But they did not work as mirror-image switches: dopamine reduced activity in a particular group of inhibitory neurons in the antennal lobe, while octopamine reduced the main neural response without changing that measured inhibition. The findings show how chemical signals can tune an odor-processing circuit—not how humans subjectively experience smell.
What the locust study found
The study, “Distinct mechanisms mediate dopamine-octopamine opponency in an insect model of olfaction,” was published in The Journal of Neuroscience on September 14, 2026. Researchers studied both sexes of Schistocerca americana and compared the effects of dopamine and octopamine on odor-evoked neural activity and an appetitive behavior. The PubMed abstract reports the central results; WashU’s October 5, 2026 report describes the behavioral response.
The behavior was palp opening: the locust spread appendages near its mouthparts that help it touch or grasp food. Researchers presented several odorants, which WashU described for readers with analogies such as grass, citrus, rose, almond, and a spicy floral scent. These labels help identify the experimental odors; they do not mean locusts perceive or name smells as people do.

The abstract and institutional account do not provide numerical effect sizes or sample counts, so the comparison is directional rather than a statement about how large either effect was.

Where odor processing happens in the locust
Odor information begins at sensory neurons in the antenna. Those neurons send signals to the antennal lobe, the first central olfactory circuit. There, local neurons and projection neurons shape the incoming activity; projection neurons carry processed information onward to higher brain regions, including the mushroom body.
GABAergic local neurons provide inhibition within the antennal-lobe network. One useful way to think about the findings is that neuromodulators can adjust the circuit’s gain—the relationship between incoming odor signals and the circuit’s output. Dopamine reduced the activity of one inhibitory subgroup, which was associated with greater principal-neuron responses. Octopamine also lowered the principal response, but not by changing the local-neuron inhibition measured in this experiment.

Why the two modulators are not simple opposites
Both chemicals affected responses to odors, but the neural routes differed. Dopamine’s effect was associated with reduced activity in a GABAergic local-neuron subgroup and a release from that measured inhibition. Octopamine lowered odor-evoked principal activity even though the measured local-neuron inhibition did not change.
Barani Raman, the study’s senior author, summarized the contrast: “What we found was that octopamine did not affect the activity of local neurons at all,” while dopamine “suppressed one subpopulation of local neurons. It released the circuit from inhibition to get that boost in the neural network output. Octopamine did not do that.” The WashU McKelvey Engineering report also quotes Raman describing a simple antennal-lobe model with one neuron subgroup that increases behavioral response and a second that reduces or suppresses it. That model helps explain the results, but it does not establish that every cell group or downstream step has been identified.

The authors’ intrinsic-excitability explanation for octopamine should also be kept distinct from a direct measurement of a specific projection-neuron mechanism. The result establishes reduced principal neural responses alongside unchanged measured local-neuron inhibition; the precise alternative mechanism remains an interpretation to test.

Serotonin is relevant to locust olfaction, but it belongs to a separate line of evidence. A 2024 review by Zhang and Xu focuses on serotonin receptor 2 and odor input at the locust antenna, the sensory periphery rather than the antennal-lobe comparison in the dopamine–octopamine study. The review notes that neuromodulation in peripheral olfactory systems is less understood than modulation in the antennal lobe. Read the 2024 review.

Earlier locust work summarized in a university report suggests serotonin’s behavioral effect can depend on odor identity. That does not mean serotonin was tested as a central result of the 2026 dopamine-and-octopamine experiments, nor does it make serotonin interchangeable with either modulator.
Octopamine also appears in research on locust learning, but at a different circuit site and in a different question. In work on the mushroom-body β-lobe, octopamine delivered after spike-timing-dependent plasticity could selectively modify responses at synapses previously tagged by activity, affecting odor-evoked output. This concerns odor-specific plasticity in a higher brain area; it is not a replication or direct explanation of the antennal-lobe effect. What this teaches—and what it does not
- It shows that odor responses are actively shaped inside a circuit. The antennal lobe does not simply pass an unchanged antenna signal onward; local inhibition and other properties influence its output.
- Opposing behavioral outcomes can come from different mechanisms. Dopamine and octopamine moved measured neural and palp-opening responses in opposite directions, but the reported circuit changes were not opposites in the same pathway.
- The findings are specific to the studied locust and conditions. They do not establish a universal effect of dopamine or octopamine on smell in insects, mammals, or humans.
- Neural responses and subjective smell are not the same measurement. The study measured activity in an olfactory circuit and a food-related behavior, not what an odor felt like to the animal.
