In 2015, a study of photosystem II argued that the enzyme’s four-manganese catalyst follows a high-valent oxidation-state pattern as it advances through the S-state cycle. The authors found that their model could account for the full cycle, while the low-valent alternatives they tested could not provide a consistent account across all states. That was the paper’s conclusion—not proof that the debate had ended.
What question did the study address?
Photosystem II (PSII) is the biological catalyst that splits water during photosynthesis. Its oxygen-evolving complex contains a cluster of four manganese ions. As the catalyst accumulates the chemical changes needed to form oxygen, it passes through five states, conventionally labeled S0 through S4.

The dispute was how to assign oxidation states to those manganese ions in each state. An oxidation-state assignment is a way to describe how electrons are distributed among atoms; it is not a direct photograph of each ion’s charge. The competing schemes differ over which manganese ions are assigned as Mn(III) or Mn(IV), and therefore how the cluster changes as the cycle proceeds.

The report summarized the study by Vera Krewald and colleagues, published in Chemical Science on 9 January 2015: “Metal oxidation states in biological water splitting”.

How do the high- and low-valent assignments differ?
The clearest comparison in the paper’s account is the S2 state. The two schemes assign different numbers of Mn(III) and Mn(IV) ions:
| Scheme | S2 manganese assignment | What the assignment implies |
|---|---|---|
| High-valent | Mn(III, IV, IV, IV) | One Mn(III) and three Mn(IV) ions |
| Low-valent | Mn(III, III, III, IV) | Three Mn(III) and one Mn(IV) ion |
The labels are shorthand for the four ions in the cluster; they do not, by themselves, show the cluster’s full structure or explain every spectroscopic signal. The key test was whether an assignment could work coherently across the whole catalytic sequence, rather than fit one state in isolation.

What did Krewald and colleagues conclude?
The authors constructed a consistent structural and theoretical framework for the S-state cycle and tested candidate models against experimental constraints. Their high-valent model described a progression from S0, assigned Mn(III, III, III, IV), to S3, assigned Mn(IV, IV, IV, IV). In their calculations, the low-valent alternatives did not produce a consistent model of the complete cycle.

It supports the high-valent assignment within that analysis; it does not establish that every competing interpretation was disproved or that the field reached a lasting consensus. Chemistry World’s report recorded disagreement at the time.
What evidence did the study bring together?
The team compared its calculations with multiple kinds of experimental observation, including extended X-ray absorption fine structure (EXAFS), X-ray free-electron laser diffraction (XFEL-XRD), electron paramagnetic resonance (EPR), electron-nuclear double resonance (ENDOR), and manganese K pre-edge X-ray absorption near-edge structure (XANES). The paper also included new low-temperature 55Mn ENDOR data for S2.
Using different measurements matters because each constrains a different aspect of the system. Structural observations can restrict the arrangement of atoms, while spectroscopic signals provide information about electronic and magnetic properties. The study’s central move was to examine these constraints together in a shared framework, rather than treating an oxidation-state assignment as a conclusion from one measurement alone.

Why did the interpretation remain contested?
In the 4 February 2015 Chemistry World report, computational quantum chemist Rob Stranger argued that “the argument for the low-valent model is by no means dead.” He pointed to other interpretations of spectroscopic results, disagreements over crystal structures, and substrate-water exchange kinetics as reasons the low-valent interpretation remained viable in his view.
Marcel Swart, a computational chemist at the University of Girona, praised the systematic combination of theory and experiment, calling it “a complete and convincing story” for high-valent assignments along the cycle. He also identified unresolved work concerning the oxygen-forming step and the return to S0. These comments describe positions reported alongside the 2015 study; they should not be read as a summary of later developments.

Corresponding author Dimitrios A. Pantazis characterized the study’s conclusion as offering “a definitive answer.” That was his description of the paper’s claim. The distinction between a strong conclusion within a study and a settled scientific consensus is important, especially when other researchers identify evidence they believe still supports an alternative.

The cited 2015 report and paper establish what the authors concluded and why the assignment was disputed at publication. They do not establish whether subsequent work has settled the high- versus low-valent question. On this evidence, the accurate takeaway is historical and bounded: Krewald and colleagues found the high-valent scheme better able to account for the full S-state cycle in their models, while contemporaneous critics maintained that low-valent interpretations remained viable.
