In a 2006 experiment, short synthetic collagen-like fragments joined end to end through overhanging “sticky” peptide segments, assembling into fibrils. Some measured fibrils exceeded 400 nm—longer than the approximately 300 nm natural collagen length used for comparison in the study. The work concerned peptide materials, not collagen supplements or a treatment.
How did the sticky ends make the fragments join?
Frank W. Kotch and Ronald T. Raines designed short collagen-like peptide fragments with three peptide strands connected by disulfide bonds. The bonds arranged the strands in a staggered configuration, leaving peptide segments protruding at the ends. Those overhangs acted as “sticky ends”: they could associate with overhangs on other fragments, promoting intermolecular triple-helix formation and extending the assembly.
The design addressed a length limitation of earlier synthetic collagen triple helices, which the paper describes as less than 10 nm. The fragments were not simply longer individual helices; the strategy used association between fragments to build larger fibrils.
What did the study measure?
The researchers used several complementary methods to examine the fragments and their assemblies: circular dichroism spectroscopy, dynamic light scattering, analytical ultracentrifugation, atomic force microscopy and transmission electron microscopy. Together, these measurements supported the interpretation that the sticky-ended fragments self-assembled into fibrils with a collagen-like appearance.

These are the paper’s experimental comparison figures, not product specifications or a claim that every fibril had the same length. The study reported fibrils that resembled natural collagen; resemblance does not mean the synthetic material was identical to collagen made by living tissue.
What could the approach be useful for?
The paper proposed that this self-assembly strategy might provide synthetic collagen-mimetic materials for varied applications. Kotch and Raines wrote, “We anticipate that our self-assembly strategy can provide synthetic collagen-mimetic materials for a variety of applications.”
A contemporary Chemistry World report discussed possibilities such as wound-healing materials and metal-coated nanowires for nanotechnology. These were prospective ideas, not outcomes demonstrated by the fibril experiment. The study did not establish a therapy, clinical benefit, approved product or commercial use.
