Short, positively charged peptides can help RNA bind to model membranes in laboratory experiments, offering one possible way the ingredients of primitive cell-like compartments could have stayed together. A 2015 study found this effect with both phospholipid and fatty-acid vesicles, but it did not show that the process happened on early Earth.

What does “peptide glue” mean?

“Glue” is a metaphor for a two-part interaction, not a substance that cements a membrane. In the proposed mechanism, a peptide’s hydrophobic portion associates with the membrane, while its positively charged, or cationic, portion attracts negatively charged RNA. That electrostatic attraction can concentrate RNA at the membrane surface.

First author Neha Kamat described the idea in Chemistry World’s 2015 report: “The peptides essentially act as a kind of glue to bind membranes and then attract and hold the RNA at the membrane surface.”

What did the 2015 experiment find?

Kamat, Tobé, Hill, and Szostak tested whether short, basic amphipathic peptides could promote RNA binding to vesicle membranes. Their paper reports RNA localization in two model membrane types: zwitterionic phospholipid membranes and anionic fatty-acid membranes. The authors summarize the result as showing that “electrostatic interactions provided by short, basic, amphipathic peptides can be harnessed to drive RNA binding” to both types.

The shortest reported peptides

The primary paper reports that amphipathic peptides as short as three amino acids could drive RNA localization to model membranes. This is the precise minimum reported in the study; broader descriptions of very short peptides in contemporary news coverage should not be mistaken for the paper’s stated minimum.

RNA inside vesicles

The study also reports that peptides could cross vesicle bilayers and localize RNA that had been encapsulated inside. That finding extends the proposed mechanism beyond RNA already outside a vesicle, while remaining a result from model membrane experiments.

Why compare phospholipid and fatty-acid membranes?

The experiments used two membrane compositions with different charge characteristics. Zwitterionic phospholipids have both positively and negatively charged regions and are overall electrically neutral; the fatty-acid membranes in the study were anionic. RNA binding was reported in both model systems, suggesting the peptide-mediated mechanism was not limited to one of these membrane classes.

The comparison does not establish that either model reproduces every membrane that could have existed on early Earth. The Chemistry World report describes measurements of vesicle surface charge, along with selective fluorescence resonance energy transfer and microscopy to examine RNA–membrane association.

How does this fit origin-of-life ideas?

RNA matters in origin-of-life models because it can carry genetic information and catalyse chemical reactions. Membranes can create compartments and bring molecules together. A mechanism that associates RNA with a membrane could therefore help explain how genetic material and a cell-like boundary might have become co-localized.

The experiment supports that possibility, not a historical reconstruction. It did not create a living cell, demonstrate that these peptides existed in the required conditions on the prebiotic Earth, or prove that peptide-mediated RNA binding was how the first cells formed. The authors describe the mechanism as one that “could have been important” for primitive cellular evolution.
