There is no universally best label-free particle detector. Optical methods read light-based contrast, electrical methods measure impedance changes as particles pass a sensing region, and acoustic methods detect particle-related mechanical responses. The right choice depends on the particle, size range, sample matrix, concentration, and whether you need counting, sizing, imaging, sorting, or classification.
Use the comparison below to match the physical signal and workflow to your application. Treat throughput, resolution, and cost figures as method- and study-specific rather than universal specifications.
What “label-free” means in particle detection
A label-free assay detects a particle through an intrinsic physical property rather than an attached fluorescent, magnetic, enzymatic, or molecular tag. The measured property might be light scattering, absorption, phase change, electrical impedance, particle volume, acoustic impedance, or photoacoustic absorption.

Two systems both advertised as label-free can produce entirely different data and have different limits of detection, sample requirements, and failure modes. “Optical,” “electrical,” and “acoustic” are useful families, but a meaningful comparison must identify the specific technique inside each family.

How the three detection families work
Optical detection: contrast from light
Label-free optical systems illuminate a sample and measure naturally occurring optical contrast. Depending on the modality, contrast can arise from scattering, absorption, refractive-index differences, or phase-related changes. Imaging systems use that contrast to visualize cells or other particles; non-imaging systems can count events or measure signal intensity as particles flow through a detection zone.
Optical methods are non-destructive in many biomedical workflows and can provide spatial information that electrical pulse measurements do not. Imaging can reveal morphology, position, and sometimes internal structure. However, “optical” does not specify a single detection limit or event rate. A scattering detector, a quantitative phase system, and an absorption-based instrument have different sensitivity to particle composition, size, background, and illumination conditions.
- Typical information: presence, count, morphology, position, scattering or absorption-related properties, and image-derived classifications.
- Strength: direct visual or image-based evidence, with no tagging step.
- Constraint: optical background, transparency, refractive-index contrast, focus, and image-processing choices can determine whether a particle is visible.
Electrical detection: impedance and the Coulter principle
Impedance-based flow cytometry is a non-optical, label-free approach. In a Coulter-principle arrangement, particles suspended in an electrolyte pass through a narrow aperture. The displaced conductive liquid changes the electrical resistance and generates a voltage pulse. Pulse amplitude is proportional to the particle’s volume under the measurement conditions.

A practical impedance-cytometry workflow includes single-particle measurement, signal conditioning and processing, calibration, and subtype identification. The electrical waveform can contain more information than a simple count, but interpretation depends on aperture geometry, fluid conductivity, particle position, flow conditions, and instrument electronics. Electrical methods can also report properties related to particle electrical behavior, not just volume, when the system measures frequency-dependent impedance or multiple signal components.

A review of extracellular-vesicle characterization reported impedance-based flow-cytometry resolution above 300 nm for the method literature it examined. That is a context-specific review finding, not a specification for every impedance instrument or a universal lower size limit.
- Typical information: event count, volume-related size estimate, size distribution, concentration, and electrical-property features.
- Strength: high-speed single-particle measurements without optical labels.
- Constraint: conductivity, aperture condition, particle position, coincidence events, and calibration strongly affect the result.
Acoustic detection: mechanical response in the sample
Acoustic particle detection measures sound or mechanical waves associated with particles and their interaction with the surrounding medium. In a reported microfluidic flow-cytometry study, acoustic contrast was associated with acoustic-impedance mismatch between a particle and the surrounding fluid. The particle’s size, density, compressibility, and the medium’s properties all influence that response.
Photoacoustic detection is related but not identical. A short light pulse is absorbed by the particle or its contents; the resulting rapid thermoelastic expansion launches a mechanical wave that is detected acoustically. In other words, the excitation is optical, while the measured signal is acoustic. A system can combine acoustic and photoacoustic measurements, but a demonstration of simultaneous analysis does not establish that one configuration works for every particle or matrix.
- Typical information: acoustic response, contrast related to mechanical properties, and—when photoacoustic excitation is used—absorption-linked information.
- Strength: mechanical or absorption-sensitive contrast without a fluorescent label.
- Constraint: coupling, acoustic reflections, medium properties, transducer bandwidth, and particle position can alter the signal.

Choose by the question you need to answer
If you need images or morphology
Start with an optical imaging method. Images can preserve particle position and shape, which is valuable when classification depends on morphology rather than a single scalar signal. Define the optical modality before comparing performance: phase, scattering, and absorption measurements should not be treated as interchangeable.

