Label-free optical detection identifies particles from the light they scatter, absorb, or alter as it passes through them—without attaching a fluorescent tag. Some methods can detect or track particles moving in a fluid; others deliberately sense only the region close to a sensor surface. The distinction matters: a surface-sensitive reading is not the same as observing particles throughout a sample volume.
What “label-free” means—and what it does not
In fluorescence microscopy, a fluorescent tag attached to a target supplies a conspicuous signal. In label-free optical detection, the measured signal instead comes from the particle or analyte’s own interaction with light. Depending on the method, that interaction may appear as scattered light, a change in phase, absorption, or a refractive-index contrast.
Removing the tag can avoid photobleaching—the loss of fluorescence under illumination—and reduce the ways a label may perturb a target. A 2025 review in npj Biosensing notes that label-free methods do not suffer from label photobleaching, which can support prolonged observation and real-time tracking with minimal perturbation. That does not mean every label-free experiment leaves the sample untouched: surfaces, illumination, fluid handling, and other parts of the setup can still affect it. The review was corrected on 28 October 2025.

“Label-free” also does not mean “no preparation” or “no engineered sensor.” A sample may still need to be placed in a controlled chamber, and a surface-based assay may require a prepared interface or a target that binds to it. The key question is where the instrument detects the particle: at or near an interface, or while it is moving through the fluid volume.

How a weak particle signal becomes measurable
Interferometric scattering compares the particle with a reference field
A nanoscale particle scatters only a small amount of light, so measuring its scattered light directly can be difficult. Interferometric scattering microscopy, or iSCAT, makes the weak scattered field interfere with a reference field. In many configurations, reflected light from an interface supplies that reference. The particle changes the combined optical signal, producing a detectable intensity pattern even when its own scattering is faint.
A useful analogy is comparing a quiet sound against a steady reference tone: the comparison can make a small difference easier to notice than listening for the quiet sound alone. In iSCAT, the instrument records the combined light, and analysis uses its changes to locate or characterize the particle. The result depends on optical configuration and sample conditions; interference is a detection strategy, not a guarantee of a particular sensitivity in every setup.
A 2019 Annual Review of Physical Chemistry review describes iSCAT as an all-optical method for label-free imaging, tracking, and quantitative characterization of nanoscale objects. It covers applications including particle tracking, biomolecular assembly, and mass measurement. The review explains why the reference field is central to detecting weak scattering.

Other methods shape or interpret the optical field differently
Not every label-free technique measures the same signal. Some approaches use plasmonic structures or resonators to enhance a local field or respond to a change in the surrounding refractive index. Others use holographic methods to record information that can be reconstructed as a complex optical field. These are related ways of extracting information from light, but their signal mechanisms and preferred geometries differ.
Which methods can see particles away from a surface?
“Beyond a sensor surface” can mean two different things: detecting particles that remain in the fluid instead of binding to a sensor, or sensing a region that extends some distance into the liquid from an interface. Those are not equivalent. A particle can be near a surface without being tethered to it, while a surface-sensitive method may still be largely blind to particles farther into the sample.
Free-solution tracking observes motion in the fluid
Optical tracking can follow Brownian motion—the continual random movement of small particles—or other motion in a fluid. In a free-solution geometry, the target need not land on or bind to a sensor surface for observation. This can be useful when the experimental question concerns how a particle moves or behaves in solution rather than how it attaches to an interface.

The setup is a system, not simply a microscope objective: the source, sensors, optics, and stages all contribute to what can be detected and tracked. A 2016 NIST review discusses these components and how tuning illumination and detection changes the capabilities of particle-tracking systems. NIST’s overview provides instrumentation context for tracking nanoscale particles in microscale environments.

Surface-plasmon and evanescent-field approaches concentrate optical fields near an interface. A 2023 Chemical Society Reviews article reports that the field associated with surface plasmons penetrates roughly 30–200 nm into the adjacent liquid. Changes in local refractive index within that region can therefore affect the optical response.
That distance describes the field’s penetration into the liquid, not a general particle-detection limit or a universal maximum viewing depth for every optical technique. The geometry is especially relevant when targets interact with the sensing surface. It should not be described as equivalent to tracking freely suspended particles throughout a bulk fluid volume. The review discusses this approach in the context of label-free optical biosensing, which it characterizes as predominantly a research tool. Read the 2023 review.

