Detecting airborne mould means collecting air with a method chosen for the question, then analyzing the sample by microscopy, culture, PCR, or another suitable assay. Collection and analysis are separate steps: culture detects organisms that grow under selected conditions, microscopy can count fungal structures, and PCR detects genetic material from its chosen targets. A result describes the sampled place and time; it is not a complete measure of exposure or a health-risk verdict.

Is sampling or testing for mould necessary?

Usually not as a first step. If you can see mould, the U.S. Environmental Protection Agency (EPA) says sampling is generally unnecessary: address the moisture source and mould growth. CDC/NIOSH also does not recommend routine mould air sampling as part of building air-quality evaluations. A musty odour or visible growth can warrant investigation and remediation without an air count.

Sampling is most useful when a defined question could change a decision—for example, investigating a suspected hidden problem or evaluating a specific environmental condition. EPA recommends using professionals experienced in designing sampling protocols, selecting methods, and interpreting results. If a suitable plan and enough samples cannot be obtained to answer the question, EPA cautions that it may be better not to sample than to rely on misleading results.

What an airborne mould sample can—and cannot—tell you

An air sample is a snapshot. EPA says it reports what was in the air when sampling took place; CDC/NIOSH warns that a negative short-term result may not represent actual exposure. Airborne particles vary with location, time, activity, and environmental conditions, so one result cannot describe every room or every period.

  • Furniture made of wood is a high-risk area for mold, especially black mold. It contains 12 separate tests and is of high value. Take it with you to check your cabinets, wardrobes, and beds!

There is no universal health threshold for interpreting the result. EPA says no federal limits have been set for mould or mould spores in the air. CDC/NIOSH says short-term spore or culture counts cannot be interpreted as health risks. CDC healthcare guidance likewise notes that standards linking fungal spore levels to infection rates have not been established, and that protocols and culture methods vary.

As CDC/NIOSH puts it, “No indoor space is completely free from mold spores” (Mold, Testing, and Remediation, February 25, 2025). Detecting spores therefore does not, by itself, show that a building is unsafe; not detecting them in one short sample does not prove that exposure never occurs.

How collection methods differ

The sampler collects airborne particles; the laboratory assay determines what can be detected from what was collected. No single sampler-and-assay combination captures and enumerates every airborne organism. Choose the combination according to whether the goal is to find viable, culturable organisms, count fungal structures, or detect selected genetic material.

Collection or analysis method What it can contribute Key limitation
Solid impaction Deposits particles onto a solid surface, such as coated material or agar, for microscopy or culture. Sampler design, airflow, collection efficiency, and compatibility with the assay affect results.
Liquid impingement Collects particles in liquid; CDC describes liquid impingers as practical for microbial air sampling. Recovery depends on the sampler and organism conditions, so select it for the intended analysis.
Filtration or dry collection Traps particles on a filter and can support nonculture analysis such as PCR. May not preserve viability; the collection method must suit the assay.
Culture Shows organisms able to grow on the selected media and can provide viable/culturable counts. Can miss organisms that do not grow under those conditions, and results require time to develop.
Microscopy Can resolve and count fungal structures or spores collected on a suitable medium. Identification detail and interpretation depend on the method and the analyst’s expertise.
PCR Detects targeted DNA, including from organisms difficult or impossible to culture. DNA detection does not establish viability; results are limited to the targets selected.
Settle plates Provide simple, qualitative gravity-based collection. CDC says they should not be used to quantify airborne fungal spores: they miss suspended individual spores and do not measure sampled air volume.

These distinctions matter when comparing results. A culture count is not a count of all fungal material in the air, and a PCR detection is not proof that the detected organism was alive. CDC’s environmental sampling guidance explains that sampler and assay choice, particle size, and collection efficiency affect what is recovered.

How to plan a meaningful sampling investigation

Start with the decision the sample is meant to inform, not with a device or a desired number. CDC guidance recommends accounting for the organism and whether viability matters, compatibility between collection and analysis, particle-size sensitivity, expected concentration, air volume, sampling duration, background contamination, ambient conditions, and collection efficiency.

  1. Define the question. Specify the location and condition of concern, and decide whether you need evidence of viable growth, a count of fungal structures, or detection of particular genetic targets.
  2. Choose the assay and laboratory. Confirm that the laboratory can perform the intended analysis and that the planned sampler and collection medium are compatible with it.
  3. Select the collection approach. Consider particle-size performance, expected concentration, collection efficiency, and the air volume and duration needed to answer the question.
  4. Plan for conditions and handling. Account for ambient conditions and background contamination. Arrange adequate supplies and refrigeration if prompt analysis will not be possible.
  5. Interpret within the plan’s limits. Record the sampled place and time, method, volume, duration, conditions, and laboratory method. Use the result only to answer the question that the plan was designed to address.

CDC’s sampling guidance and EPA’s Mold Course Chapter 3 explain why inadequate sampling can produce results that do not answer the intended question.

How to interpret a result without overclaiming

  • A positive culture means organisms grew under the laboratory’s selected conditions; it does not account for organisms that those conditions did not grow.
  • A positive PCR result means targeted genetic material was detected; it does not establish that the source organism was viable.
  • A microscopy count depends on the collection medium, identification detail, and analyst expertise.
  • A negative short-term air sample does not establish that exposure never occurs or that a suspected hidden source is absent.
  • A raw count cannot be converted into a general safe/unsafe judgement using a federal airborne-mould limit, because EPA says no such federal limit has been set.

For these reasons, air sampling should be interpreted alongside the actual building conditions and the purpose of the investigation, not as a stand-alone health test. CDC/NIOSH’s mould guidance covers routine testing and the limits of short-term results; EPA’s sampling guidance addresses when testing may be useful and the value of professional protocol design.
