Researchers tracked newly made sugar chains in mice by feeding the animals an azide-bearing sugar precursor, then attaching a detectable Flag tag to the resulting glycans with copper-free click chemistry. The key qualification: the reported Flag assay was performed on extracted cells, not by directly imaging sugar synthesis inside a living mouse.
How the mouse experiment worked
Glycans are complex sugar chains assembled by cells and found inside cells and on their surfaces. Their patterns can vary among cell types and tissues, and can change with cell state, development, or disease. Conventional tools such as antibodies and lectins have limits when researchers want to study those patterns in living animals.
In the method described by Chemistry World in its report on Pamela Chang and colleagues at the University of California, Berkeley, researchers supplied a synthetic sialic-acid precursor carrying an azide group. Cells used the modified precursor in glycan biosynthesis, placing the azide handle into newly synthesized glycans.

The researchers then used a strained cyclooctyne linked to a Flag peptide. The cyclooctyne reacted selectively with the azide in a copper-free click reaction, attaching the Flag tag to the labeled glycans. A standard biological assay could detect that tag.

What “in live animals” means here
The labeling began in the living mouse: the precursor entered the animal and was incorporated into glycans through cellular biosynthesis. But the reported readout depended on extracting cells and detecting the attached Flag tag afterward. This was not direct, noninvasive imaging of glycan synthesis inside a live mouse.
Chang described the approach as a way to monitor glycan-expression patterns across cell types and tissues, saying, “In this way we can monitor the expression of patterns of glycans in different cell types and tissues.” That describes the method’s potential for pattern analysis, not an imaging result from the mouse experiment. Chang identified attaching an imaging agent to the cyclooctyne as a future goal.

How it compares with the earlier zebrafish demonstration
Copper-free click chemistry had also been used in a distinct 2008 study of living zebrafish embryos. That work incorporated azide-modified sugars and reacted them with strained fluorescent alkynes, allowing optical imaging of sugar patterns during embryonic development. It reported bursts of sugar synthesis in the jaw, pectoral fins, and olfactory organs at 60 hours after fertilization, according to Monica Harrington’s 2008 Lab Animal summary.

| Study | Model | Reported readout |
|---|---|---|
| Chang and colleagues, reported by Chemistry World | Mice | Flag-tag detection in extracted cells |
| Study summarized by Monica Harrington in Lab Animal, 2008 | Zebrafish embryos | Optical imaging of sugar patterns |
Both studies illustrate metabolic labeling followed by click chemistry, but their readouts differ. The zebrafish imaging result should not be attributed to the mouse experiment.
Why the approach mattered—and what it did not establish
Labeling glycans through biosynthesis offered a way to investigate where particular sugar-building activity occurs across tissues, rather than relying only on conventional recognition reagents. The report discussed embryonic development and cancer spread as motivations for studying glycan patterns; it did not establish a clinical diagnostic or show that the method could track cancer in patients.

Nor does the described method show that every glycan can be tagged. It concerns an azide-bearing sialic-acid precursor and glycans into which that precursor was incorporated. The report’s proposed imaging direction was a possible next step, not a demonstrated capability of the mouse study.
Later work on azido-sugar reagents
A 2024 ChemSusChem paper by Frohnmeyer and colleagues reported enzymatic gram-scale production of UDP-6-azido-GalNAc, an unnatural nucleotide sugar relevant to glycan engineering and click-chemistry applications. The authors reported production of up to 2.1 g, yields up to 97% across five consecutive batch cycles, and 99.96% HPLC purity for purified product.
