Long-duration spaceflight changes how human cells regulate genes and maintain chromosomes. NASA’s year-long Twins Study found shifts in gene expression, telomere length, DNA methylation and markers associated with genome instability. Most measured gene-expression changes moved back toward preflight levels after astronaut Scott Kelly returned to Earth, although a smaller set remained altered months later. The evidence does not show that spaceflight permanently rewrote his inherited DNA sequence or gave him a wholly different genome from his identical twin.

What the NASA Twins Study actually compared

NASA compared retired astronaut Scott Kelly during a nearly year-long mission aboard the International Space Station with his identical twin, Mark Kelly, who stayed on Earth. Because identical twins begin with extremely similar inherited genomes, Mark provided an unusually strong personal control for age, family background and many genetic differences that complicate ordinary astronaut studies.

The project combined molecular, physiological and cognitive measurements from 10 investigations involving 84 researchers. The integrated, peer-reviewed findings were published in Science in 2019. It remains a detailed case study of one spaceflight participant, not a population estimate for every astronaut.

The comparison followed Scott before launch, during flight and after landing. That timing matters: a molecular signal detected in orbit can represent a temporary response, a slower recovery process or a change that persists after return to Earth.

Which genome-related systems changed?

Gene expression changed across many pathways

Gene expression is the process by which cells use DNA instructions to make RNA and proteins. It can change without altering the underlying DNA letters. NASA reported broad changes in Scott’s gene-expression profile during and after the mission, including pathways connected with immune activity, DNA repair, bone formation, low-oxygen responses (hypoxia) and high carbon-dioxide responses (hypercapnia).

NASA’s Twins Study overview says 91.3% of Scott Kelly’s gene-expression levels returned to baseline within six months of landing. A separate NASA preliminary-findings summary rounded the result differently, saying 93% of his genes returned to normal after landing. Those figures come from different NASA descriptions and should not be treated as interchangeable measurements; the 91.3% figure is the more specific value tied to the six-month follow-up.

Telomeres responded quickly

Telomeres are protective end sections of chromosomes that help preserve genomic stability. In Scott’s white blood cells, telomeres became significantly longer during the space mission. Most of that length was lost within two days after he landed, showing that the response was highly dynamic rather than a simple, permanent lengthening.

Telomere length is only one biological indicator. A longer telomere measurement during flight does not automatically mean improved health, and a later shortening does not by itself prove irreversible genetic injury. The significance depends on the cell type, the measurement time and other evidence about cellular function.

DNA methylation shifted in sampled immune cells

DNA methylation is an epigenetic chemical mark that can influence whether genes are more or less active without changing the DNA sequence. Methylation patterns changed in Scott’s sampled immune cells during flight. NASA’s overview also notes that the degree of epigenetic change in Scott was no greater than the change observed in Mark while he remained on Earth. Therefore, a methylation difference should not automatically be labeled a spaceflight-specific effect; ordinary variation, stress or other terrestrial factors can contribute.

Chromosomal and DNA-damage signals appeared

The integrated Twins Study paper reported genome-instability and DNA-damage-related findings, including chromosomal changes. These are molecular or cellular signals measured in particular samples. They do not mean that every cell acquired a permanent new genome, nor do they establish that Scott’s inherited DNA sequence was rewritten.

What recovered after landing—and what did not?

Recovery was uneven. Many molecular measurements moved toward Scott’s preflight baseline after he returned to Earth, while a smaller subset remained altered at the end of follow-up. The residual signals included pathways involved in immune regulation, DNA repair, bone formation, hypoxia and hypercapnia.

NASA also reported lingering or partly lingering changes outside the genome itself. Immune responses, metabolism, gut microbiota, carotid-artery measures, ocular structure, body mass and cognitive performance all changed during the mission. Many trends moved back toward baseline, but some vascular, ocular, cognitive, telomere and DNA-related signals were still different at the final follow-up point described by the study.

“Recovery” therefore does not mean that every measurement instantly returned to its launch value. It means that the direction or magnitude of a signal moved closer to the individual’s earlier state, sometimes over days and sometimes over months.

Does a short mission produce the same effect as a year in orbit?

