A laboratory study published in 2024 offers one possible answer to a puzzle about early life: how could microbes use molybdenum-dependent nitrogenase when oxygen-poor ancient oceans had little dissolved molybdenum? The study found that the modern bacterium Rhodopseudomonas palustris could obtain molybdenum from the mineral molybdenite and use it for nitrogen fixation under anoxic laboratory conditions. That makes mineral-derived molybdenum a plausible source—not proof that this was how nitrogen fixation worked across the Archean Earth.
Why nitrogen fixation created a molybdenum puzzle
Nitrogen is essential to proteins and nucleic acids, but atmospheric nitrogen gas (N₂) is chemically stable and unavailable to most organisms in that form. Biological nitrogen fixation converts N₂ into ammonia, which life can use. The predominant enzyme for this process, molybdenum nitrogenase, requires molybdenum.
The difficulty is that dissolved molybdenum was scarce in low-oxygen conditions. Yet a 2015 study of ancient rocks placed molybdenum-nitrogenase-mediated nitrogen fixation by 3.2 billion years ago, according to a 2024 report by Chemistry World. That historical inference raises a question: what could have supplied the metal before oxygen-rich conditions made it more available in the environment?

The study, published in Earth and Planetary Science Letters on 1 December 2024, paired the anoxygenic phototroph R. palustris with molybdenite (molybdenum disulfide, MoS₂) under anoxic conditions. The researchers tested whether the bacterium could mobilize molybdenum from the mineral and use it to fix nitrogen.

They assessed nitrogen fixation using acetylene-reduction and ¹⁵N-labelled methods, alongside analyses of trace-metal mobilization and uptake, metallophore production, proteins, microscopy, and mineral surface chemistry. The paper reports that nitrogen-fixation rates increased in correlation with molybdenite concentration; the accessible report does not provide a numerical rate or effect size.
How the bacterium accessed molybdenum
The study reports that R. palustris secreted two molybdenum-binding compounds, rhodopetrobactin A and B, and expressed molybdenum transport proteins. Together, these findings support a mechanism in which the bacterium mobilizes mineral-bound molybdenum and brings it into the cell. Surface-sensitive analyses also detected changes to the molybdenite after interaction with cells.

In this proposed chain, the mineral is not itself the nitrogen-fixing enzyme’s usable metal source until the organism makes molybdenum available for uptake. The observed concentration correlation is consistent with the idea that more molybdenite can support greater nitrogen fixation under the tested conditions.

What the result says about early Earth—and what it does not
The experiment is a laboratory demonstration that molybdenite can supply molybdenum for nitrogen fixation by this modern model organism in an anoxic setting. It gives a plausible mechanism for reconciling a Mo-dependent enzyme with environments where dissolved molybdenum was limited.

It does not show that R. palustris lived on early Earth, establish how common mineral-derived molybdenum was, or prove that this pathway was ecologically important in ancient environments. Nor does it rule out other sources. Eva Stüeken, a University of St Andrews geochemist and co-author of the 2015 study, told Chemistry World that the results “definitely offer a plausible mechanism for life to obtain molybdenum on the Archean Earth.” She also noted that deep-sea hydrothermal vents remain another potential source to investigate.

These possibilities are not mutually exclusive. The experiment addresses whether one mineral-based pathway can work; it does not determine which sources dominated in particular ancient environments.

Why the finding matters
The study connects a mineral interaction to a central biological process: a microbe obtains a scarce metal and uses it in an enzyme that makes nitrogen biologically available. That is a concrete mechanism for further testing against geological evidence. The result is important because it makes one proposed solution experimentally plausible, while leaving the scale and prevalence of the process on early Earth unresolved.