Holography can add characterization information, not automatic sensitivity
Holographic methods record information that can be reconstructed as a complex optical field. That field can help estimate particle size and refractive index and, in some cases, distinguish material classes from their optical response. A 2025 Nanoscale review describes these capabilities alongside interferometric approaches for characterizing dielectric nanoparticles.
More information in a measured field does not necessarily mean a lower detection limit. The review identifies detection limit as a historical drawback of holographic microscopy compared with some other approaches. Whether holography is suitable depends on the particle, sample, optical system, and whether the goal is merely to detect a particle or also to infer properties from its response. The review’s discussion of optical label-free microscopy covers these trade-offs.

| Approach | What is measured | Surface relationship | Useful when the goal is |
|---|---|---|---|
| Interferometric scattering microscopy (iSCAT) | Interference between particle-scattered light and a reference field | Can use reflected light from an interface as the reference; that does not by itself mean the particle must be tethered | Imaging, tracking, or quantitative characterization of nanoscale objects |
| Surface-plasmon or evanescent-field sensing | Changes in the local optical response, including refractive-index changes near an interface | Strongly emphasizes the interface; the reported surface-plasmon penetration into liquid is roughly 30–200 nm | Sensing targets or changes in the region near a surface |
| Holographic microscopy | A reconstructed complex optical field | Depends on the specific setup; the cited review does not establish one universal surface geometry | Estimating size or refractive index, or distinguishing some materials by optical response |
| Optical particle tracking in fluid | Particle position and motion over time | Can observe particles without requiring them to land on a surface; some other scattering approaches instead rely on tethering or surface interaction | Following Brownian motion or other particle motion in a fluid |
When comparing instruments or experimental designs, work through these questions:
- Does the particle have to bind or approach a surface? If the study concerns freely suspended particles, distinguish a bulk-fluid tracking geometry from a surface assay or a method that needs tethering. Reviews of optical scattering methods explicitly distinguish surface-tethered and free-solution approaches. The 2024 review focuses on label-free analysis of single biomolecules.
- What optical signal carries the answer? Scattering, interference, phase, a resonance change, and a reconstructed field are different measurements. Choose based on what the experiment needs to observe or infer rather than treating “optical detection” as one technique.
- What are the particle and sample conditions? Particle concentration, optical contrast, and background from the sample can all affect suitability. Reviews discuss limitations across methods, but do not support a universal sensitivity ranking across every sample and configuration.
- Do you need detection, tracking, or characterization? Finding that a particle is present is a different task from following its trajectory or estimating properties such as size, mass, or refractive index. Check that the method supports the intended output.
- What hardware and preparation are required? Consider illumination, optics, sensors, stages, surface preparation, and analysis. Specialized configurations may be necessary; an ordinary microscope should not be assumed sufficient for nanoscale label-free detection.
A broad overview of optical systems for detecting unlabeled nanoparticles discusses the range of approaches and their limitations, rather than identifying a single configuration for every use. The 2022 review can help frame that landscape.
What label-free detection does well—and where it remains difficult
Removing fluorescent labels avoids label photobleaching and can reduce label-related perturbation, which may be valuable for prolonged observation. But a particle’s native optical signal can be weak, particularly when the particle scatters little light or the sample background complicates measurement. Methods address that challenge in different ways, including interference with a reference wave, plasmonic enhancement, resonators, and nanoscale apertures.
Quick wins for a faster PC:
Repair Windows errors before they cause bigger problemsFix Now →Fix the driver behind crashes, sound loss and screen glitchesFind Drivers →Those strategies introduce their own design choices. Enhancement tied to an interface can favor near-surface measurements; a configuration intended to track particles in fluid has different optical and analysis requirements. Signal quality, robustness, versatility, and performance in a particular sample depend on the method and how the system is configured. A 2025 overview describes sensitivity, robustness, and versatility as ongoing challenges in the field, rather than problems already solved by a single architecture. Its review of label-free single-molecule optical detection surveys enhancement strategies and current challenges.
There is no single consumer instrument or universal detection threshold established for this broad topic. The methods discussed in the literature are research-oriented optical systems whose fit depends on the target, sample, geometry, and measurement goal. For that reason, compare the experimental requirements and the signal each system measures—not just whether it is called “label-free.”
