Can nitrogen dioxide and ozone act synergistically? Some controlled laboratory and animal studies found effects from exposure to both pollutants that were greater than the effects expected from either alone. But the result varied with exposure conditions and the health outcome measured. The evidence does not establish a universal synergistic effect in people breathing ordinary outdoor air.
What does “synergy” mean for air pollutants?
A synergistic effect means a combined effect greater than the effect expected by adding the effects of the individual pollutants. That comparison is specific to the chosen outcome—such as a change in lung tissue or resistance to infection—and to the method used to define the expected additive effect. A mixture can look synergistic for one endpoint and not for another; “synergy” is not an unchanging property of two chemicals.
Other possible mixture outcomes include an additive effect, in which the combined response is consistent with the individual effects added together, and a less-than-additive effect, in which the mixture response is smaller than that expectation. Researchers also have to account for dose, exposure duration, and whether pollutants are inhaled together or in sequence.
How are NO2 and ozone related in the atmosphere?
Nitrogen dioxide (NO2) is one of the nitrogen oxides, or NOx. The U.S. Environmental Protection Agency says NO2 forms quickly from emissions from cars, trucks and buses, power plants, and off-road equipment. NOx also contributes to the atmospheric formation of ground-level ozone and fine particles. That relationship helps explain why the pollutants can occur in the same air, but it does not show that inhaling them together causes a greater-than-additive biological effect.

Ozone is a reactive gas and an air pollutant at ground level. EPA describes it as “a powerful oxidant that can irritate the airways.” NO2’s role in ozone-forming chemistry and the potential effects of breathing a mixture are separate questions: atmospheric chemistry alone cannot answer whether ozone makes NO2 more harmful inside the body.

What have mixture studies found?
The studies do not all ask the same question. Some exposed animals to both gases, one tested cells in vitro, and the human study exposed participants to each pollutant separately. The concentrations below belong to their respective experimental designs; they should not be compared across study types as if they represented equivalent real-world doses.

Does ozone make NO2 more harmful to people?
The animal and in-vitro results make that a plausible question for toxicology, but the evidence summarized here does not answer it as a general claim about people. The 2024 controlled human exposure abstract tested NO2 and ozone separately, not as a mixture. The county-level study examined mixtures that also included PM2.5, so its associations cannot show that ozone specifically amplified NO2’s effect.

- Not established: That combined exposure always produces synergy, or that people face a quantified excess risk from the combination under ordinary ambient conditions.
- Not established: A combined NO2–ozone health threshold or a human effect size attributable specifically to an interaction between these two gases.
What are the known health concerns?
Uncertainty about synergy does not mean either pollutant is harmless. EPA lists respiratory symptoms, lung-function decrements, and airway inflammation among the effects associated with ozone pollution. Its ozone health information identifies people with asthma, children, older adults, and people active outdoors among groups at greater risk from ozone exposure. EPA’s NO2 overview also identifies people with asthma, children, and older adults as groups generally at greater risk from NO2 health effects. Those points concern the pollutants’ known health effects; they do not quantify a special additional risk from breathing both together.
For current health information, consult EPA’s pages on health effects of ozone pollution and ozone effects in the general population. For day-to-day exposure decisions, check official local air-quality information and follow any health guidance issued for your area.

What EPA’s assessments do—and do not—say
EPA’s oxides of nitrogen assessment page identifies the 2016 Integrated Science Assessment as the last final assessment and lists a July 2026 external review draft. A draft is not a final assessment. EPA’s ozone assessment page identifies its 2020 assessment as the latest evaluation on that page. These assessment-status details are not a combined-exposure standard: they do not establish a regulatory threshold for NO2 and ozone acting together.
