Software then uses that directional information to reconstruct a three-dimensional volume or computationally refocused planes from one camera exposure. The exposure is scanless; the finished volume still requires computation, and the method trades some spatial detail for angular information.
What a light-field microscope records
A conventional wide-field microscope forms an image on a two-dimensional plane. Its sensor records intensity at each image location, while much of the information about the directions of arriving rays is integrated together.
In a common LFM design, a microlens array sits in the optical path and divides the light into many small samples. The position of each lenslet contributes spatial information; pixels behind that lenslet sample different ray directions. Together, those measurements form a sampled light field, often represented by two spatial and two angular coordinates. A complete description of light can include depth, wavelength, and time as well, but a practical camera does not record every dimension independently.
A useful way to think about the added information is that an ordinary image records how bright a point appears, while LFM also preserves a coarse indication of the directions from which light arrived. Those directional patterns vary with depth and give reconstruction software evidence for estimating where signal belongs in a volume. The camera’s raw image is not itself a finished 3D picture.

How one exposure becomes a 3D volume
- The objective collects light from the specimen and forms an image within the microscope’s optical path.
- The microlens array partitions that light into spatial and angular samples.
- The camera records the samples together in one exposure rather than capturing a sequence of focal planes.
- Reconstruction software uses a model of the microscope’s point-spread function and an inverse method, commonly 3D deconvolution, to estimate a volume or computationally refocused planes.
“Single exposure” describes acquisition, not the time required to deliver a reconstructed volume. The primary 2013 method used iterative, GPU-accelerated reconstruction; processing requirements and speed depend on the implementation and the data.
What the speed trades away
Spatial and angular resolution
A sensor has a finite number of pixels. Using some of them to distinguish ray directions leaves fewer available to represent fine lateral detail. The balance between spatial and angular sampling affects both lateral detail and the quality of depth reconstruction. Its exact consequences depend on the optical layout, objective, microlens array, camera, specimen, and reconstruction method. Broxton et al. discuss the need to model diffraction at microscope scales rather than relying only on ray optics in their 2013 wave-optics and deconvolution study.

What a reported resolution result does—and does not—mean
In a particular optical configuration, Broxton et al. reported up to an eightfold improvement in lateral resolution over the earlier computational-refocusing limits used as the study’s comparison. That result came from a planar USAF test target; it is not a general performance figure for LFM and does not mean LFM out-resolves other microscope classes. The authors also noted that their method did not reach the diffraction-limited spatial resolution of confocal, two-photon, or wide-field deconvolution microscopy.
Signal, specimens, and computation
Because light is divided among angular samples, signal-to-noise and reconstruction quality matter alongside nominal resolution. Scattering can complicate performance, and the 2013 model was aimed at largely transparent, incoherently emitting samples. In biological imaging, fluorescent labels may also raise photobleaching and phototoxicity concerns. These are factors to assess for the specific setup and specimen, not inevitable outcomes of every LFM experiment. Review literature discusses such practical constraints, including spatial resolution, signal-to-noise, scattering, and processing speed (2025 biomedical light-field imaging review).

When researchers use light-field microscopy
LFM is useful when capturing a changing 3D scene in one exposure is more important than acquiring a higher-detail sequence of scanned planes. A scanless volume can help when the scene changes too quickly for sequential z-stack acquisition to represent a single moment. Reviews describe research applications including neuronal activity, live-cell imaging, worm locomotion, and single-molecule observations (Kim’s 2022 review of single-shot light-field microscopy). These examples do not imply that every LFM configuration will provide suitable resolution or signal for every specimen.

- Temporal sampling: Does the experiment need a volume from one exposure, or can it tolerate sequential plane scanning?
- Required detail: What lateral and axial resolution is needed, including away from the native object plane?
- Signal: Will the available light support useful angular samples and a stable reconstruction?
- Specimen: Is it fluorescent, weakly scattering, thick, or strongly scattering, and how might those properties affect the optical model?
- Illumination and sample health: For labeled biological samples, are photobleaching and phototoxicity acceptable?
- Post-capture workflow: Are the computing resources and reconstruction time acceptable for the experiment?
For a lab considering a build, the microlens array is not a universal add-on. The primary method describes matching the array’s f-number to the objective’s numerical aperture and arranging the sensor or relay optics accordingly. Choosing components requires an optical design; a generic array should not be treated as a drop-in upgrade for an existing microscope.
