A 2010 study reported that self-assembled peptide nanospheres could reach a Young’s modulus of up to 275 GPa, a measure of stiffness that is higher than the approximately 200 GPa steel comparison reported at the time. That does not mean the spheres were stronger than steel in every sense: the headline comparison concerns a nanoscale structure and, for the 275 GPa figure, a modeled particle geometry.

What the researchers found

Adler-Abramovich and colleagues reported the findings in “Self-Assembled Organic Nanostructures with Metallic-Like Stiffness,” published online on 28 September 2010 in Angewandte Chemie International Edition (volume 49, issue 51, pages 9939–9942). The study used atomic force microscopy (AFM) with a diamond-tip cantilever to examine nanoscale spheres assembled from an aromatic dipeptide. The publisher’s abstract reports a Young’s modulus of up to 275 GPa and describes the structures as potential building blocks for ultrarigid composite biomaterials. Read the paper record at Wiley.

The material was made from N-tert-butoxycarbonyl (Boc)-protected diphenylalanine. A contemporaneous Chemistry World report gives the spheres’ diameters as ranging from 30 nm to 2 μm. The researchers could not determine the dimensions of the internal cavities. Read the report.

What “stiffer than steel” means

Young’s modulus describes how resistant a material is to elastic deformation under load: broadly, how much it strains for a given applied stress. A higher modulus means greater stiffness. It is not a direct measure of tensile strength, hardness, toughness, or resistance to breaking.

The comparison reported by Chemistry World puts the peptide structure at 275 GPa, Kevlar at 130 GPa, and steel at approximately 200 GPa. Crucially, the 275 GPa value corresponds to a simulated case: a 1 μm particle with a 0.4 μm dipeptide shell. The report explains that the researchers used simulation to estimate the modulus because the basic measured stiffness varied with material thickness. These figures therefore do not establish that every sphere, or a bulk peptide material, would have the same modulus.

Material or case Young’s modulus reported How to interpret the figure
Peptide nanosphere, modeled geometry Up to 275 GPa Study authors’ reported value; the Chemistry World comparison associates it with a simulated 1 μm particle and 0.4 μm shell.
Kevlar 130 GPa Comparator value reported by Chemistry World in 2010.
Steel Approximately 200 GPa Comparator value reported by Chemistry World in 2010.

As a result, “stiffer than steel” is a shorthand for a particular stiffness comparison, not a universal ranking of material performance. The reported values do not show that peptide spheres have greater tensile strength, fracture resistance, durability, or practical performance in a finished object.

Why the result did not mean an immediate material breakthrough

The proposed uses were prospective. The researchers suggested that the nanospheres might reinforce composite materials and mentioned medical implants as a possible direction, but the 2010 report emphasized the substantial distance between discovery and practical use. It did not establish that the material was manufactured at useful scale or incorporated into a commercial composite or implant.

The mechanism behind the unusually high mechanical properties was also unresolved. The researchers suggested that molecular planarity and interactions between the π-electrons of aromatic side chains might contribute, but this remained a proposed explanation. Protein-assembly researcher Kenneth Woycechowsky, quoted by Chemistry World, raised pH-dependent structure as a possible constraint on applications.

What the experiment can—and cannot—support

  • Supported: the 2010 study reported AFM experiments on self-assembled aromatic dipeptide nanospheres and a Young’s modulus reaching 275 GPa.
  • Qualified: the comparison with steel and Kevlar is about stiffness, and the reported 275 GPa case relies on a modeled particle size and shell thickness.
  • Not established by the reported result: across-the-board superiority to steel, the performance of a bulk material, a confirmed molecular mechanism, or readiness for a medical or structural application.

In a contemporaneous interview with Chemistry World, study coauthor Itay Rousso said, “When we applied force to measure these particles, diamond probes were the only thing that actually made an indentation,” and cautioned, “There is a large gap between discovery and usage.” Those remarks are the coauthor’s comments as quoted in the report, not claims from the paper itself.
