files and relevant metadata.
  • The instrument or acquisition tools and important settings.
  • Sample identity, conditions, and relevant acquisition timing or sequence.
  • Processing software and material transformations.
  • Clear explanation of crops, separate panels, and any boundaries or rearrangements.
  • Scale, calibration, controls, and color or intensity mapping when relevant.

When substantial irregularities compromise a published finding, a journal may need to correct the record or consider retraction. COPE’s retraction guidelines discuss when retraction may be warranted: COPE Retraction Guidelines. The presence of an irregularity alone is not proof that it was intentional.

A practical comparison checklist

When evaluating two or more images, use these questions rather than assigning a numerical reliability score:

  • Fidelity: Can the displayed figure be traced to original data without unexplained changes?
  • Acquisition: Were the images captured under equivalent conditions for the comparison being made?
  • Processing: Are adjustments disclosed, consistent, and non-selective?
  • Scale and resolution: Are magnification, scale, and image quality adequate to support the claim?
  • Calibration and display: Are quantitative values calibrated, and is color or intensity mapping explained where needed?
  • Context: Are controls, artifacts, sample conditions, and relevant surrounding information available?
  • Traceability: Can readers evaluate source files and the documented processing trail?
