The MasSpec Pen is a research-stage probe designed to help identify cancerous tissue during an operation by collecting molecules from a tissue surface and sending them to a mass spectrometer. The instrument produces a chemical profile that may help distinguish cancer from normal tissue; the pen itself is not a visual scanner or a standalone diagnostic device.
How does the MasSpec Pen work?
The device described by Chemistry World in 2021 combines a handheld sampling probe with flexible tubing and a mass spectrometer. Its 3D-printed polydimethylsiloxane tip has three ports: one connects to vacuum, one lets compressed gas operate the system, and one delivers a small water droplet to the tissue.
- A small amount of water contacts the tissue surface and absorbs molecules from it.
- After about three seconds, vacuum draws the water and collected molecules through the tubing.
- A mass spectrometer analyzes the sample. Chemistry World described the complete process as taking around ten seconds.
The resulting mass spectrum contains signals associated with molecules such as metabolites, lipids, proteins, and fatty acids. The probe therefore gathers a sample; the mass spectrometer performs the analysis. Chemistry World’s 2021 feature describes the device and workflow.
Why might a chemical profile help during surgery?
Tissue can look similar to the naked eye even when its cellular makeup differs. The MasSpec Pen’s premise is that cancerous and normal tissue can have distinguishable molecular profiles. A rapid chemical readout could give a surgical team additional information while an operation is underway, rather than relying on appearance alone.

That is a proposed aid to intraoperative assessment, not evidence that the device is better than pathology or other established methods. The 2021 feature does not establish such a comparison.
What results were reported?
Chemistry World reported in 2021 that seven surgeons had trialled the device in more than 100 human surgeries at a Houston hospital. The feature also reported 715 in vivo and ex vivo analyses across thyroid, parathyroid, lymph-node, breast, pancreatic, and bile-duct tissues. It reported over 90% accuracy for distinguishing normal and cancerous pancreas, ovarian, lung, thyroid, and breast tissues.

Is the MasSpec Pen approved or available for routine use?
The cited feature said clinical tests had finished in 2020 and described the team’s plans for further work as of 2021. It does not establish the device’s present-day regulatory status, commercial availability, or adoption in routine surgery. The MasSpec Pen should therefore be understood here as a research-stage approach, not as an approved or standard-of-care product.

What was the goal behind the device?
Livia Schiavinato Eberlin, a professor at the University of Texas at Austin, described the aim as making mass spectrometry easier for surgeons to use: “I wanted surgeons to take advantage of the amazing analytical technique that is mass spectrometry through a user-friendly interface or device that did not require extensive expertise for use,” she told Chemistry World in 2021.
