DNA and RNA released outside cells can help activate blood-clotting reactions in laboratory experiments, particularly through the contact or intrinsic pathway. DNA-rich neutrophil extracellular traps (NETs) may also provide a physical framework that gathers platelets and fibrin. These are mechanistic findings, not evidence that nucleic acids are a practical treatment for bleeding: much of the evidence comes from plasma assays and animal models, and clotting in disease can be harmful.
How can nucleic acids help blood clot?
Coagulation is a chain of reactions that ultimately generates thrombin, an enzyme that helps convert fibrinogen into fibrin. Extracellular DNA or RNA can interact with proteins in the contact (intrinsic) pathway, including factor XII and factor XI, and promote their activation in experimental systems.
In a human-plasma study, adding DNA or RNA increased thrombin generation through the intrinsic pathway and shortened clotting time. Histidine-rich glycoprotein (HRG), a plasma protein that binds nucleic acids, reduced their effects at several points in the experiments. These results describe behavior in plasma assays; they do not establish the size or clinical effect of this mechanism in an intact person. Vu et al. (2016)
How NETs can support clot formation
Neutrophils can release neutrophil extracellular traps, or NETs: webs made of DNA, histones, and antimicrobial proteins. NETs are part of the innate immune response to infections, as the authors of the 2010 paper put it. In an experimental model, NETs promoted platelet adhesion, activation and aggregation, recruited red blood cells, and supported fibrin deposition. DNase or heparin dismantled the scaffold in that model. Fuchs et al. (2010)

This finding concerns the combined NET structure, not DNA acting alone. It also illustrates why a clot can be more than a cascade of soluble proteins: a physical scaffold can help bring clot components together. The experiment does not show that NETs should be used to stop bleeding in people.
Why results depend on the experiment
“Nucleic acids promote clotting” is not a universal rule. Outcomes can vary with DNA versus RNA, chain length and structure, concentration, preparation, and the method used to measure clotting. A comparative study warned that purification itself can create misleading results: silica particles shed by some silica-based extraction columns can promote clotting, while polyphosphate may co-purify with cellular DNA or RNA. Under that study’s experimental conditions, long-chain polyphosphate was much more potent than the tested cell-derived nucleic acids. Smith, Gajsiewicz, and Morrissey (2018)

Accordingly, a nucleic-acid preparation that appears procoagulant does not necessarily show that its DNA or RNA caused the effect. Comparisons need contamination controls and clear descriptions of sample preparation and assay conditions. A review also noted substantial variation in methods for measuring circulating nucleic acids and a lack of standardized procedures, which limits comparisons between studies and interpretation as clinical biomarkers. What animal studies add—and what they do not
An experimental study of extracellular RNA reported coagulation activity in vitro and found that RNase pretreatment delayed occlusive thrombosis in a mouse arterial-thrombosis model. The authors also reported a 15- to 20-fold elevation of serine-protease activity in their nucleic-acid-supplemented plasma experiments. That figure belongs to the paper’s specific assay; it is not a general estimate of what happens in people. Kannemeier et al. (2007)

Together, plasma and animal-model findings support a possible mechanistic role for extracellular nucleic acids in the studied settings. They do not establish an RNA-based treatment, or show that increasing nucleic acids would safely improve normal hemostasis—the body’s control of bleeding. Thrombosis, by contrast, is pathological clot formation and can obstruct blood flow.

The evidence described here does not establish an available clinical nucleic-acid treatment for bleeding. These findings are not a reason to apply DNA, RNA, laboratory reagents, or experimental materials to a wound. For an actual injury, use appropriate first aid and seek medical care when needed; the experimental clotting mechanisms discussed here are not home remedies.

How to interpret claims about nucleic acids and clotting
- Check whether the evidence comes from a plasma assay, an animal model, or a clinical study; these answer different questions.
- Ask whether the material was free DNA or RNA, NET-associated material, or a preparation that might contain polyphosphate or silica.
- Look for the nucleic-acid structure, concentration, preparation method, and contamination controls before comparing results.
- Distinguish normal hemostasis from thrombosis: promoting clot formation may be useful in one context and dangerous in another.
