Electron-beam imaging is a group of microscopy methods that forms images from signals produced when a focused beam of electrons interacts with a specimen. Depending on the method, electrons may scan a surface or pass through a thin sample, revealing surface features, internal structure, or—when analytical detectors are used—information about composition and bonding.

How does an electron microscope work?

An electron source generates and accelerates electrons in a vacuum. Apertures and electromagnetic lenses shape and focus the beam. When it reaches the specimen, the interaction changes or produces signals, which detectors collect; software maps those measurements to image data.

The details depend on the microscope mode. In scanning electron microscopy, the beam moves across the surface point by point, and detectors can collect emitted secondary electrons. In transmission methods, electrons that pass through or scatter from a thin specimen are measured. The U.S. Department of Veterans Affairs describes 5–100 keV as a typical accelerating-energy range, but that is a general description, not a specification that applies to every instrument.

What is the difference between SEM, TEM, and STEM?

Mode How the beam interacts with the sample What it is commonly used to examine
SEM (scanning electron microscopy) A focused beam scans the specimen surface; detectors collect signals such as secondary electrons. Surface shape and texture, and, with suitable analytical detectors, spatially resolved composition.
TEM (transmission electron microscopy) Electrons pass through an electron-transparent specimen to form an image or diffraction pattern. Internal structure, defects, and structural or compositional variation.
STEM (scanning transmission electron microscopy) A fine focused probe scans a thin specimen, while detectors measure transmitted or scattered electrons at each position. Localized transmission contrast and analysis across a thin sample.

SEM is the surface-scanning choice; TEM and STEM examine electron-transparent samples. STEM uses a scanning probe in transmission geometry, so its image is assembled from measurements made as the probe moves across the specimen. What information can electron-beam imaging provide?

An image records contrast from electron signals, but microscopy can also include analytical measurements. Energy-dispersive X-ray spectroscopy (EDS, also called EDX) can provide elemental information. Electron energy-loss spectroscopy (EELS) can provide compositional information and, in suitable circumstances, information about bonding or oxidation state. Diffraction can reveal structural information, while tomography can be used to reconstruct three-dimensional features.

These measurements depend on the instrument, detector configuration, sample, and operating conditions. An electron microscope does not automatically produce a chemical map, a three-dimensional reconstruction, or an atomic-resolution image. In STEM, for example, bright-field, dark-field, and high-angle annular dark-field detectors collect different ranges or directions of scattered electrons, producing different kinds of contrast.

When is each method useful?

  • Choose SEM when the question is about a specimen’s surface, such as its topography or texture. With suitable detectors, SEM can also support spatially resolved compositional analysis.
  • Choose TEM when the question concerns the inside of a thin, electron-transparent specimen, including internal structures, defects, or diffraction patterns.
  • Choose STEM when scanning localized regions of a thin specimen in transmission is useful, especially when image contrast and analytical measurements need to be collected across the same area.
  • Use spectroscopy or diffraction alongside imaging when the question is chemical or structural; an image alone does not necessarily identify elements, bonding, or crystal structure.

Examples described by Arizona State University’s Eyring Materials Center include biological TEM studies of cell interiors and biological complexes, as well as materials investigations of crystal defects, structure, and composition. Its facility also lists SEM surface work and TEM/STEM capabilities such as elemental mapping, diffraction, and 3D imaging. Those examples describe facility capabilities, not results guaranteed for every specimen or microscope.
