Yes—but the clearest evidence is about skeletal muscle, not a general boost to every cell. Leucine is an essential amino acid that becomes part of proteins and also acts as a nutrient signal linked to the muscle protein-building process. That short-term signaling response is not proof that leucine supplements reliably add muscle, strength, or athletic performance over time.
What leucine does in the body
Leucine is one of the branched-chain amino acids (BCAAs), alongside isoleucine and valine. It is essential, meaning the body cannot make enough of it and must obtain it from food. Like other amino acids, leucine is incorporated into proteins. In skeletal muscle, it also has a signaling role associated with the regulation of protein translation—the process by which cells assemble proteins.
That second role is the basis for saying leucine does more than supply a building block. It does not mean leucine is a universal cellular fuel or that it switches every cell into a higher-performance state. The best-supported account concerns nutrient signaling and muscle protein synthesis.
How leucine signaling relates to muscle protein synthesis
Research describes leucine and insulin-related signals as inputs that converge on mTOR-related machinery. One key component is mTORC1, which regulates parts of the translation process. Downstream regulators discussed in this context include 4E-BP1 and S6K1. In broad terms, these pathways help regulate whether a muscle cell initiates protein production.

Human studies of leucine-enriched essential amino acids, particularly in post-exercise nutrition, have found acute increases in muscle signaling and protein synthesis. These are measured responses over a limited period after nutrient intake and exercise, not evidence by themselves of lasting muscle growth or improved performance. The precise way human muscle senses and responds to nutrients is not fully resolved; mechanistic explanations draw on both human research and experimental work.
A 2023 review reported that phosphorylation of mTORC1 substrates was about three times greater with leucine than with the other essential amino acids in the evidence it assessed. That is a review-reported molecular comparison, not a finding that leucine produces three times as much muscle growth. The same review discusses the transient nature of protein synthesis after feeding and the influence of exercise, age, and inactivity on muscle response: the 2023 review in Clinical Nutrition.

Experimental cell research also helps explain possible pathways. A 2007 study in C2C12 mouse muscle cells reported changes in AMPK and mTOR signaling and increased measured protein synthesis after leucine treatment. Results in a cell model provide mechanistic background; they do not establish the size of an effect in people: the C2C12 cell study.

A rise in a signaling marker or short-term muscle protein synthesis is one step in a larger process. It does not establish that taking leucine alone will lead to more muscle, greater strength, faster recovery, or better athletic performance after weeks or months.

The NIH Office of Dietary Supplements says studies have not consistently shown that BCAA supplements—or any one of the three BCAAs taken singly—enhance exercise performance, build muscle mass, or aid recovery. Its fact sheet summarizes the evidence and distinguishes these outcomes from claims based on the amino acids’ biological roles: NIH ODS, Dietary Supplements for Exercise and Athletic Performance.
A critical review of resistance-exercise nutrition found no differences in strength or muscle-mass outcomes between protein types in the long-term resistance-training evidence it assessed when the stated leucine threshold was reached. That conclusion belongs to the studies covered by the review; it is not a universal threshold or a personalized dosing rule: the 2016 critical review.

Food protein versus leucine supplements
Complete protein foods provide leucine together with the other essential amino acids needed to make proteins. NIH ODS notes that whey contains more leucine than casein or soy. A whey protein powder can therefore be one convenient complete-protein source, but it is optional—not a requirement for someone who already gets adequate protein from food or other sources.
Isolated leucine powder supplies leucine without automatically supplying the full range of essential amino acids. The evidence does not establish that people who already consume sufficient complete protein need an isolated leucine supplement. If the practical question is whether to use a supplement, consider whether your overall diet supplies adequate protein and whether a clinician or qualified dietitian has advised supplementation for your circumstances.

What the safety figures do—and do not—mean
NIH ODS reports that up to 20 g per day of BCAA supplements in divided doses appears safe. It also cites an upper safe leucine intake of 500 mg per kg of body weight per day in studies of healthy young and older men—about 38 g per day for a 75 kg man. These are source-reported safety findings, not recommended targets, effective muscle-building doses, or guidance for every person. The fact sheet does not state a publication year for these figures.
People with health conditions or dietary restrictions should seek individualized advice from a qualified clinician before using supplements. A safety limit should not be treated as a reason to take a high dose, particularly when the evidence for performance or muscle-building benefits is inconsistent.
