Iodine can play two different roles in complex organic chemistry: it can become part of an organic molecule, or it can be part of a reagent that transforms a molecule without necessarily remaining in the product. The distinction matters because direct iodination and hypervalent-iodine oxidation are different strategies, with different purposes and evidence.

Two meanings of iodine in organic chemistry

An organoiodine compound has iodine bonded within an organic structure. A hypervalent iodine reagent, by contrast, uses iodine in a higher oxidation state—commonly iodine(III) or iodine(V)—to bring about a chemical transformation. The iodine-containing reagent may be changed or consumed during that reaction; its presence does not mean the product will contain iodine.

Hypervalent iodine compounds are established reagents for selective oxidative transformations, and the field also includes catalytic applications. Their role is therefore not interchangeable with simply attaching an iodine atom to a carbon framework. Wiley’s overview of polyvalent iodine compounds discusses this reagent chemistry.

How iodine can help make a more complex molecule

Use a hypervalent iodine reagent to transform a substrate

In an oxidative transformation, the reagent helps change the structure or bonding of another organic molecule. This approach is useful when the desired outcome is a selective transformation; the aim is not necessarily to make an iodinated product. Hypervalent iodine chemistry also has applications in catalysis, as discussed in a review of cyclopropanes and hypervalent iodine reagents in synthesis and catalysis (PubMed).

Install iodine directly into an organic scaffold

Direct iodination instead puts iodine into the product. That iodine-bearing intermediate can be useful as a starting point for further synthesis. One specific example is a 2024 study by Thotathil and coauthors, which converted coumarin-3-carboxylic acids into 3-iodocoumarins by decarboxylative iodination. The abstract describes heating the acids with molecular iodine and potassium hydrogen phosphate in acetonitrile. The paper reports isolated yields of 46–93% across its reported products; those results vary by substrate and are not a general yield expectation for iodination. Some substrate classes gave poor results or failed. Read the study.

The authors describe 3-iodocoumarins as precursors to more complex coumarin-containing compounds. They frame molecular iodine as an inexpensive and environmentally benign option in the context of their method; that characterization should not be generalized into a claim that iodine chemistry as a whole is safe or sustainable.

Where organic iodine occurs beyond synthesis

Biological molecules

Iodine also occurs in naturally relevant organic molecules. The World Iodine Association describes iodine as capable of bonding to carbon, oxygen, or hydrogen in organic molecules and gives thyroid hormones as an example of complex organic iodine compounds. Coastal atmospheric aerosols

Organic iodine compounds have also been detected in coastal atmospheric aerosol. A 2020 study by Yu and coauthors used nontarget mass-spectrometric analysis of size-resolved aerosol samples collected during iodine nucleation events and reported 440 molecular formulas of iodinated organic compounds. The figure refers to formulas, not 440 fully structurally identified compounds. The study proposed that certain oxygenated or nitrated organic species contributed to particle growth; this is an atmospheric chemistry finding, not evidence that these compounds are biological molecules or synthetic reagents. Which iodine chemistry is relevant to a particular question?

Approach or context What iodine does Example or evidence
Hypervalent iodine reagent Helps drive an oxidative transformation; iodine need not remain in the product. General synthetic chemistry described in Wiley’s overview.
Direct carbon iodination Is incorporated into the organic product. The 2024 coumarin study reports 3-iodocoumarins, with substrate-dependent results.
Biological organoiodine Is part of an organic molecule found in a biological context. Thyroid hormones are an example cited by the World Iodine Association.
Atmospheric organoiodine Is present in organic compounds detected in aerosol. The 2020 coastal study reports 440 molecular formulas in samples collected during iodine nucleation events.

These categories answer different questions: what a reagent does in a reaction, whether iodine is present in a product, or where iodine-containing organic compounds occur. They should not be treated as one reaction family.
