Choose a cell-free protein synthesis (CFPS) kit by matching its system to your target protein and intended downstream use—not by picking the kit with the biggest headline yield. Before ordering, verify the template type the kit accepts, its protocol and reaction format, and the full cost per usable reaction. No single CFPS platform is established as best for every target.
Start with the protein and what you need to do with it
Write down the target protein, the template you plan to use, and the experiment’s endpoint. A kit for a quick expression test may not be the right choice for producing material for a specific downstream assay. A broader review also highlights target origin and complexity, reagent availability, convenience, downstream processing, and cost.
Use these factors to shortlist systems, then check the manufacturer’s current protocol for each product. Platform-level descriptions can guide where to start, but they cannot establish how a particular target will perform.
Compare the main CFPS system families
Bacterial extract
Escherichia coli extract is a common option when productivity is important. Its metabolism can provide energy for protein synthesis, but that does not guarantee a particular yield for your protein. Consider it a starting point when its system and workflow fit your target and experiment.

Eukaryotic extracts
Commercial alternatives include wheat-germ, rabbit-reticulocyte, insect, CHO, and HeLa extracts. Investigate these when the target or intended downstream work makes a eukaryotic system appropriate. The available reviews do not establish a universal ranking of eukaryotic and bacterial platforms or identify one that will work best for every target.
Purified-component systems
Most commercial CFPS products use cell extracts. PURExpress is a different approach: the 2019 review describes it as a reconstitution of purified E. coli transcription and translation components, rather than an extract. A defined composition may matter when you want a system built from specified components; it is not evidence that the system is more productive or better suited to every experiment.

Confirm the template and workflow before comparing prices
Template compatibility differs across platforms and products. Check the product protocol for the exact DNA or mRNA template you intend to use, and confirm whether the kit supplies any transcription components your workflow requires.
Some workflows separate template preparation, transcription, and translation. The MilliporeSigma Next Generation Cell Free Protein Expression Kit (Wheat Germ) protocol is one example. Do not assume that a product labeled “cell-free” accepts your template as supplied or includes every step and reagent you need.

Evaluate performance claims in context
A 2014 review reports typical yield ranges across commercial CFPS system families, but those figures come from historical literature and different sources. They are not a standardized, current head-to-head comparison or a guarantee for a specific protein. The reviewed sources do not establish a contemporary matched-condition dataset that ranks currently marketed kits.

A 2019 review gives an illustrative historical estimate of $0.019 per microliter of reaction for in-house E. coli CFPS and $0.15–$0.57 per microliter for commercial lysate-based kits, attributing the figures to earlier work. These are not current vendor quotes and should not be used as present-day prices.

For a purchase decision, use current product pricing and account for required components, the number of reactions you can actually run, expected failures, labor, and any optimization the protocol entails. A lower list price may not mean a lower cost per usable result. The 2019 review’s authors note: “Commercial kits enable users to implement CFPS easily, but for extensive usage, they may not be cost-effective.”
Use a practical shortlist before ordering
- Define the experiment. Record the target, intended downstream application, template, and amount of protein you need.
- Choose plausible system families. Compare bacterial extract, relevant eukaryotic extracts, and a purified-component option if system definition matters to your work.
- Read the current product protocol. Verify template compatibility, included transcription and translation components, reaction format, and setup steps.
- Check evidence that matches your use. Prefer results with comparable targets, templates, reaction conditions, scales, and measurement methods; treat broad historical ranges as context only.
- Estimate usable-reaction cost. Include required reagents, labor, and likely optimization or failed reactions alongside the current kit price.
This process narrows the options without pretending that a platform-level yield figure can identify a winner for an untested target.
