Yes—nanopores can change when and how some drug molecules crystallize in laboratory experiments. The outcome depends on pore geometry and size, the material’s surface chemistry, and the interaction between the drug and pore walls. Researchers are investigating these effects as ways to control crystal formation or stabilize amorphous drug material; they have not established nanopore-engineered medicines as routine clinical treatments.
How can a nanopore affect drug crystallization?
A nanopore is a tiny space within a material. When drug molecules are confined in or near that space, the pore can influence the molecular arrangement and interactions involved in nucleation—the initial formation of a crystal. Depending on the pore and drug, confinement may hinder nucleation, promote it, affect the crystal state, or help keep the drug amorphous.
That means “nanopores promote crystallization” is not a general rule. Shape, diameter, surface chemistry, drug–surface interactions, and processing conditions all influence the result.

What experiments show about pore shape and size
Aspirin: angular and spherical pores behaved differently
A 2011 study by Diao and colleagues patterned polymer films with pores 15–120 nm in diameter. Under the study’s conditions, spherical pores hindered aspirin nucleation, while angular pores of the same size promoted it. The angular-pore effect depended on favorable interactions between the surface and aspirin; the authors suggested that molecular orientational order near pore angles may help explain the result. Fenofibrate: pore diameter was linked to the resulting state
Dwyer and colleagues studied fenofibrate in controlled-pore glass with ten pore sizes ranging from 12 to 300 nm. They reported drug loading above 20 wt% for pores larger than 20 nm. Nanocrystalline fenofibrate formed in pores above 20 nm; the study did not report the same crystalline result in smaller pores. The confined nanocrystals showed melting-point depression consistent with a Gibbs–Thomson relationship, as well as enhanced dissolution rates in the experiment. Those are laboratory findings, not evidence of improved patient outcomes. Royal Society of Chemistry, 2015.
Nanopores can also help stabilize amorphous drug material
Crystallization is not always the goal. Rengarajan and colleagues described using nanoporous hosts with strongly interacting pore walls to increase the lifetime of amorphous drugs. Confinement can affect thermodynamics and crystallization kinetics, so the direction of the effect depends on the host and the drug–pore interaction. Journal of Materials Chemistry, 2008.

What researchers need to control
Experimental approaches are best compared by what they are designed to achieve and how the material is built and processed—not as consumer treatment options.

- Pore geometry and diameter: shape and size can influence nucleation and the state of the confined drug.
- Host material and surface chemistry: the pore walls may interact with the drug, and the strength and nature of those interactions matter.
- Target outcome: an experiment may aim to trigger nucleation, favor a particular polymorph, form nanocrystals, or stabilize an amorphous state.
- Process conditions: temperature, pH, and other preparation conditions can affect what forms.
- Where crystallization occurs: crystals forming outside pores can limit dissolution, even when the pores themselves are intended to control the drug.
A 2020 review discusses mesoporous silicon’s loading capacity, tunable pore size, and adaptable surface, while identifying outside-pore crystallization and process conditions such as temperature and pH as translation concerns. Pharmaceutics, 2020.

Are nanopore-engineered medicines already in clinical use?
The cited studies and review describe laboratory research strategies, not proof that nanopore-engineered medicines are routinely prescribed or produce clinical benefits. In particular, a faster dissolution rate measured for confined fenofibrate does not establish better absorption, effectiveness, or outcomes for patients. Results from one drug, pore material, and set of conditions also cannot be assumed to apply to another.
Researchers have also explored templates beyond rigid nanoporous solids: hydrogel microparticles have been studied as a way to crystallize small-molecule drugs. This broadens the experimental approaches but does not establish a ready-to-use medical product. Chemical Communications, 2017.
