An experimental compound that directly activates the energy-sensing enzyme AMPK extended lifespan in fission yeast, nematode worms and fruit flies in a study published in Aging Cell on 29 September 2026. The reported increase was more than 25% in some cases—not a result established for every species or experiment. The study did not show that mice or people lived longer.
What the longevity study found
The researchers tested compound 991, a direct activator of AMP-activated protein kinase (AMPK), in three organisms: fission yeast (Schizosaccharomyces pombe), nematode worms (Caenorhabditis elegans) and fruit flies (Drosophila melanogaster). They reported lifespan extension in all three. UK Research and Innovation described the increases as “in some cases by over 25%,” which is a selective result, not a universal percentage for every organism, dose or experimental condition.
The available institutional summary does not give a single percentage that can be applied to all three species. The paper and its figures are the appropriate place to assess species-, dose- and condition-specific results. The headline-level finding alone does not justify assigning the same effect size to each organism.
Why AMPK matters—and what compound 991 tests
AMPK is an enzyme that helps cells respond to energy status. Compound 991 binds to and activates AMPK directly, allowing researchers to probe the consequences of activating that target. The study reports that the lifespan effect disappeared in worms and yeast without functional AMPK, supporting the interpretation that AMPK was necessary for the effect in those models.

This is not evidence that every way of influencing energy metabolism will produce the same outcome. Metformin, for example, acts through a more complex set of mechanisms and can affect other metabolic pathways. The study’s authors distinguish it from 991; the compounds should not be treated as interchangeable longevity interventions.
Dose mattered: more activation was not always better
Queen Mary University of London’s summary says higher doses shortened lifespan in some experiments. The findings therefore depend on dose and experimental conditions; they do not support the assumption that stronger AMPK activation would yield greater longevity. A result from a controlled experiment with an experimental compound also does not establish a safe or effective dose for people.
What the mouse experiments did—and did not—show
Mice received treatment for three weeks. Researchers observed liver protein-expression changes consistent with AMPK activation, including changes associated with ATP production, mitochondrial biogenesis and suppression of the growth-promoting mTOR pathway. These are molecular findings, not evidence that the mice lived longer or that their health improved.

Does this mean AMPK activation can extend human lifespan?
No human longevity benefit has been established. The lifespan results came from yeast, worms and flies; the mouse work measured molecular changes over three weeks rather than lifespan. These experiments make AMPK a subject for further study, but they do not show that 991—or another AMPK-targeting treatment—extends human life or improves human health.
The study also does not support taking a supplement for longevity. Study co-author Dr Charalampos Rallis cautioned: “I would not want anyone to reach for a supplement on the back of this.”

How to read claims about longevity research
- Check the organism: a lifespan effect in a simple model organism is not a human result.
- Separate lifespan from biomarkers: protein changes consistent with pathway activation do not prove longer life or better health.
- Look at dose and conditions: the report says higher doses shortened lifespan in some experiments.
- Distinguish mechanisms: directly activating AMPK with 991 is not the same intervention as taking metformin or a supplement.
- Keep percentages attached to their scope: “over 25%” describes some cases in the institutional summary, not all species or trials.
