Water droplets can model selected features of cell membranes when researchers coat them with lipids and bring them together. The lipid layers form a thin bilayer where the droplets meet—a droplet interface bilayer, or DIB. Researchers can use that interface to study transport and electrical behavior, but a DIB is a simplified experimental model, not a recreated living cell.
What is a droplet interface bilayer?
A DIB forms when two aqueous droplets, each coated with lipids, touch. The lipid layers rearrange at their shared boundary to create a bilayer: two layers of lipid molecules separating the droplets. The droplets remain distinct compartments, so one can act as a donor and the other as an acceptor in an experiment.
That arrangement lets researchers examine selected membrane behaviors, including whether molecules cross the bilayer and how much transport occurs. DIBs can also support electrophysiological measurements and can be connected into networks. A 2022 perspective describes DIBs as models that can reproduce some features of cell membranes more effectively than traditional systems such as liposomes and black lipid membranes, while cautioning that no model captures everything. The authors write that “the perfectly biomimetic, yet bespoke, model membrane has yet to be built.” Read the 2022 Nature Chemistry perspective on DIB capabilities and limitations.

What can researchers learn from the droplets?
The key advantage is experimental separation: two droplets provide distinct aqueous compartments, and the bilayer between them is a defined boundary to investigate. Depending on the setup, researchers can measure molecular transport, record electrical behavior, or connect compartments in a network. These are targeted measurements of membrane-related processes; they do not establish that the droplets perform all the functions of a cell.

Why do temperature and lipid composition matter?
Bilayer formation depends on the experimental recipe and conditions. In a 2021 microfluidic study using naturally derived phospholipids, Korner and Elvira examined phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylserine (PS), and phosphatidylinositol (PI)—lipid classes the paper identifies as abundant in mammalian cell membranes. Under the conditions tested, these lipids formed DIBs only above their phase transition temperatures. The study also reports that formation usually occurred above the highest transition temperature of a single lipid in a bespoke formulation.
Those findings describe the study’s microfluidic platform and tested lipid formulations, not a universal temperature rule for every DIB. How DIBs differ from other artificial-cell models
“Artificial cell” describes several experimental architectures, not one standard design. Their compartments and demonstrated functions differ, so the useful choice depends on the question being asked rather than on a universal ranking.

Hydrogel-encapsulated communicating droplets
A 2017 study described aqueous droplets stabilized in an oil/lipid mixture and encapsulated in hydrogel. Protein nanopores crossing the lipid bilayer enabled electrical and chemical communication, and the arrangement allowed adjoining bilayers. This differs from a simple pair of droplets meeting in a DIB experiment. Read the 2017 Scientific Reports study.

All-aqueous droplet-in-droplet compartments
A 2025 Nature Communications paper reported an all-aqueous structure combining coacervate and aqueous two-phase system components. The authors reported separating transcription and translation spatially between compartments. This is a related artificial-cell strategy, not another name for a DIB. What the droplet models do not show
These systems help investigate selected physical and chemical functions under controlled conditions. Their ability to model transport, electrical signals, communication, or compartmentalized reactions is not evidence that a complete living cell has been recreated. Each architecture simplifies different parts of cell organization, and the 2022 review emphasizes that a fully biomimetic model membrane remains an unmet goal.

A brief historical example: droplets producing electrical output
A 2009 account from the National Institute of Standards and Technology described a simplified model cell made from a salt-containing water droplet enclosed by lipid. When two such droplets contacted, their lipid arrangement formed a double bilayer. A difference in salt concentration could drive electrical output through a circuit with electrodes. This was a demonstration of a model system, not evidence of a practical battery product. Read NIST’s 2009 account.
