There is no universal recipe: template design, reagents, temperatures, and timing vary among wheat-germ, bacterial-lysate, and purified-component systems.
What cell-free protein expression does
Cell-free protein synthesis (CFPS) produces protein outside living cells. A reaction supplies the transcription and translation machinery and reagents that a cell normally provides; you supply a nucleic-acid template encoding the protein. Depending on the system, that template may be plasmid DNA, linear DNA, or mRNA. Some kits use cell lysate, while others use purified components. New England Biolabs’ overview of cell-free protein expression explains these system differences and describes applications including screening and protein engineering.
Before you start: check the kit and the template
Read the current manual for the exact kit you have before preparing a reaction. Do not transfer a volume, temperature, or incubation time from another kit or from an example protocol.
- Template type and design: Check whether the kit accepts plasmid DNA, linear DNA, or mRNA, and whether the template needs a promoter, ribosome-binding site, or other sequence elements. For example, Promega’s S30 T7 E. coli system calls for cloned DNA with a T7 promoter and ribosome-binding site. Its technical manual describes that system’s requirements.
- Reaction contents: Confirm which reagents are supplied and which must be prepared or purchased separately. Systems can differ substantially in composition and setup.
- Handling and scale: Note the specified reaction volume, storage temperature, extract handling limits, and whether the instructions support the scale you intend to use.
- Detection method: Decide how you will check for expression. The appropriate assay depends on the target protein and kit.
- Controls: Check whether the manual recommends a positive control or other controls for your first run.
Follow the kit’s workflow
The exact steps depend on the product. Sigma-Aldrich’s CFPS700 wheat-germ protocol illustrates a workflow in which DNA is transcribed to mRNA first, and the mRNA is then used in a separate translation reaction. Other kits combine transcription and translation in one reaction.

- Prepare the template. Use the template form and sequence design specified for your kit. In the CFPS700 example, the process starts with a DNA transcription template.
- Transcribe DNA to mRNA if the protocol requires it. The CFPS700 protocol uses T7 RNA polymerase for transcription at 37 °C for three hours, with up to six hours specified in the protocol. These conditions apply to that product, not to cell-free kits generally.
- Purify and check the mRNA if instructed. The CFPS700 workflow calls for mRNA purification and confirmation before translation.
- Set up the translation reaction. Follow the kit’s exact component volumes and order of addition. In the CFPS700 example, translation uses wheat-germ extract, an amino-acid mix, and mRNA. Its example batch reaction is incubated at 16 °C overnight for more than ten hours. The protocol warns that adding more than 10 µL of wheat-germ extract to its 110 µL example mixture may reduce yield.
- Measure the result. Use an assay suited to the protein and the kit’s instructions; a reaction completing its incubation does not by itself confirm that the desired protein was made.
Those temperatures, times, and volumes are specific to the cited CFPS700 protocol. For a different kit, follow its own manual. For example, Promega describes a coupled S30 T7 E. coli system containing T7 RNA polymerase and components for translation, while NEB distinguishes its lysate-based NEBExpress system from its purified-component PURExpress system. NEB’s system overview provides that distinction.
Choose a system for the target and experiment

Protect the reaction and troubleshoot methodically
RNA is vulnerable to RNase contamination, so use the RNase-free handling practices specified by the kit. Handle and store the extract according to its manual: CellFree Sciences’ wheat-germ kit manual, copyright July 2024, specifies storage at −80 °C and warns that repeated freeze-thawing can inactivate the extract.

For an initial experiment, use a small-scale reaction and the manual’s recommended positive control when available. If the target is not detected, check the following before changing several conditions at once:
- Confirm that the template is intact and has the required design elements.
- Check that reagents were stored and handled as directed, including limits on extract freeze-thawing.
- Review RNase-free technique, especially when preparing or handling mRNA.
- If the source of failure is unclear, test transcription and translation separately when the kit’s instructions support it. The CellFree Sciences manual recommends separating the stages as a troubleshooting approach.
Because each system has its own chemistry and operating conditions, troubleshoot against that product’s manual rather than applying settings from another platform.
