To make biological research data reusable, document enough for someone else to identify the dataset, understand its biological subject and context, interpret how it was produced, assess access and reuse conditions, and connect it to related resources. Start with the FAIR principles—Findable, Accessible, Interoperable, Reusable—then use the metadata standard suited to your research community. FAIR is guidance, not a universal schema or a requirement to make every dataset openly accessible.

What metadata makes biological data reusable?

Metadata is information that describes data and the context needed to interpret them. A useful record goes beyond a title: it explains what was observed or measured, how the work was done, what each value means, where the data came from, and how another researcher can access and cite them.

Use this cross-domain checklist as a starting point, then add fields required by the relevant community standard and repository. It is not a universal set of fields mandated by FAIR.

  1. Identify and cite the dataset. Provide a globally unique persistent identifier, title, creators or responsible organization, release or publication date, version, and a citation or link to the data record. Make clear which dataset the metadata describes.
  2. Describe the biological subject and context. Record the organism or taxon, material, sample or occurrence identity, relevant location and time, and biological context needed to interpret the study. Choose terms that fit the type of record.
  3. Explain methods and acquisition. Document experimental or observational design, collection and sampling procedures, measurement methods, and relevant instruments or computational workflows. Describe processing and transformation steps as well.
  4. Define variables and formats. Explain fields and variables, units, allowed values or controlled vocabularies, file formats, and relationships needed to interpret the data. Use shared vocabularies and qualified references to related data where appropriate.
  5. Record provenance and versions. State who created or changed the data, how they were processed, which source records or samples they derive from, and which dataset version the record describes.
  6. State access and reuse conditions. Tell readers where and how to retrieve the data, whether authentication or authorization is required, what restrictions apply, and which data-use license governs reuse. Keep descriptive metadata accessible even if files are restricted or later removed.
  7. Link related resources. Use explicit, qualified references to publications, protocols, code, samples, instruments, and related datasets rather than leaving those connections implicit.
  8. Name the standards and versions used. Identify the schema, vocabulary, or community standard applied, including its version when relevant. In life sciences, this choice affects whether other researchers can interpret and connect the records.

How FAIR principles guide the metadata

FAIR stands for Findable, Accessible, Interoperable, and Reusable. The principles describe qualities that make data easier to locate and use; they do not prescribe one software implementation or one metadata schema. The GO FAIR Foundation’s guiding-principles page emphasizes reuse as the goal.

  • Findable: Give the dataset a persistent identifier and provide searchable, adequately described metadata.
  • Accessible: Explain how to retrieve the data and any authentication or authorization requirements. Accessible does not necessarily mean unrestricted or open.
  • Interoperable: Use shared formats, vocabularies, and references so data can be connected and interpreted alongside other data.
  • Reusable: Provide accurate attributes, provenance, licensing, methods, and community-standard alignment so potential users can judge whether and how the data fit their work.

The original FAIR paper, “The FAIR Guiding Principles for scientific data management and stewardship” (2016), and the current GO FAIR guidance both support treating these principles as a framework rather than a fixed field list. In particular, data access may be restricted while descriptive metadata remains retrievable.

Which metadata standard fits your biological data?

Choose a standard based on the biological object and data type, the research community and repository, the context and provenance needed for intended reuse, interoperability with adjacent disciplines, support for access and licensing information, and the standard’s maintenance status and version. These are practical comparison considerations, not a validated scoring system.

Life-sciences workflows across domains

ISO 20691:2022, Biotechnology — Requirements for data formatting and description in the life sciences, addresses consistent formatting and documentation of data and corresponding metadata across life-sciences research and development. Its scope includes manual and computational workflows, experimental or procedural data, machine-derived data, and datasets of different sizes. It names genomics, metagenomics, transcriptomics, proteomics, metabolomics, synthetic biology, and systems biology among its applications.

ISO 20691 is a formal reference for broad life-sciences documentation, not a claim that every researcher must buy or implement it. The official ISO preview identifies the first edition as published in November 2022 and lists paper and digital formats.

Biodiversity occurrences, specimens, and samples

Darwin Core, maintained by Biodiversity Information Standards (TDWG), is a glossary of terms for sharing biodiversity information. Its focus includes taxa and their occurrence in nature as documented by observations, specimens, samples, and related information. Its stated scope does not cover non-biodiversity data or purely taxonomic data, so it is not a universal metadata standard for biology.

Simple Darwin Core is a predefined subset of terms commonly used in biodiversity applications. The cited version was issued on 2023-09-13 and is designed for simple sharing structures such as rows and columns. It has no mandatory fields: select terms that make each record meaningful for its purpose rather than treating flexibility as permission to omit needed context.

Biodiversity data with sequence or omics information

Datasets that combine biodiversity and sequence information may need terms from more than one community standard. A 2023 paper, “Aligning Standards Communities for Omics Biodiversity Data: Sustainable Darwin Core-MIxS Interoperability”, describes work convening TDWG, the Genomic Standards Consortium, and other stakeholders. The group produced mappings between Darwin Core and Minimum Information about any (x) Sequence (MIxS) keys and a MIxS-DwC extension intended to incorporate MIxS core terms into Darwin Core-compliant metadata. This is a documented interoperability approach, not evidence of universal adoption or a solution to every interoperability issue.

How to apply the checklist to a dataset

  1. Define the intended reuse. Identify what another researcher should be able to understand or do with the data. The biological context, methods, variables, and provenance to document depend on the research object and the intended use.
  2. Choose the relevant community standard. Check its scope, fit with your repository and research community, coverage of necessary context and methods, connections to adjacent standards, and current version. Use a domain standard alongside broad guidance where needed.
  3. Complete the record, not just the file description. Add identifiers, citations, biological context, methods, variable definitions, provenance, versions, formats, access conditions, and relationships to related resources. Use controlled vocabularies and qualified references where suitable.
  4. Check that the record supports access and interpretation. A prospective user should be able to find the dataset, understand how to obtain it and any restrictions, interpret its values and methods, and identify applicable reuse terms.
  5. Preserve the metadata record. Keep descriptive metadata retrievable even if the underlying files are restricted or later removed, so the dataset remains identifiable and its conditions are clear.
