The finding adds a surprising candidate to the study of proteins associated with these folded DNA structures—but it does not show what HK1 does at G4 sites inside living cells.
What are G-quadruplexes, and what did the study ask?
G-quadruplexes, often shortened to G4, are folded DNA structures formed by guanine-rich sequences. The study focused on a human telomere G4 sequence and asked which proteins could be found close to it. The researchers reported their method in Communications Chemistry on 27 August 2026.
Finding G4-associated proteins is challenging: a protein recovered near a DNA structure may be a direct binder, a nearby molecule, or a candidate whose association needs further testing. The study therefore used proximity labeling to identify candidates and then tested HK1 separately with purified protein.
How did the blue-light labeling method work?
The researchers attached a photocatalyst—a ruthenium complex or BODIPY—to the human telomere G4 probe. They added a light-responsive MAUra labeling reagent and exposed the mixture to light. The reaction tagged proteins near the probe, after which the team enriched labeled proteins and identified them using quantitative proteomics. The method is described in the primary paper.

Unlike an approach that uses a small-molecule G4-binding ligand to capture proteins, this design does not rely on occupying a G4 binding pocket with that ligand. That may help avoid displacing some native interactors, but proximity labeling still identifies candidates rather than proving that every tagged protein binds G4 directly. Probe design and local conditions also matter: the paper reports different capture profiles when the photocatalyst was placed at a G4 terminus versus in a loop.
In the paper’s purified-protein photolabeling protocol, the sample received 455 nm LED irradiation for 30 seconds. That is a specific laboratory protocol, not a general recipe for consumer blue lights. Reproducing the experiment also requires modified oligonucleotides, the labeling reagent and photocatalyst, controlled reaction conditions, and proteomic analysis.

What proteins did the screen identify?
The Tohoku University-authored summary reports that the method flagged more than 1,000 candidate binders. This is a count of candidates from the screen, not more than 1,000 independently confirmed G4-binding proteins. Each candidate would require follow-up evidence to establish whether it binds G4 directly and what that interaction means. See the Phys.org summary.
HK1 stood out in comparisons between the G4 probe and controls. HK1 is a metabolic enzyme, making it an unexpected candidate in research on DNA structure. The authors followed up with purified-protein assays rather than treating its appearance in the screen as proof of binding.

What evidence supports HK1 binding to G4 DNA?
Electrophoretic mobility shift assays with purified HK1 supported binding to G4 DNA, but not to the tested double-stranded DNA. The team also used microscale thermophoresis (MST) with Texas Red-labeled human telomere G4 DNA. Under the reported assay conditions, the measured dissociation constant was 11.8 ± 2.0 nM (n=3), as detailed in the paper and its methods.
That value describes one assay setup, one labeled G4 sequence, and the study’s experimental conditions. It should not be read as a universal measure of HK1’s affinity for every G4 sequence or as a prediction of binding strength inside a cell.

What does the finding not establish?
The work supports a previously unreported direct interaction between purified HK1 and G4 DNA in vitro. It does not establish that HK1 binds G4 structures in living cells, what function it might have there, or whether the interaction affects gene regulation. Nor does it demonstrate a causal link to cancer or metabolic disease; those are open questions, not outcomes shown by the study.
The broader contribution is a way to profile proteins near G4 DNA using photocatalytic proximity labeling. Its candidate list depends on probe chemistry, catalyst placement, reaction conditions, and the biological sample. As with other proximity-labeling approaches, a tagged protein’s presence near the probe is a lead for validation, not by itself proof of a stable, functional complex.

Why the result matters
HK1 illustrates how a structural-DNA screen can reveal candidates outside the expected set of DNA-associated proteins. The purified-protein follow-up gives the candidate stronger support than proximity labeling alone, while leaving the biological significance unresolved. Future work would need to test whether the interaction occurs in cells and determine whether it changes any cellular process.
