Mirror-image peptides can enter cells, but how well they do depends on the cargo and delivery system. In a 2026 laboratory study, researchers attached matched L- and D-form cargoes to the same cell-penetrating peptide and found that the L cargoes were taken up more efficiently in the tested cell models. The result describes one delivery setup—not a rule that D-peptides cannot enter cells.
What “hitch a lift” means in this study
A cell-penetrating peptide (CPP) is a short peptide used as a delivery scaffold: researchers attach a molecular cargo to it and test whether the construct can enter cells. The “lift” is the CPP carrying its cargo; it does not mean that the cargo independently crosses the cell membrane or necessarily reaches the cell’s cytosol.
Nassar, Donthoju, Hasan, and Brik compared three model cargoes made in L and D forms. They attached each matched pair to the same cyclic deca-arginine scaffold, cR10, to keep the delivery peptide constant while examining the effect of cargo chirality. Their paper, published online June 17, 2026, in the Journal of the American Chemical Society, reports that L cargoes were internalized more efficiently than their mirror-image D counterparts across the tested assays and cell models. What the 2026 experiments tested
The researchers synthesized three L/D cargo pairs of increasing size and structural complexity, then assessed uptake with flow cytometry, gel analysis, and confocal microscopy. The accessible figure descriptions identify HeLa, U2OS, and Jurkat cells. They describe flow-cytometry comparisons at 0.5, 1, and 2 micromolar after one hour, and confocal imaging in U2OS cells at 5 micromolar after one hour. The abstract reports the direction of the result but does not provide complete numerical uptake ratios for each cargo pair in the accessible record. It is therefore accurate to say that L cargo was taken up more efficiently in this cR10 setup; a specific percentage advantage is not established here. And “internalized” should not automatically be read as “delivered intact into the cytosol”: uptake assays can measure cell-associated or internalized material without proving its final location or function.
Why another study successfully delivered D-form cargo
An earlier study used a different delivery machine: anthrax protective antigen (PA) together with the N-terminal domain of anthrax lethal factor (LFN). PA binds cell receptors, is processed into a pore-forming assembly, and—after endocytosis and acidification—supports translocation of LFN-associated cargo. With this system, Rabideau, Liao, and Pentelute reported delivery of mirror-image peptide and protein cargo comparable to L-form counterparts. Their article explains the system and its results.

That paper also reported that a delivered D-peptide binder perturbed the intracellular p53/MDM2 interaction in cancer cells. This was a cell experiment, not evidence of a clinical treatment. For the delivered D-protein cargo, the authors reported cytosolic delivery but noted that they lacked tools to assess whether those proteins folded correctly or remained active inside the cytosol.

These studies are not a head-to-head comparison. They differ in delivery mechanism, constructs, cargoes, and readouts. The 2026 result asks how cargo chirality affects uptake when cR10 is held constant; the earlier work tests transport using a pore-forming translocation system. A successful result with one mechanism does not predict the outcome with another.

Does a D-peptide’s stability make it enter cells better?
No. A D-form peptide can resist proteolytic degradation better than its L-form counterpart, but resistance to degradation and cellular uptake are separate properties. Uptake, endosomal escape, cytosolic delivery, target binding, safety, and therapeutic efficacy are also distinct questions.
A 2023 study found that D-form cell-penetrating peptides retained delivery activity for antisense morpholino oligomers under the conditions tested and resisted proteolysis better than the native form. It is another example of a different CPP, cargo, and delivery context—not a replication of the cR10 cargo comparison. See the 2023 study.

What the findings do—and do not—show
- Shown: In the 2026 study, three matched cargo pairs attached to cR10 were internalized more efficiently in L form than in D form across the tested assays and cell models.
- Not shown: That D-peptides cannot enter cells, that every CPP favors L cargo, or that one delivery system is better than another.
- Not established: A clinical benefit, a treatment recommendation, or a complete numerical uptake advantage for each cargo pair.
The useful conclusion is specific: cargo chirality affected uptake in this cR10 laboratory system. To predict whether a particular mirror-image molecule will work, researchers would need to test that cargo with its intended delivery scaffold and measure the outcome that matters—such as intact cytosolic delivery or biological activity—not rely on chirality alone.