No. Mission duration, radiation exposure, spacecraft environment, workload, sleep, exercise, diet and sampling schedules all affect interpretation. A three-day flight and a nearly year-long International Space Station mission should not be treated as equivalent experiments.

Study or mission Mission length Control and setting Genome-related observations What the comparison can show
NASA Twins Study Nearly one year in orbit Identical twin Mark Kelly remained on Earth; repeated preflight, in-flight and postflight samples Gene-expression shifts, telomere lengthening followed by rapid shortening after landing, DNA-methylation changes and genome-instability signals Detailed before-and-after adaptation and recovery patterns in one astronaut, with an unusually strong personal control
Space Omics and Medical Atlas analysis of Inspiration4 Three days Commercial orbital mission; not an identical-twin, year-long control design Rapid telomere-length dynamics were reported during the mission Shows that telomere responses can occur on short flights, but does not measure the same exposure or recovery timeline as the Twins Study

The 2024 Space Omics and Medical Atlas findings complement rather than replace the Twins Study. They indicate that telomere behavior can change during a mission lasting only three days, while the year-long study provides more information about prolonged exposure and the months following landing.

Does space travel change an astronaut’s DNA?

The most accurate answer is: spaceflight can change gene regulation and produce DNA- and chromosome-related cellular signals, but current evidence does not show that it permanently rewrites an astronaut’s inherited DNA sequence.

That distinction separates four different ideas:

  • DNA sequence: the chemical order of DNA bases inherited from a person’s parents. The Twins Study did not establish a permanent, whole-body rewrite of this sequence.
  • Gene expression: which genes are more or less active at a given time. This changed substantially during Scott Kelly’s mission.
  • Epigenetic marks: regulatory marks such as DNA methylation that can alter gene activity without changing the sequence. These changed in sampled cells, but some comparable change occurred in Mark on Earth.
  • Genome stability: the maintenance of chromosomes and DNA integrity in cells. The study detected instability and damage-related signals, which are risk indicators rather than proof that every cell suffered permanent damage.

How long can an astronaut stay in space before genetic changes occur?

There is no established exposure threshold at which a person suddenly crosses from “normal” into permanent genetic damage. The available findings show different timescales: telomere changes were detectable during a nearly year-long mission and mostly reversed within two days of landing, while the Inspiration4 analysis found telomere dynamics during a three-day flight. Gene-expression and other molecular changes can persist for months, but the magnitude and duration vary by pathway and person.

Radiation environment, microgravity, isolation, disrupted sleep, exercise, nutrition and carbon-dioxide exposure can all contribute. The Twins Study cannot determine how long every astronaut can remain in orbit safely because it involved effectively one flight subject and was not designed to calculate a universal limit.

Why the findings matter for future missions

NASA describes the body as resilient and capable of adapting to many spaceflight stresses, but adaptation is not the same as absence of risk. Persistent changes in immune regulation, DNA repair, bone pathways, vascular function, vision or cognition could matter more on missions that last longer than a year or that place crews farther from Earth, where medical support and radiation shielding differ.

The practical value of the findings is monitoring and countermeasure design. Future programs can track molecular signals alongside cardiovascular, ocular, skeletal, immune and cognitive measurements, then test whether exercise, habitat design, radiation protection, sleep scheduling or other interventions shorten recovery. Longer missions and larger groups are needed to determine which changes are common, which are individual, and which predict clinically important outcomes.

What the evidence cannot yet establish

  • The Twins Study cannot provide a percentage risk for all astronauts because Scott Kelly was one flight participant.
  • A molecular change is not automatically a disease or a permanent mutation.
  • Changes seen in one tissue, such as white blood cells, cannot be assumed to occur identically in every organ.
  • Findings from a three-day mission cannot be used to calculate the effects of a year in orbit, and a year in orbit cannot by itself predict a multiyear Mars mission.
  • Recovery at the end of the reported follow-up does not prove that every biological effect has resolved or that no later effect can occur.

The strongest conclusion is measured rather than dramatic: long stays in space alter gene regulation, telomere behavior and other cellular systems, many responses show substantial recovery after landing, and a smaller set can persist. Scientists have not shown that an astronaut returns to Earth with a permanently rewritten inherited genome.
