Scientists do not test for extraterrestrial life with a single detector. They combine geological context, chemical measurements and, where possible, microscopic observations, then ask whether the evidence fits biology better than nonbiological chemistry. Finding a place that could support life, detecting a possible biosignature and confirming life are three different steps.

What counts as evidence of life?

A biosignature is a feature that may indicate life. It is not proof by itself: nonbiological processes can produce some of the same molecules and patterns. Scientists interpret candidate signals alongside the environment where they were found and look for independent observations that support the same explanation.

NASA’s Ladder of Life Detection is a framework for organizing possible indicators and measurements, not a pass-or-fail test. NASA’s 2018 discussion describes 15 features proposed by the astrobiology community as indicators of life. The number of features is not a threshold: a chemical detection does not become proof just because it matches one or more items on a list.

  • Habitability: Was there liquid water, useful chemistry and an energy source in an environment where life might persist?
  • Biosignature detection: Is there a molecule, structure or pattern that could be associated with life?
  • Confirmation: Do multiple lines of evidence hold up when scientists test nonbiological explanations and account for how the sample was collected and preserved?

As NASA Senior Scientist for Astrobiology Mary Voytek has put it, “Many of the molecules that are used by life can be formed without life.” A signal that is unexpected in its environment can be worth investigating, but its meaning depends on context and competing explanations.

What do scientists measure?

Geology and environmental context

Images and spectroscopy from orbit help scientists map terrain and identify minerals that can guide the choice of sampling sites. Landers and rovers then inspect rocks and soils in place. Mineralogy, textures and the surrounding geology can help establish whether water or energy sources were present and whether conditions might have preserved evidence over time. Context helps scientists decide both where to look and what a chemical result could mean.

Organic molecules and chemical patterns

Instruments can analyze organic molecules in rock, soil or material collected from a plume. Organic molecules are carbon-based compounds, not a synonym for organisms: NASA notes that even amino acids can form without life. Scientists therefore consider the kinds and relative abundance of molecules, how complex they are, what the local environment would produce without biology, and whether other evidence supports a biological interpretation.

Cell-scale structures and activity

Microscopy can provide a different kind of evidence from bulk chemistry by examining structures or cell-like properties in a sample. JPL’s OWLS instrument suite is designed to pair high-resolution microscopy with chemical analysis of collected water samples. That combination illustrates a complementary measurement strategy; OWLS is instrument-development work, not a report that life has been detected or necessarily a payload selected for a mission.

Unexpected chemistry and independent checks

Molecules in unusual proportions or a chemical pattern that the environment does not readily explain may strengthen a case for further study. They do not settle it. Scientists test whether geology, radiation, alteration or other nonliving chemistry could produce the same signal, and seek agreement among different measurements rather than relying on one striking result.

Sample preservation and contamination

A measurement is only as useful as the sample and its history. Scientists must consider where material came from, how radiation or chemical alteration may have changed it, whether spacecraft or handling introduced contaminants, and whether an analysis itself could damage fragile evidence. NASA’s life-detection planning explicitly treats sample fragility, destruction, environmental alteration and tolerance as questions that affect interpretation.

How the search differs on Mars and icy moons

Question Mars Europa and Enceladus
Where can samples come from? Rovers can examine exposed rocks and soils directly. The oceans are beneath ice. Researchers can investigate material brought to or ejected above the surface, such as Enceladus plume particles, or consider sampling surface ice. The subsurface ocean itself is difficult to access.
What is the main environmental question? Whether ancient environments had conditions capable of supporting microbial life, and whether rocks preserve evidence of those conditions. Whether subsurface oceans and their chemistry could provide conditions suitable for life, and whether accessible material carries clues from those environments.
What measurements are relevant? Imaging, mineralogical analysis and measurements of organic chemistry in rocks and soils. Analysis of plume or ice material can investigate chemistry; a collected water sample could also be examined at molecular and cellular scales using approaches such as those described for OWLS.
What can a result establish? Evidence about past habitability or a candidate biosignature; neither alone establishes that life existed. Evidence about ocean-related chemistry or habitability, or a candidate biosignature; chemical ingredients alone do not establish organisms.

This comparison reflects the mission descriptions and instrument concepts discussed by NASA and JPL, not a complete inventory of every proposed mission or instrument. Radiation, alteration and contamination are relevant to both settings, though the sample pathways and preservation challenges differ.

How do Mars rovers investigate possible signs of life?

NASA describes Curiosity’s work as investigating whether ancient Mars had environments capable of supporting microbial life, including the presence of water, carbon and energy. Rover imaging, mineralogy and organic-chemistry measurements help characterize the rocks and soils in their geological setting. These investigations address habitability and possible evidence; they have not confirmed life.

Perseverance also studies rocks and soils, and collects and seals cores for possible future return to Earth for detailed laboratory analysis. A returned sample could be examined with a broader range of laboratory methods, but caching samples is not itself a detection, and possible future analysis should not be mistaken for a completed result.

What can scientists learn from Europa and Enceladus?

Europa and Enceladus attract astrobiology interest because subsurface oceans may provide liquid water and chemically useful materials. Their ice shells make direct access to those oceans difficult, so scientists look for ways to study material available at or above the surface.

NASA’s Cassini mission found saltwater and organic chemicals in material from Enceladus’s plume. That is evidence of chemically interesting material, not evidence that the plume contains life. For Europa, NASA describes Europa Clipper as a habitability-reconnaissance mission, not a direct life-detection mission. A NASA Europa Lander page describes a proposed concept to sample near-surface ice for possible biosignatures; it should not be treated as an operational mission or a confirmed payload.

Instrument concepts show how future sample analysis could combine approaches. JPL’s OWLS description pairs chemical analysis with microscopy for collected water. NASA’s EMILI technical report describes an instrument concept combining chemical separation with optical and mass-spectrometry detection. These are descriptions of instrument design work, not completed detections or proof that either system has been deployed on a mission.

What would make a life-detection claim convincing?

A strong claim would require more than an organic molecule, a cell-like shape or a potentially habitable environment. Scientists would need to establish what the sample is, preserve its provenance, evaluate plausible nonbiological explanations, and show that multiple independent observations converge on a biological interpretation. The evidence must also be robust to how the sample was altered, measured or handled.
