In a 2010 demonstration, researchers combined scanning transmission electron microscopy (STEM) with electron energy-loss spectroscopy (EELS) to measure the electronic and bonding structure of individual carbon atoms at a graphene edge. Their spectra distinguished atoms bonded to one, two or three neighboring atoms—not simply by making a sharper image, but by analyzing how each atom interacted with the electron beam.

What the technique measured

STEM focuses an electron probe onto a small region of a specimen. In EELS, scientists measure how much energy electrons lose after interacting with that specimen. The energy-loss near-edge fine structure—the detailed shape of a spectrum near an element’s characteristic energy-loss edge—can reveal information about an atom’s electronic state and bonding.

In their paper, “Atom-by-atom spectroscopy at graphene edge,” Kazu Suenaga and Masanori Koshino of Japan’s National Institute of Advanced Industrial Science and Technology (AIST) positioned the probe at individual carbon atoms along a graphene boundary. They used the resulting fine structure to distinguish carbon atoms with single, double and triple coordination. Here, coordination describes how many neighboring atoms are bonded to the carbon atom.

This is different from elemental identification alone. Identifying an element tells scientists what kind of atom is present; the reported spectra also provided information about the bonding environment of a particular carbon atom in its local context.

How the researchers made an atom-specific measurement

  1. Focus the electron probe: The team directed a tightly focused beam at a chosen site along the graphene edge.
  2. Measure energy loss: They recorded how electrons lost energy as they passed through and interacted with the specimen.
  3. Read the fine structure: They analyzed the near-edge spectral features to infer the electronic and bonding structure at that site.

A contemporary account in Chemistry World reported a probe diameter of 0.1 nm and an accelerating voltage of 60 kV for this experiment. These are the reported settings for the demonstration, not universal requirements for EELS or guarantees that a sample will avoid damage. The low-voltage setup was intended to provide a suitably short electron wavelength for high-resolution spectroscopy while reducing the risk of specimen damage.

Why the result mattered—and what made it difficult

Atomic-resolution images can show where atoms are, but a site-specific spectrum can add evidence about their electronic and bonding structure. Suenaga described the change from measurements averaged over much larger areas to measurements of a single atom: “In the past, if you were using some other technique, you could do spectroscopy for a local area of 1,000 atoms or a million atoms, something like that. Now, with a very tiny probe, we can measure from single atoms.” The quotation and experimental parameters were reported by Chemistry World in 2010.

The measurement was challenging because fine-structure signals from individual light atoms are extremely weak, while the electron beam can damage the specimen. Those constraints make it important to distinguish the demonstrated capability from a claim that any atom can be analyzed easily or without altering the sample. The original Nature paper describes these limitations in its account of the work: “Atom-by-atom spectroscopy at graphene edge”.

What the demonstration does—and does not—establish

The reported result was specific: site-resolved EELS of carbon atoms at a graphene boundary, with spectra distinguishing single-, double- and triple-coordinated carbon. It did not establish a general-purpose method for reading every property of every atom, nor did it demonstrate routine measurements across all materials or specimens.

The authors proposed that the approach could help investigate local electronic structure in nanodevices and individual molecules. Those were prospective applications; the demonstration itself concerned carbon atoms at a graphene edge. The Nature paper is available at Nature.
