People generally cannot regrow an amputated arm or leg because human healing does not activate the coordinated, pattern-forming program salamanders use to rebuild a missing limb. Salamanders form a blastema—a growing collection of cells beneath the wound surface—and rebuild the missing tissues in the right arrangement. Humans can repair some tissues, but do not ordinarily form a salamander-like blastema after limb loss. Scientists are studying why; human limb regrowth is not a current medical treatment.
How does a salamander regrow a limb?
After an amputation, cells cover the wound and form a wound epidermis. Beneath it, dividing progenitor and other cells accumulate into a blastema. In a sequence of growth, patterning and differentiation, that developing tissue contributes to replacement skin, muscle, bone, cartilage and other structures. Signals from nerves and nearby tissues, together with positional information, help organize the growth so it reconstructs the missing part appropriate to the injury site rather than forming an unpatterned mass. A blastema is not simply a uniform pool of generic stem cells. Multiple cell populations contribute, and researchers continue to investigate how cells retain or regain suitable identities and how the correct pattern is specified.
Why is human healing different?
Human wound repair can close an injury and restore some tissue, but it generally does not reconstruct a complete limb with its coordinated bones, joints, muscles, nerves, skin and blood vessels. Most mammals, including humans, do not form salamander-like blastemas after limb loss, as the National Institute of General Medical Sciences (NIGMS) explains.

The important difference is not simply that salamanders heal faster. Their response can rebuild a patterned appendage; human repair usually closes the wound and may restore only some tissues, often with scarring, rather than rebuilding the whole limb and its function.
People do have limited regenerative abilities
Human regeneration is not all-or-nothing. Skin and hair grow back, bone can heal after a fracture, and remaining liver tissue can enlarge after part of the liver is lost. These examples, described by NIGMS, are limited tissue repair or compensation—not regrowth of an entire limb.

What biological factors may explain the gap?
No single established switch separates salamander regeneration from human healing. Researchers study several interdependent parts of the process:
- Cell sources and identity: Blastemas involve cells from multiple sources. Which cells contribute and how they acquire the right roles are active questions.
- Positional information: A replacement limb must form the structures appropriate to where it was cut. Salamander blastemas respond to positional cues; how to reproduce that outcome in mammals is not established.
- The wound environment: The wound epidermis and surrounding tissue provide a context for growth. The injury site must support regeneration, not only closure and scar formation.
- Nerve-related signals: Nerves contribute signals implicated in salamander regeneration, alongside signals from nearby tissues.
- Immune activity: Reviews discuss immune-system modulation, including macrophage roles, as part of the regeneration environment. The process is not explained by one immune-cell type alone.
These factors work together; the available evidence does not establish a checklist that can simply be switched on in people. The 2021 review and a 2014 review of vertebrate limb regeneration discuss these mechanisms and open questions.

Did humans evolve to lose the ability to regrow limbs?
That is not established. Salamander limb regeneration is unusual among vertebrates, and its evolutionary history remains under investigation. The 2014 review discusses salamander-specific genes and local evolutionary changes as possible contributors, while noting that why salamanders are the only adult tetrapod vertebrates able to regenerate limbs remains controversial. It would overstate the evidence to say humans simply lost a complete adult limb-regrowth ability they once had.

Salamanders offer researchers a way to study how wounds, cell behavior, nerves and positional cues coordinate tissue regrowth. NIGMS describes work to identify pathways involved in detecting tissue loss and developing a blastema, while noting that scientists do not yet fully understand how to activate such processes in humans. A 2025 National Institute of Child Health and Human Development (NICHD) workshop summary describes limb regeneration in mammals as a research goal and covers nearer-term challenges such as wound healing and muscle regeneration in amputees.

This is basic and translational research, not evidence of a treatment that can make a person regrow an amputated limb. The scientific challenge is to understand and safely coordinate the many processes needed to rebuild a correctly patterned, functional appendage—not merely to make cells grow.