If you need fast counting or a volume-related size estimate
Impedance flow cytometry is a logical candidate when the sample can be prepared in a conductive medium and particles can pass consistently through an aperture. The Coulter pulse gives a direct volume-related signal, while additional impedance features may support subtype discrimination. Verify the instrument’s calibration range and coincidence controls with particles that resemble your target.
If mechanical properties or acoustic contrast matter
Evaluate acoustic detection when density, compressibility, or acoustic-impedance mismatch is central to the analysis. Acoustic and photoacoustic signals can add information unavailable from a purely optical or electrical measurement, but the transducer, fluid path, and coupling design are part of the measurement—not interchangeable accessories.
If you need sorting or other particle handling
Separate the sensing question from the handling question. Label-free microfluidic platforms can combine detection with screening, sorting, or enrichment. Reviews group electronic sensors, optical and deformation techniques, hydrodynamic methods, and acoustic approaches as distinct options. A detector that measures particles well may still require a separate or integrated mechanism to route them into collection channels.
Throughput, integration, and cost evidence
A 2021 review comparing electrical impedance spectroscopy flow cytometry (EIS-FC), fluorescence-activated cell sorting (FACS), and imaging flow cytometry (IACS) reported the following review-era estimates. These are dated estimates from that review, not current quotations or guaranteed capabilities.

| Platform in the 2021 review | Estimated installation cost | Reported event rate | Character of the measurement |
|---|---|---|---|
| EIS flow cytometry | Below $100,000 | Tens of thousands of events per second | Electrical, label-free, with dimensionality and integration determined by the configuration |
| FACS | $50,000–$1 million | Tens of thousands of events per second | Fluorescence-based comparison platform |
| IACS | Above $1 million | Around 100 events per second | Imaging-based comparison platform |
Do not use those figures as a purchasing budget or as a universal ranking. Installation cost varies with lasers, detectors, fluidics, sorting hardware, automation, service contracts, and facility requirements. Event rate also depends on concentration, coincidence limits, sample preparation, and whether the stated rate is a theoretical, instrument, or application value.
A practical comparison workflow
- Define the target. Record whether the particles are cells, extracellular vesicles, synthetic particles, droplets, or another material. Specify the expected size range, concentration, and heterogeneity.
- Describe the matrix. Note conductivity, viscosity, optical background, suspended debris, bubbles, and components that may absorb or scatter light or alter acoustic coupling.
- State the required output. Decide whether you need a yes/no detection, concentration, size distribution, morphology, electrical properties, acoustic properties, classification, or a sorted fraction.
- Choose the physical signal. Select optical contrast for image or light-property information, impedance for volume-related and electrical measurements, or acoustic/photoacoustic contrast for mechanical or absorption-linked information.
- Check the flow path. Confirm aperture dimensions for impedance systems, optical path and focus for imaging, or transducer placement and coupling for acoustic systems. Ensure the geometry matches the particle size and expected concentration.
- Plan calibration and controls. Use standards or reference particles appropriate to the target, run blank matrix controls, and test for coincidence, background, drift, and carryover. Calibration should cover the operating range rather than a single convenient point.
- Validate with the real sample. A method that performs well in a clean buffer may behave differently in serum, lysate, process fluid, or another complex matrix. Compare recovery, false events, repeatability, and classification stability in the intended sample.
Common failure modes and fixes
“Label-free” is treated as a performance guarantee
It is not. Label-free only identifies how contrast is obtained. Fix the comparison by naming the exact modality and reporting the particle, matrix, calibration, and analysis conditions alongside any detection limit or resolution claim.
Optical background overwhelms small particles
Scattering, absorption, debris, and out-of-focus material can create events or hide weak contrast. Improve sample cleanup, optical alignment, focus control, and blank subtraction, then verify that image-processing thresholds do not remove genuine low-contrast particles.
Impedance pulses vary unexpectedly
Check electrolyte conductivity, aperture condition, bubbles, particle coincidence, flow stability, and calibration. A pulse interpreted as a size change may instead reflect an off-axis trajectory or a transient fluidic disturbance.

Acoustic signals drift or ring
Inspect coupling, trapped bubbles, reflections, transducer bandwidth, temperature, and fluid composition. Acoustic measurements are sensitive to the complete mechanical path, so changing tubing or channel geometry can change the baseline and response.
Numbers from different studies appear contradictory
They may measure different particles, matrices, definitions of resolution, or event-rate limits. Keep each number attached to its source context and compare like with like rather than selecting the largest throughput or smallest reported size.
What the evidence supports—and what it does not
The available comparisons establish distinct physical principles and show that label-free microfluidic detection can support screening, analysis, and, in some platforms, sorting. They do not establish a single cross-modal detection threshold, a universal ranking, or a current vendor specification. The reported extracellular-vesicle resolution above 300 nm applies to the reviewed impedance literature, and the installation-cost and event-rate figures come from a 2021 comparison.
