A 2025 Science paper reports an iron-catalyzed method for accessing donor, neutral, and acceptor carbenes, then using these reactive intermediates to make cyclopropanes. The work is a synthetic-method development result—not a new drug or a demonstrated treatment—and its proposed use inside living cells remains a future possibility.
What the researchers developed
The paper, by Khue N. M. Nguyen and colleagues, presents a catalytic approach designed to generate three classes of carbene distinguished by their electronic polarity: donor, neutral, and acceptor. These highly reactive intermediates are useful building blocks in synthesis, but methods for making them can rely on limited or hazardous precursor strategies.
The institutional summary describes the approach as combining iron catalysis with chlorine-based molecules that generate free radicals. Those radicals enable formation of selected metal carbenes, which can add to another molecule to produce a cyclopropane—a strained ring containing three carbon atoms. Ohio State News explains the reaction concept; the publication record identifies the article as “Harnessing carbene polarity: Unified catalytic access to donor, neutral, and acceptor carbenes,” in Science, volume 389, pages 183–189 (2025). The Nagib Research Group publication list links to the paper.
Why carbene polarity matters
Donor, neutral, and acceptor describe different electronic character, which affects how a carbene behaves in a reaction. A method that can reach all three types offers chemists a broader synthetic toolkit than one limited to a single carbene class. The paper’s central contribution, as presented in its title and institutional account, is this unified access paired with cyclopropane formation.
#1 Best Overall
- FOR BASIC TEACHING TO ADVANCED SCIENCE: 444 pieces molecular model kit, including 136 atoms, 158 bonds and 150 parts for Carbon-60(Fullerene), provides to students from Grade 7 to Graduate level.
- TWO CHEMICAL STRUCTURE MODELS: The ball-and-stick models use spheres to represent atoms and sticks to represent chemical bonds. In the space-filling model, the spheres are drawn to scale and are next to one another as atoms are in real molecules.
- CHEMISTRY EDUCATIONAL MOLECULE MODEL IN 3D: It can display chemical structure, molecular bond, and bond angle in all directions. Demonstrate fundamental molecular geometry, chemical molecular structure, stereochemistry with 3D modeling studies.
- EASY TO LEARN: The universal standard adopted for each atom's color makes it easier for you to use and learn. Atoms and chemical bonds combine tightly and firmly and can be easily disassembled by disconnecting tools.

Cyclopropane rings appear in medicinal and agrichemical chemistry, making ways to build them of interest to synthesis researchers. That relevance does not mean this method has produced a medicine, improved patient outcomes, or established a biomedical application.

The “about 100 times better” comparison
Ohio State News attributes to David Nagib the statement that the approach is “about 100 times better” than chemical tools his lab produced over the preceding decade. The accessible account does not define the metric or the comparison conditions, so this is an attributed, underspecified comparison—not a general independent performance benchmark.

Choosing a synthetic method requires experimental details that are not established in the accessible institutional and news accounts. In particular, those accounts do not verify the paper’s detailed reaction scope, yields, selectivity, catalyst loading, substrate limitations, or full safety information. Those factors—and the exact alternatives used for comparison—are necessary to judge how the method performs for a particular synthesis.

Nagib described the group’s aim as exploring new ways to access carbenes: “Our goal all along was to determine if we could come up with new methods of accessing carbenes that others hadn’t found before,” he told Ohio State News. He also summarized the group’s focus: “Our lab is very much a tool development lab.”
