Researchers have designed a peptide that self-assembles into a nanopore and can detect DNA and a single polypeptide chain. Other protein designs sense targets through light, or modify an existing pore rather than create one entirely from scratch. These are distinct laboratory approaches—not parts of one finished biosensor—and the cited studies do not establish a clinical test or consumer product.
What does it mean to design a nanopore or biosensor from scratch?
In de novo protein design, researchers specify or redesign a protein’s structure to give it a desired function. For a nanopore sensor, the goal may be a channel through a membrane that lets researchers detect molecules by measuring changes in electrical current. A designed protein biosensor can instead change shape when it binds a target and produce a different signal, such as light.
The distinction matters: a designed pore, a designed protein switch, and a sensor built by modifying a natural pore are related engineering strategies, but they are not the same device. In particular, a luminescent protein switch is a biosensor, but it is not a nanopore.

Shimizu and colleagues reported SV28, a chemically synthesized β-hairpin peptide designed to assemble into pores in lipid bilayers. In this experimental membrane system, the pore provides a channel through which molecules can affect the electrical signal. The study reported SV28 pore diameters spanning 1.7 to 6.3 nm and demonstrated DNA detection. The study, published online on 22 November 2021 in Nature Nanotechnology and in the journal’s 2022 volume, is a proof of design and sensing capability, not evidence of a packaged diagnostic.

Why redesign the pore as SVG28?
The reported SV28 pore population varied in size. The researchers introduced a glycine-kink mutation to create SVG28, which they reported formed a monodisperse 1.7 nm pore. In a distinct demonstration from the DNA detection with SV28, SVG28 detected a single polypeptide chain. The study therefore illustrates two design goals: controlling pore dimensions and detecting a molecule with a designed channel.

What can designed protein switches detect?
Quijano-Rubio and colleagues reported modular, tunable protein biosensors that use target binding to shift a designed switch from a closed, dark state to an open, luminescent state. Their examples included proteins, toxins, and antibodies, among them HER2, cardiac troponin I, and SARS-CoV-2 spike. The 2021 Nature study demonstrates a route to designing protein sensors, but its light-based readout should not be confused with ionic-current measurement through a nanopore.

Rather than constructing an entire channel from newly designed components, Schnaider and colleagues integrated designed subunits into the natural CsgG pore. Their 2026 study reported a complex containing 18 subunits with a mass of 315 kDa. The authors reported altered current–voltage behavior relative to the native pore, including rectification, and used cryo-electron microscopy to confirm the intended lumen architecture. The study is evidence for engineering a natural pore scaffold with designed components—not for a wholly de novo pore.

How can a pore be made target-specific without redesigning the pore?
Another strategy attaches a programmable antibody-mimetic binder to a monomeric nanopore. The binder supplies the recognition function, and its interface can be changed to adapt the sensor to different protein targets. This separates the job of recognizing a target from the job of providing a pore. A 2023 Nature Communications study describes this modular approach; it should be classified as a binder-functionalized pore rather than a pore designed entirely from scratch.

How should readers interpret the reported results?
- They are research demonstrations. The cited studies report designed structures and sensing behavior in experimental systems. The evidence here does not establish a validated clinical test, routine patient use, or a consumer biosensor.
- Different readouts answer different questions. Ionic current is relevant to pore-based sensing; luminescence is the signal in the protein-switch platform. A result from one format does not establish performance in the other.
- Architecture affects what “designed from scratch” means. SV28 is a designed peptide pore; a luminescent switch is a designed sensor but not a pore; the CsgG system combines designed components with a natural scaffold; and the binder-functionalized sensor retains a nanopore and adds a recognition module.
- One adjacent 2026 result is not a de novo pore result. Huang and colleagues reported 98.7% overall accuracy across their analyte-identification task using an engineered MspA pore with a chemical adaptor. MspA is an existing natural pore, so the figure belongs to that specific task and setup, not to the designed pores or sensors above. The Nature Biotechnology study is useful context for nanopore engineering, not a like-for-like benchmark.
