Oxygen isotopes and trace-element ratios can help estimate past sea-surface conditions, including temperature, but they are calibrated proxies, not direct readings of ancient water. Biology, shell structure and environmental factors can all affect the signals.
How does a giant clam shell preserve a record?
Giant clams are Indo-Pacific bivalves that add shell material as they grow. Their shells preserve both visible growth patterns and carbonate whose chemical composition can be analyzed. Sclerochronology—the study of growth patterns in hard tissues that accumulate over time—helps scientists build a chronology and connect each chemical sample to a point in the clam’s life.
In practice, researchers can cut and polish a shell section, image or enhance its growth increments, and measure their spacing. They then align that physical record with a chemical sampling path, or transect, through the shell. A 2018 study of Tridacna maxima prepared two sections along the shell’s maximum growth axis, polished and imaged the increments, and matched the dated growth record to the chemistry transect. Komagoe et al., 2018.
How the chronology is assigned
In that study, researchers counted increments from the shell edge, where the collection date was known, toward older shell material. They treated the increments as daily and used the count to assign dates to positions along the chemistry transect. That is one study’s age-model procedure, not a guarantee that every shell increment forms daily or can be counted unambiguously. Fossil shells also need age control and an assessment of preservation; a 2025 review identifies fossil dating as an area requiring continued attention. What do scientists measure to estimate past ocean conditions?
Oxygen isotopes
Scientists measure the oxygen-isotope composition of shell carbonate, written δ18O. It is an established palaeoclimate proxy, but it does not provide a temperature reading on its own. Interpreting it requires calibration and consideration of seawater composition and other influences. In the 2018 T. maxima study, δ18O and δ13C were measured from shell material using a stable isotope ratio mass spectrometer. Komagoe et al., 2018.
Trace-element ratios
Researchers also examine ratios such as strontium to calcium (Sr/Ca), magnesium to calcium (Mg/Ca), barium to calcium (Ba/Ca), and iron to calcium (Fe/Ca). These proxies can address different environmental questions and do not have identical uncertainties. Growth patterns and microstructure
Growth increments provide the time framework and help researchers target samples. Microstructure also matters when interpreting chemistry: a variation in a shell measurement may reflect how the clam formed the shell as well as changes in the surrounding environment. In the 2018 study, Ba/Ca was measured along a transect using laser-ablation inductively coupled plasma mass spectrometry (LA-ICP-MS), and the chemical record was aligned with the growth increments. Komagoe et al., 2018.
Why Sr/Ca is not automatically a temperature signal
Giant-clam Sr/Ca can vary with the daily light cycle, so a change in this ratio should not automatically be interpreted as a temperature change. In a 2012 study of a cultivated giant clam, researchers measured Sr/Ca, Mg/Ca and Ba/Ca with a high-resolution secondary-ion mass spectrometer at 2 μm resolution. They reported diurnal Sr/Ca variation and proposed that light-enhanced calcification and elemental transport involving the clam and its symbiotic algae could help explain the pattern. The proposed mechanism is an interpretation of the measured signal, not a direct measurement of temperature. Sano et al., 2012.

What a fossil giant clam study shows—and what it does not
Hori and colleagues studied a fossil Tridacna gigas collected at Shiraho Coast on Ishigaki Island in southern Japan. Radiocarbon dating placed it in the middle Holocene. They measured a longer Sr/Ca profile at 50 μm resolution and performed finer-scale work at 2 μm resolution. The fossil showed repeating Sr/Ca variation resembling patterns in cultivated giant clams; the authors interpreted fine alternating bands as potential evidence of daily light-cycle variation at a multi-hour scale. Hori et al., 2015.
This is a demonstration of using a shell proxy to investigate a recurring signal, not proof that Sr/Ca directly records ocean temperature in every giant clam. The authors also reported that two winter intervals differed: the first showed variation similar to their modern cultivated samples, while the second had relatively small Sr/Ca ratios and less variation. Those observations describe that specimen and study, not a universal seasonal pattern.

How to compare giant-clam reconstructions
Two shell records are not directly comparable just because both come from giant clams. To judge what a reconstruction can support, check the evidence behind its chronology, measurements and interpretation.
- Species, location and age: Identify the clam and where and when it lived. A fossil from the middle Holocene and a modern cultivated shell do not represent the same setting.
- Chronology: Check how growth increments were identified and dated, and whether the age model is supported by collection dates or independent age control.
- Sampling resolution: Find out how closely spaced the measurements were. Fine-scale sampling used to investigate daily patterns answers a different question from a longer-profile temperature reconstruction.
- Proxy and calibration: Note whether the study measured δ18O, an elemental ratio or several signals, and what modern or instrumental observations were used to interpret them.
- Preservation and structure: Consider whether the shell’s chemistry and microstructure remain suitable for interpretation, particularly for fossils.
These distinctions matter because proxy calibration, shell microstructure and fossil dating remain challenges in the field. A fine-scale Sr/Ca study of a daily light-cycle signal should not be treated as interchangeable with an oxygen-isotope reconstruction aimed at estimating temperature.
