Researchers distinguish quantum excitations in individual molecules by comparing how each molecule absorbs and emits light, responds to vibrational probes, and changes over time. A peak alone rarely settles the assignment: transition frequency, linewidth, polarization, decay, vibrational fingerprints, and sample geometry provide complementary evidence. The right combination depends on the molecule and the question.

Why measure molecules one at a time?

An ensemble spectrum combines signals from many molecules. That average can obscure differences between molecules and short-lived changes in an individual one. Single-molecule measurements retain that variation, allowing researchers to examine how transition energies, linewidths, or dynamics differ from molecule to molecule.

This matters because an observed feature may reflect not just the molecule’s state, but also its local environment. Comparing individual measurements can help separate a recurring molecular signature from variation across the sample.

What counts as a different excitation?

An electronic excitation changes the molecule’s electronic state. A vibrational excitation involves motion of its nuclei in a particular molecular mode. A vibronic transition involves electronic and vibrational structure together. An exciton is an electronic excitation whose behavior can involve interactions or energy sharing among molecules in a multichromophoric system.

These categories are not always identified by a unique spectral label. Researchers infer them from multiple observables and from how the signal responds to a particular measurement. A vibrational progression accompanying an optical transition, for example, provides different evidence from a change in signal over femtoseconds.

How optical excitation and emission spectra differ

Excitation spectra

An excitation spectrum records the detected outcome while the excitation wavelength is scanned. It can reveal which transitions prepare a detectable excited state, including structure associated with excited-state vibrations and, in some cases, short-lived intermediate states.

Emission spectra

An emission spectrum records photons released as an excited molecule decays. Its vibrational progression can show how the emitting state connects to vibrational levels of the final state. It therefore answers a different question from an excitation scan: what emits during decay, rather than which prepared transitions lead to the measured outcome.

Researchers can also compare transition frequency, linewidth, excited-state lifetime, polarization, and single-photon statistics. Taken together, these measurements can help distinguish transitions or states and reveal changes associated with the molecule’s surroundings. No one of these quantities should be treated as a universal identifier on its own.

How vibrational measurements identify molecular modes

Vibrational methods probe mode-specific responses, giving researchers a fingerprint that can help distinguish molecular motions and local interactions. Raman spectroscopy is one way to measure vibrational features. Resonance with a vibronic transition can selectively enhance particular Raman signals.

For example, a 2024 preprint reports scanning-tunneling-microscope (STM) resonance Raman measurements on individual deprotonated phthalocyanine molecules. In that configuration, tuning the laser to vibronic transitions between the electronic ground state and vibrational levels of an excited state enhanced selected Raman signals. This is a specific reported arrangement, not a general recipe for every molecule or sample.

Single-molecule vibrational spectroscopy has had to contend with weak signals. A 2026 perspective describes progress in far-field vibrational spectroscopy and imaging under ambient conditions, and discusses the chemically interpretable information these methods can provide. The particular technique still depends on the molecule and experimental geometry.

What ultrafast measurements add—and what they cannot settle alone

Femtosecond spectroscopy follows signals as they evolve, revealing temporal behavior that a steady-state spectrum cannot show. At the single-molecule level, such measurements have been used to investigate electronic and vibrational coherence, wavepacket interference, and quantum control.

An oscillating time-dependent signal is an observation; its explanation is an assignment. Some oscillations have been attributed either to excitonic coherence or to vibrational wavepacket motion. Time resolution adds valuable evidence, but does not automatically resolve that ambiguity. A careful interpretation distinguishes the measured pattern from the proposed mechanism and explains what supports the assignment.

When STM is useful

STM-based methods apply to molecules adsorbed in a surface junction. Tunneling electrons can couple to molecular electronic and vibrational states; with optical illumination and photon detection, a localized surface plasmon can also excite molecular states. These methods can provide local access to a surface-bound molecule and, in suitable setups, support mode-selective measurements.

The junction and substrate are part of the measurement geometry. An approach designed for an adsorbed molecule should not be assumed to transfer directly to a molecule in solution or in a crystal.

How researchers build a defensible assignment

  1. Choose the observable for the question. Use optical excitation or emission to study optical transitions and decay, vibrational methods to examine molecular modes, ultrafast measurements to follow dynamics, or STM-based methods for a suitable surface-bound molecule.
  2. Compare more than peak position. Examine linewidth, lifetime, polarization, vibrational structure, and photon statistics where the method provides them.
  3. Keep the sample geometry in view. Record whether the molecule is measured optically in a far-field setup or adsorbed in an STM junction; conclusions depend on the environment and configuration.
  4. Separate signal from interpretation. Report what was measured and what mechanism is proposed, especially when a time-dependent feature has more than one plausible explanation.

There is no established universal ranking of these techniques by sensitivity, resolution, cost, or suitability across all molecules. The appropriate approach depends on the target state, molecular environment, sample geometry, and timescale of interest.
