Optimize a biologics process by designing it around the product’s intended quality, using risk-based studies to learn which materials and process variables matter, and verifying that the resulting controls remain suitable at the intended manufacturing scale. Scale-up is not simply a matter of copying setpoints: changes in equipment, site, materials, or operating context can affect whether the process continues to deliver the desired product.
What does quality-focused, scalable process development involve?
It is a connected development chain: define the intended product quality, identify the attributes and process factors that could affect it, study the important relationships, establish justified controls, and carry the resulting knowledge into scale-up and lifecycle validation. Quality and scalability are therefore related objectives, not separate targets to optimize in isolation.
The framework is grounded in FDA-adopted ICH guidance. Q8(R2) Pharmaceutical Development provides the pharmaceutical-development and quality-by-design (QbD) framing; Q9 supplies the risk-management framework; and Q10 describes a pharmaceutical quality-system model. FDA’s May 2026 page lists the final Q8, Q9, and Q10 Questions and Answers (R5), intended to clarify implementation of those guidances.
These principles do not prescribe universal cell-culture settings, purification conditions, assay panels, acceptance limits, or scale-up ratios. Those choices depend on the molecule, expression platform, unit operations, development stage, commercial scale, and applicable jurisdictions.

How should development start from intended product quality?
Define the product and its quality goals
Begin with the product’s intended quality profile, then identify candidate quality attributes that development and manufacturing need to understand and protect. FDA’s Q8(R2) guidance supports connecting the desired product profile to quality attributes, process variables, development studies, and the proposed control strategy.
For biological products, FDA’s document index lists ICH Q6B, which addresses specifications and testing. Q6B can inform the relevant specifications and testing context, but it does not make one analytical panel or acceptance limit appropriate for every biologic.
Build a scientific rationale
Document why an attribute or process variable matters, what evidence supports its classification, and what uncertainties remain. A list of parameters without a rationale does not explain how the process protects product quality. FDA’s Q11 Development and Manufacture of Drug Substances addresses process understanding for drug substances, impurity-reduction steps, and information for relevant CTD sections.

How do you decide what to study or control?
Use risk assessment to prioritize learning
Risk assessment helps focus development on material attributes and process parameters that could affect product quality. FDA’s Q8/Q9/Q10 implementation material also identifies interactions, equipment, and scale as relevant considerations. Use prior knowledge to guide study design, but assess whether it applies to the specific product and process rather than assuming it transfers unchanged.

For each important risk, record the reasoning behind the criticality decision, the evidence gathered, and any residual risk. A variable’s importance may depend on interactions with other variables or on the equipment and scale in which it operates.
Design studies to resolve uncertainty
Choose experiments that can distinguish the effects of plausible material attributes and process variables, including interactions where relevant. The purpose is not to generate a large volume of data for its own sake; it is to learn enough to justify controls and understand the process at the scale and context that matter.

The specific study design and operating ranges cannot be set from a general framework alone. They require product- and process-specific evidence, including the relevant development phase and manufacturing context.
How do you scale up while maintaining quality?
Treat scale-up as a change in process context
A control strategy that works in one setting may not remain suitable after a change in scale or operating environment. FDA’s Q8/Q9/Q10 implementation material identifies differences in equipment, facilities or sites, raw-material source or lot, personnel capability, and technology experience as factors that can affect control-strategy suitability.

Use relevant prior process knowledge and appropriate scale-up studies to assess these differences. Matching a nominal setpoint alone does not establish that the process is comparable or that the control strategy remains adequate. The evidence should address the particular differences and risks introduced by the intended scale-up.

Verify design-space suitability at the relevant scale
FDA’s implementation material says the entire design space does not necessarily have to be re-established at commercial scale, but its suitability should be initially verified before commercial manufacture. Further verification may be appropriate after changes such as a site, scale, or equipment change; risk assessment should guide the need and extent.
Design-space verification and process validation are distinct activities. Verification addresses whether the design space is suitable in the relevant manufacturing context; validation addresses whether the manufacturing process can consistently meet requirements under the applicable approach.

FDA’s January 2011 Process Validation: General Principles and Practices covers biological products. The ICH implementation material describes traditional process validation, continuous process verification (CPV), or a combination. It does not establish one universal validation recipe for every biologic.

The table describes options identified in the guidance framework; it is not a recommendation that one approach is preferable for all products. The choice and supporting evidence depend on product and process complexity, prior knowledge, remaining risk, and applicable regional requirements.
How does process understanding carry through the product lifecycle?
Development knowledge should remain useful after transfer to manufacturing. The control strategy, rationale for criticality decisions, scale-up evidence, and known uncertainties provide context for interpreting manufacturing performance and quality information. FDA describes ICH Q10 as a model for an effective pharmaceutical quality system, which supports lifecycle decisions and continual improvement.
Ongoing process-performance and quality monitoring can help identify when observed performance warrants investigation or further action. The monitoring plan and response should be appropriate to the product and process; the cited guidance framework does not establish universal alert limits or a one-size-fits-all monitoring set.
What should be confirmed before applying the framework?
The FDA guidance provides a framework rather than product-specific operating instructions. Before translating it into development or filing decisions, establish the details that change the technical and regulatory answer:
- Product and platform: the biologic modality and expression system.
- Process scope: upstream, downstream, formulation, or integrated manufacturing, including the unit operations involved.
- Development context: the phase, intended scale, equipment, facility, and whether a site or process change is planned.
- Regulatory context: the target jurisdictions and their applicable current requirements.
FDA’s Q8/Q9/Q10 training appendix dates to August 2012. It remains useful for its implementation discussion of risk-based scale-up, design-space verification, and lifecycle validation, while FDA’s May 2026 R5 listing is the newer clarification status for the Q8/Q9/Q10 questions and answers. Confirm current regional guidance before making filing recommendations; QbD does not remove regional requirements or GMP expectations.
