There is no proven leucine threshold that everyone must hit at every meal. A useful starting point for many exercising adults is 20–40 g of high-quality protein per meal (or about 0.25 g per kilogram of body weight), with a broad leucine range of 700–3,000 mg alongside the other essential amino acids. These are practical guidelines, not an on-off switch for muscle growth.
Why leucine matters—but is not enough by itself
Leucine is an essential amino acid and one of the three branched-chain amino acids. It helps signal muscle protein synthesis (MPS), but building muscle protein requires all nine essential amino acids (EAAs), not leucine alone. A meal with a high leucine number but too little complete protein is not equivalent to a balanced protein serving.
The NIH Office of Dietary Supplements says most complete proteins are about 40% EAAs. By that estimate, a 25 g serving of complete protein supplies about 10 g of EAAs. NIH ODS explains protein and amino acids for exercise.

The International Society of Sports Nutrition (ISSN) position stand for exercising people recommends an acute dose of 20–40 g of high-quality protein, or about 0.25 g/kg, and says doses should contain 700–3,000 mg of leucine and/or a higher relative leucine content, alongside a balanced array of EAAs. The stand does not define one exact leucine amount that guarantees an MPS response in every person or meal. Read the ISSN protein and exercise position stand (2017).

Use the figures as a broad guide for a complete protein serving, not as a test you must pass at every sitting. The response depends on the full amino-acid supply and the person; a mixed meal cannot be reduced to a leucine number alone.
What the “leucine trigger” evidence does—and doesn’t—show
The leucine-trigger hypothesis proposes that leucine helps initiate MPS after a meal. A 2021 systematic review examined 29 eligible studies; 16 supported the hypothesis as an explanation for differences in post-meal MPS responses. That is a count of studies, not a prediction that 16 out of 29 people will respond.

The review found the model applied most clearly to older adults and to isolated protein sources, rather than protein-rich whole foods. Leucine concentration alone did not explain all MPS responses, and the authors called for further study of factors beyond blood leucine profiles. Do older adults need more leucine?
Aging can reduce the muscle-building response to protein, sometimes described as anabolic resistance. The 2023 ISSN position stand on EAAs notes that aging may require a greater proportion of leucine to overcome this effect, but it does not establish a universal leucine-only dose. It discusses EAA supplementation more broadly, with EAA-induced MPS occurring across doses and appearing to plateau at around 15–18 g. Read the ISSN EAA position stand (2023).

A 2021 review of nutrition interventions in older adults reports a proposed dose of 10–15 g of amino acids containing at least 3 g of leucine. The review also describes substantial variation in dose, timing, intervention length, and adherence. Treat that figure as a proposal discussed in the review, not a settled requirement for every older adult. See the older-adult nutrition intervention review.
How to apply the guidance to meals and supplements
Start with complete protein
Build meals around a source that supplies protein and the full range of EAAs. Food or a complete protein powder can do this; an isolated leucine supplement does not provide the other EAAs needed to build protein.

Use protein labels to compare products
Compare grams of protein per serving, serving size, and whether the product supplies a complete amino-acid profile. Check the label for leucine or total EAA amounts if listed, but do not assume every protein serving contains the same amount. NIH ODS notes that whey contains more leucine than casein or soy. Whey powder can be a convenient way to meet protein needs, but it is optional.

NIH ODS finds the evidence inconsistent that BCAA supplements stimulate MPS beyond adequate amounts of high-quality protein. That makes a high leucine claim by itself a weak reason to choose a product when a complete protein serving is available.

How meal timing and training fit in
Resistance exercise and protein ingestion each stimulate MPS, and ISSN describes them as synergistic when protein is consumed before or after resistance exercise. NIH ODS summarizes athlete guidance of about 0.3 g/kg of high-quality protein after exercise, then every 3–5 hours. These are athlete-oriented guidelines, not proof that one leucine-rich meal can make up for inadequate protein across the day or replace training.
