Embryonic genome activation (EGA), also called zygotic genome activation (ZGA), is when an embryo begins transcribing genes from its own genome. It starts at different times in different species—and “begins” does not mean the same thing as the larger wave of activity that follows. Current literature describes low-level transcription in one-cell human and mouse embryos, followed by a major wave at the four-to-eight-cell stages in humans and the two-cell stage in mice.
What embryonic genome activation means
In the earliest stages after fertilization, development relies substantially on RNA and other molecules deposited in the egg by the mother. EGA is the embryo’s transcription of its own genes. The term describes that specific activity, not the entire change in developmental control.
The broader process is called the maternal-to-zygotic transition (MZT). It includes the start of embryonic transcription as well as changes to chromatin—the structure that packages DNA—and the remodeling or clearance of maternal products. Researchers sometimes use EGA and ZGA interchangeably; MZT is the more inclusive term for the coordinated handoff.
When does EGA happen?
Timing depends on the species and on whether a study means the first detectable transcription or the larger, “major” wave. A 2025 perspective by Maki Asami and Anthony C. F. Perry distinguishes early activity from the later major wave:

The mouse and human early-onset findings and major-wave timings are summarized in the 2025 Asami and Perry perspective. The zebrafish timing comes from a comparative review of animal development. These timings should not be generalized across species: cell-stage labels and elapsed time are not interchangeable between animals.
Why sources may give different timings
Traditional summaries often place human EGA at the four-to-eight-cell stages and mouse EGA at the two-cell stage because those are the prominent waves. Evidence for low-level transcription in one-cell embryos points to earlier activity, not the disappearance of the later wave. The distinction is between onset and major activation, and recent findings about immediate EGA are an interpretation of transcriptomic evidence rather than terminology used uniformly across the field.

What changes during the maternal-to-zygotic transition?
As embryonic transcription increases, developmental control shifts progressively from maternal products toward gene expression directed by the embryo. This is accompanied by changes in chromatin and cell-cycle conditions, while maternal RNAs and other factors are remodeled or cleared. Reviews describe this as a coordinated transition; they do not establish one universal trigger that switches EGA on in every species.
In the mouse analysis discussed by Asami and Perry, 1,777 genes were reported as upregulated during immediate EGA at the study’s stated false-discovery threshold. That is a study-specific result, not a universal count of genes activated during EGA.

Why embryonic genome activation matters
Embryonic transcription supplies gene products needed as development advances. Evidence from model organisms illustrates why that activity matters: a cross-species review reports that inhibiting transcription in zebrafish and Xenopus can allow some early cell divisions but prevent embryos from gastrulating. These findings concern animal models; they do not establish an outcome for a particular human embryo or pregnancy.
