Senolytics are intended to kill senescent cells; senomorphics are intended to change harmful effects of those cells, often by reducing their secretions, without necessarily removing them. Both are experimental research approaches—not proven, general anti-aging treatments. The distinction is the intended outcome, not a guarantee that a compound acts only on senescent cells or only on one pathway.

What are senescent cells?

Cellular senescence is a cell state, not another word for aging. After stress or damage, a cell may stop dividing while remaining biologically active. Some senescent cells release a changing mix of signals called the senescence-associated secretory phenotype, or SASP. These can include inflammatory and tissue-remodeling factors.

Senescence has useful roles: it can help with wound repair and suppress tumor growth. But senescent cells that persist in certain contexts may contribute to inflammation and tissue dysfunction. The therapeutic question is therefore not simply whether a cell is senescent, but whether that particular population is harmful and can be targeted without disrupting useful functions.

The SASP is not one fixed substance. Its composition varies by cell type, tissue, cause, environment, and time. A 2021 National Institute on Aging workshop report described the SASP as involving more than 400 proteins; that figure is from the report, not a count that applies uniformly to every senescent cell or tissue. How do senolytics and senomorphics differ?

These are conceptual categories, not guarantees about a drug’s full effects. Compounds can affect multiple pathways, and a label does not establish clinical benefit. Both approaches face the challenge of identifying which senescent cells matter, confirming that a treatment hits its intended target, and demonstrating safety and meaningful outcomes.

How do senolytics work?

Senescent cells can resist apoptosis, the process of programmed cell death, through senescent-cell anti-apoptotic pathways (SCAPs). Senolytic research aims to disrupt survival mechanisms on which some senescent cells depend, making them more likely to die. Candidate targets include BCL-2-family proteins and other pro-survival networks.

“Selective” describes the goal, not a promise of perfect precision. Healthy cells may also use some of the same survival pathways, and different senescent-cell populations may rely on different mechanisms. Dasatinib, quercetin, and fisetin are examples discussed in early senolytic research; their mention does not make them established anti-aging medicines or personal treatment recommendations.

How do senomorphics work?

Senomorphics aim to change harmful effects of senescent cells rather than clear the cells. Many approaches focus on reducing or reshaping the SASP by modulating signaling pathways such as mTOR or JAK. Because the SASP varies, changing one pathway may not suppress every harmful output. And reducing SASP signaling does not show that the senescent cells themselves have been removed.

The NIA workshop report describes a practical distinction for research design: senolytics may be studied using intermittent exposure, while senomorphics that need to keep suppressing cell behavior may require continuous administration. That is a research consideration, not dosing advice; sustained use would require a strong long-term safety case.

Why context and cell diversity matter

Senescent cells are not a single, uniform target. Their characteristics and secretions differ across tissues and causes, and can change over time. NIH’s Cellular Senescence Network (SenNet) is developing maps and methods to characterize that variation. In a June 2026 release, NIH described a “senotype” framework that groups senescent cells according to where they occur and the conditions around them. Read the NIH announcement.

This diversity complicates both strategies: a senolytic may miss cells that do not rely on its target pathway, while a senomorphic may leave harmful SASP components untouched. Nicole Kleinstreuer, Ph.D., NIH Deputy Director for Program Coordination, Planning, and Strategic Initiatives, described the goal as mapping senotypes to support more targeted therapies that focus on harmful cells while preserving beneficial ones. That is a research objective, not evidence that such therapies are already available or proven.

What is known about human benefits?

Preclinical studies, including animal research, have motivated work on these approaches, but animal outcomes do not establish benefit in people. NIH describes senolytics as experimental drugs and notes that human trials are underway while important questions remain before widespread use. See the NIH Common Fund overview. The evidence summarized here does not establish that either approach extends human lifespan or provides general anti-aging benefits, nor does it establish that one is clinically superior to the other. Any reported clinical effect needs to be evaluated for the specific compound, condition, participants, and outcome studied.

What safety questions remain?

  • Collateral effects: A compound may affect healthy cells that use the same survival pathways as senescent cells.
  • Loss of useful cells: Removing cells indiscriminately could interfere with wound healing, tissue repair, or tumor suppression.
  • Cancer and immune context: Potential concerns include impaired cancer immunosurveillance and the possibility of cell-cycle reentry by senescent cancer cells.
  • Long-term exposure: Sustained senomorphic treatment raises questions about safety over time, and a single agent may not address every SASP component.
  • Individual health circumstances: Trials need to account for multimorbidity, polypharmacy, drug–disease interactions, and contraindications, especially in older adults.
  • Measurement: Better markers are needed to identify specific senescent cell types, estimate burden, confirm target engagement, and monitor response.

These issues are why neither “kill senescent cells” nor “turn off the SASP” is a sufficient treatment plan by itself. The target, tissue, timing, and potential effects on normal functions all matter.
