Vitamin D supplements are not proven to prevent brain cancer or treat a brain tumor. Laboratory studies suggest biological mechanisms worth investigating, but they do not establish benefit in people. General cancer findings cannot be assumed to apply to brain tumors, and taking too much vitamin D can cause serious harm.
Can vitamin D prevent brain cancer?
Current evidence does not establish that vitamin D prevents brain cancer. The National Cancer Institute (NCI) describes experimental mechanisms through which vitamin D could affect cancer cells, but a proposed mechanism is not proof that a supplement changes cancer risk in people. NCI reports that most randomized trials have not found a reduction in overall or site-specific cancer incidence. Those broad results do not establish a brain-cancer-specific effect. NCI’s overview of vitamin D and cancer explains the distinction between experimental, observational, and clinical evidence.
A 2022 systematic review of observational studies examined vitamin exposure and brain-tumor risk. Its abstract says circulating vitamin D was among the measures reviewed, but the protective associations it reports name vitamin C, beta-carotene, folate, and high serum alpha-tocopherol—not vitamin D. The abstract does not provide a vitamin-D-specific effect estimate, so it cannot support a claim that vitamin D protects against brain tumors. Read the review’s PubMed abstract.

Vitamin D supplementation is not established as a brain-tumor treatment in the sources reviewed here. Experimental findings can suggest hypotheses for further study; they do not show that taking a supplement shrinks a tumor or improves outcomes in patients. The NCI’s central nervous system tumor treatment guidance describes clinical treatment information but does not identify vitamin D supplements as an established therapy. Do not replace or delay oncology care in favor of supplements.

Does low vitamin D cause brain cancer?
The evidence described here does not establish that low vitamin D causes brain cancer. Observational studies can identify associations, but they cannot by themselves show that vitamin D status caused an outcome. Randomized trials offer stronger evidence about whether supplementation changes outcomes, and NCI’s summary of those trials does not establish a brain-cancer prevention benefit. Vitamin D status and cancer risk are separate questions from whether correcting a documented deficiency is appropriate for general health.
Can vitamin D supplements be harmful?
Yes. Excessive vitamin D can cause hypercalcemia (too much calcium in the blood) and hypercalciuria (too much calcium in the urine). Severe toxicity can lead to renal failure, soft-tissue calcification, abnormal heart rhythms, or death. The NIH Office of Dietary Supplements (ODS) lists an adult tolerable upper intake level of 100 micrograms (4,000 IU) per day from all sources, a limit established by the Food and Nutrition Board in 2010. It is a population-level upper limit, not an individualized prescription or a cancer-treatment dose; a clinician may direct a different dose to treat a documented deficiency. See the NIH ODS vitamin D fact sheet.

What does a vitamin D blood test tell me?
The main indicator of vitamin D status is serum 25-hydroxyvitamin D, written as 25(OH)D. NIH ODS notes that measurements can vary by assay and that levels associated with deficiency or adequacy are not definitively established for every context. The fact sheet summarizes National Academies reference points for general health:

- 50 nmol/L (20 ng/mL) or more: considered sufficient for most people.
- Less than 30 nmol/L (12 ng/mL): associated with risk of vitamin D deficiency.
These are general health reference points, not brain-cancer prevention or treatment targets. The evidence reviewed here establishes no blood level that prevents or treats brain cancer. Ask a healthcare professional to interpret a result in light of your health and circumstances rather than trying to reach a cancer-specific target with supplements.
