A 2021 study introduced a way to join peptide chains through side chains, enabling the synthesis of cyclic, branched, and bridged peptide structures. Its key workaround is to install a benzofuran mask during peptide synthesis, then use ozonolysis to reveal a reactive salicylaldehyde ester for ligation. The results demonstrate a flexible synthetic strategy—not a proven treatment or drug candidate.
Why use a masked salicylaldehyde ester?
Salicylaldehyde esters can serve as reactive handles for joining peptide fragments, but installing them directly on aspartic acid or glutamic acid side chains is difficult. The active ester can be unstable during peptide synthesis and handling, and approaches for placing salicylaldehyde esters at a peptide’s C-terminus do not solve the side-chain installation problem.
In the strategy reported by Carina Hey Pui Cheung, Xuechen Li, and collaborators, a benzofuran group serves as a masked form of the salicylaldehyde ester. It can be incorporated during standard Fmoc solid-phase peptide synthesis (Fmoc-SPPS), where the reactive ester itself would be challenging to handle. After synthesis and global deprotection, ozonolysis converts the benzofuran into the active salicylaldehyde ester. The 2021 Chemical Science paper describes this approach and its synthetic demonstrations.

- Build the peptide with latent and partner groups. Install the benzofuran-derived precursor at a peptide side chain using Fmoc-SPPS. The design also provides a hydroxyl-amino functionality, using Dap–Ser or Lys–Ser.
- Reveal the reactive handle. Following global deprotection, treat the peptide with ozone. This converts the benzofuran mask into a side-chain salicylaldehyde ester.
- Join the peptide partners. The revealed ester reacts through Ser/Thr ligation or Cys/Pen ligation, producing the intended covalent connection.
The approach uses chemoselective ligation to form native peptidic linkages at the joining sites. Its practical appeal is that the reactive group is exposed after peptide assembly rather than carried through the synthesis in its less stable active form. What peptide structures did the study make?
The study reports several architectures made through side-chain ligation:
- Side-chain-to-side-chain cyclic peptides
- Branched and bridged peptides
- Tailed cyclic peptides
- Multi-cyclic peptides
The authors also describe ligating peptides carrying a side-chain salicylaldehyde ester to partners bearing Dap/Lys–Ser or an N-terminal Ser, Thr, Cys, or Pen. These are chemical syntheses demonstrating structural scope; the reported work does not establish that any particular structure inhibits a protein interaction or treats disease.

Which residues are incompatible with the ozonolysis step?
Ozone is a strong oxidant, so the authors identify cysteine, methionine, and tryptophan as incompatible with the ozonolysis used to uncover the salicylaldehyde ester. Those residues can react during the oxidation, limiting which peptide sequences can be prepared by this route. This is a sequence constraint of the handle-revealing step, not evidence that the ligation chemistry itself is universally unsuitable for those residues.
Does the strategy produce a therapeutic?
No therapeutic efficacy, clinical use, or marketed drug is demonstrated in the paper. The structural diversity could help researchers explore constrained peptides, including candidates intended to interfere with protein–protein interactions, but that is a prospective application rather than a result validated by this study. Chemistry World reports that the team hoped to extend the method to higher-order cyclic peptides and investigate PPI inhibitors and peptide–drug conjugates; these are future directions, not demonstrated outcomes.

What the method establishes—and what it does not
The study establishes a synthesis strategy: a benzofuran mask makes a side-chain salicylaldehyde ester more compatible with peptide assembly, and ozonolysis reveals the handle for subsequent ligation. It demonstrates multiple peptide topologies and native linkages at ligation sites, while also identifying an important ozone-related sequence limitation. It does not provide a head-to-head comparison with other ligation methods or biological validation of the resulting structures.
