The gut–brain axis is the two-way communication network between the digestive system and the brain. It includes nerve signals, hormones, immune activity and, in the microbiota–gut–brain axis, signals from gut microbes and the chemicals they produce. These pathways may be relevant to mood, stress responses, cognition and neurological health, but evidence linking microbes to a condition does not by itself show that they caused it—or that changing them will prevent or treat it.

What is the gut–brain axis?

The gut–brain axis is the network through which the digestive system and brain send and receive information. The National Institute on Aging describes it as an intricate network relaying messages between the digestive system and brain. The broader term includes communication between gut and brain generally; the narrower term microbiota–gut–brain axis emphasizes the role of gut microbes and their products.

Gut microbes are one part of the picture, not the whole explanation. The gut microbiome helps digest food, make nutrients and support immune function; its chemicals can also affect brain function. At the same time, signals from the brain can influence gut activity and microbial composition. A 2025 review by Ohara and Hsiao in Nature Reviews Microbiology emphasizes this two-way relationship.

How do messages travel between the gut and brain?

Communication can involve several routes at once. The gut detects information about its contents and condition, and the nervous system, hormones and immune signals help relay or shape it. Researchers use the term interoception for the nervous system’s sensing, interpretation and integration of signals from inside the body, including the gastrointestinal system.

Nerves and sensory signals

Sensory pathways carry information from the gut toward the central nervous system. These include vagal and spinal sensory inputs. The gut’s sensory system also includes chemosensory epithelial cells connected with sensory nerve fibers, a route highlighted in the 2025 review. Such signals provide information about conditions inside the digestive tract; they do not mean that every effect on the brain is caused by microbes.

Hormones, immune signals and microbial products

Intestinal endocrine cells release hormones that can participate in gut–brain communication. Immune activity and chemicals produced by microbes are other potential messengers. Stress-related signals can also affect gut function, illustrating the brain-to-gut direction of the network. The National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK) workshop summary describes these interacting routes, while noting that their operation is not fully mapped in humans.

How might the gut–brain axis relate to brain health?

Because gut signals can reach systems involved in brain activity and behavior, researchers are studying possible links with mood, stress responses, cognition and neurological health. Reviews discuss reported microbiome associations with depression, anxiety, Alzheimer’s disease, Parkinson’s disease and other conditions, as well as candidate processes involving immune signaling, microbial metabolites and neural communication.

These are areas of investigation, not evidence of one disease-causing “dysbiosis” pattern. The 2025 review indexed by PubMed says more work is needed to establish whether microbial changes cause or contribute to disorders and to develop evidence-based gut-focused recommendations for prevention or management. A finding that two things occur together is not proof that one caused the other, and the direction of influence can be difficult to determine.

What does the evidence show—and what does it not show?

Different kinds of findings answer different questions. Mechanistic experiments can reveal plausible pathways; animal studies can test those pathways in a living organism; observational human studies can identify associations. None of those findings alone demonstrates that changing the microbiome will prevent or treat a brain disorder. NIDDK identifies a need for direct human evidence about altered gut input to the central nervous system, as well as human interventions that specifically modulate it.

Mechanistic research How a proposed signal or pathway might operate. That it causes a clinical condition in people.
Animal studies Whether a pathway can affect an outcome in the studied animals. That the same mechanism causes the same outcome in humans or that a treatment works for people.
Observational human studies Whether gut or microbiome features are associated with a condition in the people studied. Which factor came first, whether one caused the other, or whether changing the feature will improve health.
Human intervention studies Whether a specified intervention affects a measured human outcome under the study conditions. A general benefit beyond the intervention, population and outcome actually studied.

One NIDDK workshop executive summary from April 30–May 1, 2024, reports that in one study about two-thirds of participants had gastrointestinal disorders preceding mood disorders, while one-third had mood disorders preceding gastrointestinal disorders. These are observations from that study, not population estimates, and they do not establish causation.

What did a 2026 mouse study find about gut bacteria and memory?

An NIH Research Matters report dated April 28, 2026, describes a study in mice. In the aged animals examined, the bacterium Parabacteroides goldsteinii was increased. Exposing mice to the bacterium or to its fatty-acid products was linked to inflammation, impaired vagal signaling from gut to brain, reduced hippocampal activity and worse performance on memory tasks. Researchers also tried experimental approaches to restore signaling.

This is preclinical evidence, not proof that this bacterium causes memory loss in people or that an intervention can restore human memory. NIH says that if the findings hold in humans, they could inspire future approaches; the report does not establish a human treatment.

Should you use a microbiome test or supplement for brain health?

The evidence summarized here does not establish a probiotic, prebiotic, commercial microbiome test or other microbiome product as a proven way to protect brain health or prevent or treat a brain-health condition. A microbiome association does not show that a product will alter the relevant pathway or improve a meaningful health outcome. If you have concerns about digestive symptoms, mood, memory or another health issue, discuss them with an appropriate health professional rather than relying on a test or supplement to explain or treat them.
